    Notes: The specific activities of 6 enzymes, which are involved in the synthesis and catabolism of membrane lipids, were monitored in plasma membranes isolated from petunia petals during senescence. These included phosphatidylinositol (PI) kinase (EC 2.7.1.67), phosphatidylinositol monophosphate (PIP) kinase (EC 2.7.1.68). diacylglycerol (DAG) kinase (EC 2.7.1.107), phospholipase A (EC 3.1.1.4) and PIP- and PIP2-phospholipase C˙(EC 3.1.4.3). Using endogenous substrate, the [32P]PA and [32P]PIP2 formation increased to 140 and 200%, respectively, of the day 1 value by 4 days after harvest. There was no significant change in [32P]PIP formation during the same time period. On the fifth day the petals wilted and the [32P]PA and [32P]PIP formation declined significantly. In contrast, the [32P]PIP2 formation remained high in the day 5 petals. When the lipid kinase activities were assayed in the membranes in the presence of exogenous substrate the specific activity of all of the enzymes increased. and the changes in [32P]PA production over the 5-day period were similar to those observed with endogenous substrate. When exogenous PI and PIP were added, however, there was no longer an increase in [32P]PIP2 formation by plasma membranes of day 4 petals and [32P]PIP formation significantly decreased. The relative decrease in PIP and PIP2 formation by day 4 membranes when exogenous substrate was added may have resulted from differences in the lipase activities in the day 1 and day 4 membranes. The plasma membrane A-type phospholipase activity increased throughout the 5 day period, and phospholipase C activity increased two-fold between day 1 and day 4. Such changes in the metabolism of the plasma membrane lipids during flower senescence would affect the ability of the petals to use inositol phospholipid-based signal transduction pathways.

    Notes: Abstract The macroscopic and microscopic acid-base chemistry of a series of sulfhydryl and ammonium-containing amino acids HS−R−NH3 [R=−CH2CH(COOH)−, cysteine (CYS); R=−C(CH3)2CH(COOH)−, penicillamine (PEN); R=−CH(COOH)CH2CH2CONHCH(−CH2)CONHCH2COOH, glutathione (GSH)] was characterized in water and its binary mixtures with acetonitrile (16.3, 34.2, and 53.9 mass % acetonitrile). Macroscopic acid dissociation constants were obtained by potentiometric titration using the glass-calomel electrode pair. Microscopic acid dissociation constants were calculated from ultraviolet absorption measurements at ca. 232 nm where the deprotonated sulfhydryl group absorbs. The macroscopic constants decrease uniformly as the solvent becomes enriched in acetonitrile. The microscopic constants, which characterize the relative concentrations of the two monoprotonated tautomers of the molecules (I and II) reveal that as the solvent becomes enriched in acetonitrile, the fraction of molecules existing as highly charged tautomer I decreases for CYS (0.68–0.40), PEN (0.85–0.34), and GSH (0.61–0.30). These results are related to the decreasing concentration of water as the solvent becomes enriched in acetonitrile.

    Notes: Abstract This article addresses the methodological problem of the non-linear representation of philosophical systems in a computerized knowledge base. It is a problem of knowledge representation as defined in the field of artificial intelligence. Instead of a purely theoretical discussion of the issue, we present selected results of a practical experiment which has in itself some theoretical significance. We show how one can represent different philosophies using CODE, a knowledge engineering system developed by artificial intelligence researchers. The hypothesis is that such a computer based representation of philosophical systems can give insight into their conceptual structure. We argue that computer aided text analysis can apply knowledge representation tools and techniques developed in artificial intelligence and we estimate how philosophers as well as knowledge engineers could gain from this cross-fertilization. This paper should be considered as an experiment report on the use of knowledge representation techniques in computer aided text analysis. It is part of a much broader project on the representation of conceptual structures in an expert system. However, we intentionally avoided technical issues related to either Computer Science or History of Philosophy to focus on the benefit to enhance traditional humanistic studies with tools and methods developed in AI on the one hand and the need to develop more appropriate tools on the other.

    Description / Table of Contents: Zusammenfassung Der3H-Thymidin-Aufnahmeindex in der Leber und das Serum AFP wurden bei Ratten, welche bei einer standardisierten Laboratoriumsdiät oder mit einer Cholinmangeldiät ernährt wurden, bestimmt. Es wurde festgestellt, daß die Proliferation der Leberzellen bei Tieren, die die Mangeldiät erhalten haben, signifikant erhöht war. Weiterhin wurde bei einigen dieser Ratten eine erhöhte Serumkonzentration des AFP gefunden. Die Befunde deuten darauf hin, daß die vermehrte Synthese des AFP einer Leberzellregeneration, welche sekundär zu einem Leberschaden entsteht, entspricht.
    Notes: Summary Liver3H-thymidine labelling index and serum AFP were determined in rats fed a standard laboratory or choline deficient diet. A significant increase in the rate of liver cell proliferation was evidenced in animals on the deficient diet. Furthermore, elevated serum AFP levels were detected in a proportion of these rats. It is suggested that increased synthesis of AFP corresponds to liver cell regeneration secondary to hepatic damage.

    Notes: Abstract Rat liver and brain subcellular esterase activities toward nicotinic acid esters were studied, under varying conditions, such as pH, organic solvents, protein concentration, duration of incubation, and substrate concentration. Esterases in each subcellular fraction displayed activities that obey Michaelis–Menten kinetics, although subcellular fractions are heterogeneous. The K m values were of the same magnitude, and the V max values were lower in microsomes than in cytosol of the liver. Brain activities normalized to protein concentration, were much lower than liver activities, aromatic nicotinates being the best substrates in both tissues. Myelin and brain mitochondria of nerve-ending and neuroglial origin display esterase activity toward phenyl nicotinate. In contrast to brain esterases, liver esterases appear homogeneous, and esterase activities in both tissues react differently to changes in pH. Qualitative and quantitative structure–metabolism relationships are not suggestive of tissue-specific ester hydrolysis.
