Notes: Abstract A cDNA clone (pBLT63) encoding a protein synthesis elongation factor 1α (EF-1α) was isolated from a low-temperature winter barley shoot meristem library by differential screening. The nucleotide sequence of the coding region of the low-temperature-induced barley gene shows very high homology with two EF-1α plant genes from tomato and Arabidopsis. The barley genome contains an EF-1α gene family situated on the short arm of chromosome 2 and the long arm of chromosome 5. The nucleotide sequence data reported will appear in the EMBL, GenBank and DDBJ Nucleotide Sequence Databases under the accession number Z23130.

Notes: Abstract Several low-temperature-responsive (LTR) genes from barley have been shown to have high steady-state transcript levels. Run-on transcription was used to determine the control of expression of these LTR genes. Six of these are shown to be transcriptionally regulated (blt 4/9, blt 101, blt 1015, blt 63, blt 49, blt 410) whilst three are post-transcriptionally regulated (blt 14, blt 411, blt 801). Two transcriptionally regulated genes (blt 4/9 and blt 101) and one post-transcriptionally regulated gene (blt 14) have been used in expression studies. The time course for the appearance and decay of these transcripts is given. Initial appearance and steady-state levels of individual transcripts have different temperature characteristics but no single gene correlates with the cold acclimation response. We suggest that these different response profiles may represent a means of fine-tuning the low-temperature response. One gene, blt 4/9, also accumulated high steady-state levels of transcript in response to drought and a nutrient stress. However, only drought has an acclimating effect on barley plants.

Notes: Abstract A cDNA clone of the previously unreported low-temperature-induced gene blt101 was isolated after a differential screen of a cDNA library prepared from low-temperature (6 °C day/2 °C night) grown barley shoot meristems. Southern blot analysis of barley ditelosomic addition lines was used to assign this single-copy gene to the long arm of chromosome 4. Analysis of steady-state levels of blt101 mRNA showed the induction of this transcript in shoot meristems upon transfer of barley (cv. Igri) plants from control (20 °C/15 °C) to low (6 °C/2 °C) temperature treatment. Further, the high level of this transcript is maintained at low temperatures but is reduced on transfer from low to control temperatures. The gene is not induced by drought or by foliar application of ABA. Analysis of segregating doubled haploid lines shows that there is no specific association of this gene with either spring/winter growth habit or frost hardiness. Examination of the spatial expression pattern revealed ubiquitous expression of blt101 in low-temperature (6 °C/2 °C) grown barley shoot meristems, mature leaves and roots.

Notes: Studies were made of the influence of hydrostatic pressure on the helix-coil transitions of poly (A + U) and poly (A + 2U). The results were analyzed by a thermodynamic treatment which emphasized the cooperative aspect of the transitions. The helix-to-coil volume changes were found to be small and negative indicating pressure stabilization of the coil form. The significance of the results with respect to other denaturation measurements was discussed.

Notes: The variable-temperature proton nmr spectra of the oligoribonucleotides in the series CpApX and the series ApGpX, X = A, G, C, U, together with the parent dimers CpA and ApG have been measured. A complete analysis of all the nonexchangeable base proton resonances and ribose H-1′ proton resonances was made. The presence of trends in the shielding abilities of the various bases at both the nearest-neighbor and next-nearest-neighbor positions were identified. The observed shieldings could be used to predict the chemical shifts of protons in related systems. Based on the empirical results from ribodinucleoside monophosphates, the temperature-dependent behavior of the J1′2′ coupling constants of the triribonucleotides suggested that the compounds in the CpApX series stacked from the 5′-end to the 3′-end, while those in the ApGpX series stacked from the 3′-end to the 5′-end.

Notes: Solutions of calf thymus DNA have been degraded in the presence of vibrating air bubbles in ultrasonic fields of low power which would not normally induce ultrasonic cavitation. The DNA was degraded to a limiting intrinsic viscosity, after which further irradiation by ultrasound had little or no effect. This limiting intrinsic viscosity decreased with increase in the ultrasonic intensity. Previously developed theories have-been adapted to calculate the maximum velocity gradient associated with the streaming of the solution around such vibrating air bubbles. The tensile force which is induced and which acts on the DNA has been calculated on the basis of current theories of degradation by hydrodynamic shear. These calculations indicate that the degradation of the DNA by ultrasound under conditions of “stable cavitation” is mainly the result of the shearing forces engendered in the solution around the oscillating bubbles.

Notes: Synthetic cyclic octapeptides of general structure cyclo[Glu(γOBzl)-Sar-Gly-(N-R)Gly]2 (R = n-hexyl and cyclohexyl) transport calcium ions selectively across organic phases and phospholipid membranes. We have now used proton nmr spectroscopy (360 MHz) to study the solution conformation(s) of their calcium complexes. When Ca(ClO4)2 was added to solutions of these peptides in CDCl3, nmr spectra of the resulting calcium complexes were characteristic of a single C2-symmetric conformer. From a Karplus-Bystrov analysis of vicinal coupling constants in both the peptide backbone and Glu side chain (treated as an ABCC′MX spin system), in conjuction with model-building studies, a structure was proposed in which the calcium ion is bound in an octahedral-type complex by the four (coplanar) carbonyl groups of the (all-trans) Glu-Sar and Gly-(N-R)Gly peptide bonds. Occurrence of preferred rotamers about Glu side chain Cα-Cβ bonds indicated that restricted rotation in peptide side chains arises upon calcium binding.

Notes: Copolymerization of acrylamide with allyl glycidyl ether yielded water soluble polymers containing an epoxy group; addition of a crosslinking agent, N,N′-methylenebis(acrylamide), to the copolymerization mixtures resulted in the formation of similar though water insoluble polymers. Condensation of a potent beta-adrenergic antagonist Alm—H, which contains a reactive amino group, with a polymer containing an epoxy group yeilded a pharmacologically active polymer. The polymer containing the epoxy group was also converted, by reaction with sodium thiosulfate and subsequent reduction, into a mecapto groups containing polymer which was also water soluble. The latter polymer was converted by condensation with a beta-adrenergic antagonist Alm—COCH2Br, which contains a reactive bromoacetamido group, into a pharmacologically active polymer.

Notes: An infrared method has been developed for the determination of the isotacticity of polypropylene in the 90-100% range. The method requires the annealing of thin films at 165-167°C. for 3 hr. in an inert atmosphere, followed by slow cooling to room temperature. For approximate answers the ratio of the intensity of the 10.00 μ band to that of the 10.27 μ band can be reported as the fraction of isotactic material. Better results are obtained, however, by using Figure 2 as a calibration curve and reading the values from it.

Notes: The preparation of oligocyclosiloxanes via hydrosilation reactions has been investigated. Hydrosilation reactions employing heptamethylcyclotetrasiloxane and vinyl-containing silanes and siloxanes yielded a variety of oligocyclosiloxanes with various numbers of cyclosiloxane rings connected through tri-, tetra-, penta-, hexa-, hepta-, or octafunctional junctions. The oligocyclosiloxanes were characterized by gas chromatography, IR spectroscopy, 29Si-, 1H-, and 13C-NMR spectroscopy, elemental analysis, and vapor phase osmometry. The polycyclics were redistributed with hexamethyldisiloxane and the resulting fragments analyzed by gas chromatography. All results were consistent with the formation of isomeric oligocyclosiloxanes due to a lack of absolute regioselectivity of the hydrosilation reaction. © 1993 John Wiley & Sons, Inc.
