    Notes: The Microstructural Physics group at the Cavendish Laboratory is actively involved in a considerable number of research projects which cover a broad range of materials science. In this paper, we describe briefly several such projects, with particular emphasis given to the application of parallel-detection electron energy loss spectroscopy (PEELS) on a scanning transmission electron microscope (STEM) to the analysis of materials such as stainless steels, catalysts, and high temperature superconductors. In addition, we describe a number of related projects that are currently being carried out in the group, particularly those which utilise and develop novel STEM imaging and analytical techniques. © 1993 Wiley-Liss, Inc.

    Description: Gamma-ray observations of the nearby starburst galaxies NGC 253 and M82 over the energy range (0.05-10) MeV have been obtained with the Oriented Scintillation Spectrometer Experiment (OSSE) spectrometer on the Compton Gamma-Ray Observatory (CGRO). The priority of these galaxies as OSSE targets had been established on the grounds that the average supernova rate may be high in starbursts as indicated by infrared and radio observations, and at distances of approximately 3 Mpc a significant chance of supernova gamm-ray line detection exists. NGC 253 was detected in continuum emission up to 165 keV with a total significance of 4.4 sigma and an estimated luminosity of 3 x 10(exp 40) ergs/s. The spectrum is best fit by a power law of photon index approximately 2.5. We consider the possible contribution of different emission mechanisms, including inverse Compton scattering, bremsstrahlung, discrete sources, and Type Ia/Ib supernova continuum to the measured flux. No significant continuum flux was observed from M82. A search for the gamma-ray line from the decay of the most abundant radioactive element produced in supernovae (Ni-56 yields Co-56 yields Fe-56) yielded no significant detection: the 3 sigma upper limits to the line fluxes at 0.158, 0.812, 0.847, and 1.238 MeV for both galaxies are obtained.

    Notes: Previously observed bioactivity of poly(dimethylsiloxane)-poly(ethyelene oxide)-heparin (PDMS-PEO-Hep) triblock copolymers has prompted studies of the surface and bulk character of this copolymer using angular-dependent electron spectroscopy for chemical analysis (ADESCA), static secondary mass spectroscopy (SIMS), differential scanning calorimetry (DSC), and thermogravimetric analysis (TGA). Because the low-energy PDMS phase dominates surfaces of this copolymer when solvent cast under air or vacuum conditions, attempts were made to explain surface restructuring and rearrangements induced in hydrated or aqueous environments that permit surface accessibility and bioactivity of heparin moieties. Based on comparisons with PDMS, PEO, and heparin homopolymers, PEO/heparin blends, and an unheparinized PDMS-PEO diblock copolymer, PDMS-PEO-heparin demonstrates both phase-mixed and phase-separated regions in DSC analysis. During annealing cycles above the Tg values of the copolymer constituents, phase-mixed regions become increasingly phase separated and PEO enriched. TGA analysis confirmed the presence of block copolymer constituents and presented evidence of intermolecular segmental interactions, hence phase-mixing in the copolymers. ADESCA analysis indicates that the outer 5 Å of both the PDMS-PEO and PDMS-PEO-Hep copolymers is essentially pure PDMS. However, significant amounts of PEO are detected 5 to 20 Å below the surface. Static SIMS also detects the presence of PDMS at the surfaces of the PDMS-PEO and PDMS-PEO-Hep copolymers. Compositional models based on ADESCA, SIMS, and DSC data are presented for desiccated and hydrated copolymer surfaces.

    Description: Yeast artificial chromosomes (YACs) were obtained from a 550-kilobase region that contains three probes previously mapped as very close to the locus of the fragile X syndrome. These YACs spanned the fragile site in Xq27.3 as shown by fluorescent in situ hybridization. An internal 200-kilobase segment contained four chromosomal breakpoints generated by induction of fragile X expression. A single CpG island was identified in the cloned region between markers DXS463 and DXS465 that appears methylated in mentally retarded fragile X males, but not in nonexpressing male carriers of the mutation nor in normal males.     Notes: A series of Melanin-concentrating hormone (MCH) fragments have been synthesized and their biological activities compared with the parent peptide. The substructural units, 5-14 linear and 5-14 cyclic, have been used as models for MCH -  in 1H-nmr conformational studies. Conformational features predicted by molecular dynamics analyses find support in the nmr experiments.

    Notes: Pure pencillin N α-sulfoxide (1) and penicillin N β-sulfoxide (2) were obtained by HPLC and tested as substrates for deacetoxycephalosporin C synthetase (DXCS). Neither one of the sulfoxides was utilized under conditions of conversion of penicillin N (8) to deacetoxycephalosporin C (9). The cephalosporin C α and β-sulfoxides (3 and 4, resp.) were also prepared. Relative stabilities of the sulfoxides 3 and 4 are discussed by interpretation of the 13C-NMR spectra.

    Notes: The development of gel permeation chromatography (GPC) has provided a convenient tool for the rapid determination of molecular weight distribution. The question has arisen as to the suitability of the method for specification purposes. The present work, suggested by the Naval Air Systems Command, represents an attempt to assess the precision of the method through a series of tests carried out by a number of laboratories using identical procedures on the same samples. Ten laboratories agreed to take part. Naval Ordnance Station, Indian Head, worked out standard conditions for operation of the chromatograph, for calibration of the columns, and for analysis of the GPC curves. Two samples of polystyrene were used by the various organizations for calibration of their instruments. Number-average molecular weight, heterogeneity index, and cumulative molecular weight distribution curves were determined on four samples of carboxyl-terminated polybutadiene (CTPB) and two samples of hydroxyl-terminated polybutadiene (HTPB), all unidentified except by letter code. All laboratories used identical directions for setting up CTPB and HTPB calibration curves which were based on curves determined from vapor-pressure osmometer molecular weights and GPC count numbers of fractionated material. Variation among the different laboratories was 0.15 in heterogeneity index, and a maximum of 1200 in molecular weight provided one aberrant set of values was eliminated. The six samples had heterogeneity indices from 1.15 to 1.54, while molecular weight varied from approximately 3000 to 6000. The average coefficient of variation of the molecular weight values was 6.2 ± 0.7%, which is quite acceptable. Variation in heterogeneity index was too great for specification purposes when considered among the different laboratories, but may be sufficiently good when measured by any one laboratory.

    Notes: Two independently derived distribution function methods validate both the calibration curve and the dispersion correction of the “effective linear calibration” method used in gel permeation chromatography (GPC). Experimental conditions are specified for making the method more useful by permitting linear extrapolation of the calibration line, \documentclass{article}\pagestyle{empty}\begin{document}$ V_R = C_1 + C_2 \log M, $\end{document} and for using a minimum number of standards. The independent methods quantitatively relate known differential of integral distribution functions for standard samples to their respective chromatograms. As such, they are useful calibration methods also, but are limited in scope and range.
