    Description: The Drosophila homeobox segmentation gene fushi tarazu (ftz) is expressed in a seven-stripe pattern during early embryogenesis. This characteristic pattern is largely specified by the zebra element located immediately upstream of the ftz transcriptional start site. The FTZ-F1 protein, one of multiple DNA binding factors that interacts with the zebra element, is implicated in the activation of ftz transcription, especially in stripes 1, 2, 3, and 6. An FTZ-F1 complementary DNA has been cloned by recognition site screening of a Drosophila expression library. The identity of the FTZ-F1 complementary DNA clone was confirmed by immunological cross-reaction with antibodies to FTZ-F1 and by sequence analysis of peptides from purified FTZ-F1 protein. The predicted amino acid sequence of FTZ-F1 revealed that the protein is a member of the nuclear hormone receptor superfamily.     Notes: Abstract A total of 1,427 juvenile Aplysia kurodai were tagged and released during spring tides in January and February, 1973, on the rocky shore of Nabeta Bay, Japan. Recapture, weighing and release were repeated within a defined observation area during 10 spring tides from late January to early June. Recapture rates decreased with time and became significantly low after the end of the spawning season (March and April in this area). Of 1,427 individuals released, 550 (ca. 39%) were never recaptured. Only 68 individuals (ca. 5%) were recaptured 5 to 10 times during the observation period. The growth curves of these 68 individuals displayed a common growth pattern of body weight increasing almost linearly, attaining a maximum during the spawning season and then decreasing gradually. The maximum weight varied fairly widely from one individual to another. Laboratory measurements indicated a growth pattern similar to that observed in the field, and a fairly remarkable daily fluctuation in body weight was observed under laboratory conditions. The described tagging-recapture method, with slight modifications, should prove useful for future studies on the life history and ecology of A. kurodai.

    Notes: Summary A method has been developed for the histochemical demonstration of unsaturated lipids in light microscopy. It is a peracetic acid-thiocarbohydrazide-silver protein sequence followed by a physical development procedure. In the present study on paraffin and cryostat sections of liver, brain and ovary, unsaturated lipids were visualized as distinct reaction products coloured various shades of brown and black. The reaction products are easier to see and the method is more efficient than the peracetic acid-Schiff method.

    Notes: Abstract We have developed an efficient system for obtaining myeloma mutants defective intrans-acting factors required for immunoglobulin (Ig) gene expression. The system consists of a myeloma cell line designed for this purpose and an efficient method for selecting mutants from it. The cell line is X63.653 transfected with the μ gene, whose tailpiece sequence was replaced with the transmembrane sequence of human EGF receptor to hold μ on the cell surface and whose CH1 sequence was removed to prevent μ from being retained in the endoplasmic reticulum. It efficiently and stably expressed μ chains of IgM on the cell surface (μ m + ) without light chains. To obtain mutants lacking μm (μ m − ) from the μ m + cell line by selectively killing μ m + cells, a method with ricin A-conjugated anti-μ antibody was more reliable than complement lysis mediated by anti-μ antibody. Applying the system, we obtained a variety of μ m − mutants.

    Notes: Abstract The nitriding rate in a plated invar with an Fe-Ni-Cr alloy (face-centred cubic) layer were compared by optical microscopy with the rate in an unplated invar. In the plated invar rapid nitrogen diffusion was discovered. The plated layer plays a role in enhancing the nitriding rate. The result gives a solution for an abnormal enhancement nitriding rate in austenitic stainless steels. The enhancement is explained from the viewpoint of non-equilibrium thermodynamics.
