    Notes: Circular dichroism spectroscopy has been used to investigate the influence of DNA molecular size, base composition, and the presence of intercalating agents upon the Ψ transition of DNA brought about by high concentrations of poly(ethylene oxide) and salt (Lerman (1971) Proc. Natl. Acad. Sci. (U.S.) 68, 1886-1890). A molecular weight of 0.15-3.0 × 106 daltons yields maximum formation of Ψ-DNA. Both the amplitude of the large negative CD band at 265 nm - a chief characteristic of the Ψ state - and the thermal stability of Ψ-DNA increase linearly with increasing mole fraction of guanine plus cytosine in the DNA sample. Either ethidium or proflavine, at concentrations where approximately one dye is bound per 5-10 nucleotide residues, can prevent the transition completely. Striking similarities between the Ψ-DNA produced by poly(ethylene oxide) + salt and the complexes formed between DNA and lysine-rich histone f1 suggest the presence of similar nucleic acid-nucleic acid interactions in both types of condensed phase.

    Notes: The branched nuclei from silk gland cells of larvae of Calpodes ethlius label with antibodies to actin and myosin and with rhodaminyl-phalloin, which is specific for f-actin. Optical sectioning localizes this actin and myosin to the nuclear periphery. Residual nuclear-associated fractions prepared from these cells contain sheets of nuclear lamina-like structures that bind heavy meromyosin and gold-tagged antibodies to actin and myosin. The results suggest that both actin and myosin, or a myosin-like protein, are components of a layer at the nucleocytoplasmic boundary that we call the nuclear shell. The nuclear shell appears to be associated with the nuclear envelope and may correspond to a zone on the cytoplasmic face of the envelope seen in electron micrographs of unextracted cells. The residual nuclear-associated fraction has a unique isoform of actin (43 kD, pl 6.45) that might allow the nuclei to associate with an actin network structurally and developmentally distinct from that of the cytoplasm. © 1992 Wiley-Liss, Inc.

    Notes: Labeling of silk glands with rhodaminyl-phalloin shows that most F-actin is restricted to parallel bundles that form rings around the gland lumen at the apical cell surface. The bundles are lost when larval feeding stops at moulting, and the F-actin is redistributed through the cytoplasm as coats to vacuoles and, occasionally, in variably oriented strands. After moulting there is a return to the distribution of filamentous actin in the apical periluminal rings of bundles. These events occur at the same time as F-actin in the nuclear shell [Henderson and Locke, submitted] undergoes its own set of changes. In silk gland cells two kinds of f-actin deployment take place concurrently.

    Notes: Parametric sensitivity of a two-dimensional pulverized-fuel (PF) combustion model is studied extensively for the effect of parametric uncertainty on model predictions. Results show that error in coal devolatilization/oxidation parameters has the dominant effect on predicted burnout, NOx formation, local gas temperature, and coal-gas mixture fraction. Uncertainty in the turbulent particle dispersion parameters appears to have a secondary effect, while error in the particle-gas radiation parameters has little impact on model predictions. Regions of the computational domain exhibiting sensitivity to specific parameters are identified. Specific parameter sensitivity implies the relative importance of various mechanisms in the overall process. Turbulent particle dispersion seems to be important early in the reactor with kinetic processes dominating at and following the predicted ignition point. Radiation appears to be of minor importance. These results indicate the need for a better method of predicting the overall volatiles yield and further understanding of the devolatilization/oxidation mechanism and its role in the overall PF combustion process. The study provides fundamental direction for future comprehensive model development and focuses on experimental work to better quantify critical input parameters.

    Notes: The von Karman analogy between heat transfer and momentum transfer in turbulent fluids is shown to apply to heat transfer through wavy liquid films in horizontal, stratified, gas-liquid flow. Nusselt numbers predicted from the analogy are shown to be in good agreement with experimental data for air-water flows involving three-dimensional wavy films and large-amplitude roll waves. The Nusselt numbers encountered with these turbulent water films are found to be in the same range as those associated with condensation of organics in horizontal tubes under stratified flow conditions.

    Notes: This study suggests that the evoution of head posture is bats is constrained by the demands of vocalization during echolocation. Nasalemitting microchiropteran taxa are easily identified by their characteristic rotation of the basicranium ventrally about the cervical axis, the depression of the rostrum below the basicranial axis, and by the rotation of the lateral semicircular canals so as to maintain their horizontal orientation during flighrt. The converse is true for oral-emitting Microchiroptera. The general form of the microchiropteran skull has been canalized along two distinct evolutionary paths, respectively, towards oral-emitting or nasal-emitting forms. © 1993 Wiley-Liss, Inc.

    Notes: Despite the widespread use of matrix-assisted laser desorption ionization, the processes involved in ion formation have yet to be satisfactorily explained. Several protonation processes have been suggested as possible methods for the creation of the analyte ions. In this work the existence of neutral hydrogen atoms in the ablation plume is reported. These are selectively ionized from the ground state through a (2+1) multiphoton resonant excitation scheme. The mass spectra at different delay times between the ablation and the post-ablation-ionization lasers have demonstrated the existence of hydrogen atoms with high velocities in the ablated plume.

    Notes: Three strain recovery techniques are investigated in the framework of the mixed iterative method. The need for appropriate strain recovery for low-order elements is shown. The three techniques compared are: C-lumping, strain-smoothing and a new technique called C-splitting. It is shown that the use of C-lumping has negative effects on the convergence of the mixed iterative method. Both strain-smoothing and C-splitting deliver convergent results, but the new technique converges faster.

    Notes: Polypropylene's physical properties (e.g., high tensile strength) and relatively inert behavior suggest that fabrication into an arterial substitute may result in an efficacious prosthesis. Grafts were woven from polypropylene yarn into conduits 4 mm I.D. × 50 mm in length. Control grafts were Dacron and ePTFE. Baseline platelet aggregometry on all dogs was performed with 10-5M ADP. Aspirin and dipyridamole were given for three days preoperatively and maintained for 2 weeks after surgery. Fifty-four grafts were placed into the aortoiliac position, two different graft materials per dog. The grafts were explanted at intervals of 2 weeks through 16 months; photographed for thrombusfree surface area determinations; and preserved for light, scanning, and transmission electron microscopy. Late (4-16 month) patency was 81% (13/16) for polypropylene, 69% (9/13) for Dacron, and 20% (1/5) for ePTFE. These data include one year patencies of 11/12 (92%) for polypropylene and 7/10 (70%) for Dacron. Late patency for polypropylene grafts was better than for PTFE (p 〈 0.05). Platelet aggregation status did not predict graft patency. Light microscopy of 2-week polypropylene explants showed inner capsules composed of myofibroblasts and macrophages, with patchy areas of endothelial cells lining the lumen. By 1 month, a confluent endothelialized surface was seen in all polypropylene explants. Progressive thickening of inner capsules with myofibroblasts and collagen continued through 4 months, reaching a mean thickness of 142 ± 50 μm compared to 150 ± 30 μm for Dacron. These findings suggest potential clinical efficacy for polypropylene as an arterial substitute. © 1992 John Wiley & Sons, Inc.

    Notes: Cultured skin fibroblasts from a patient with T-cell immune deficiency and an absence of purine nucleoside phosphorylase activity in red cells were assayed for their capacity to metabolize inosine and guanosine. The cultured fibroblasts were lacking activity of nucleoside phosphorylase and, compared to normal fibroblasts, could incorporate only 2% and 4% of 14C-inosine and 3H-guanosine, respectively, into acid precipitable material. Autoradiography visually confirmed the failure of the NP deficient cell line to incorporate the nucleosides into nuclear material. The physiological mechanism by which the deficiency of purine nucleoside phosphorylase causes T-cell dysfunction remains unclear.
