    Notes: Summary The properties of the novel copper enzyme nitrous oxide reductase from denitrifyingPseudomonas stutzeri are described. Multifrequency electron paramagnetic resonance spectroscopy is used to characterize the various forms of the enzyme. The features observed at 2.4, 3.4, 4.5, 9.31 and 35 GHz are explained by a mixed-valence \s[Cu(1.5)\3. Cu(1.5)\s]S=\12 species with the unpaired electron delocalized between the two Cu nuclei. This site is also present in the catalytically inactive derivative of nitrous oxide reductase which was obtained from a transposon Tn5-induced mutant with defective chromophore biosynthesis. The resemblance of the low-frequency electron paramagnetic resonance spectra to the spectra for the so-called CuA of cytochromec oxidase can be taken as a first indication that the CuA may have a structural and electronic arrangement similar to the electron-paramagnetic-resonance-detectable copper in nitrous oxide reductase. Results from oxidation/reduction experiments, and from a quantitative determination of sulfhydryl and disulfide residues in the various forms of nitrous oxide reductase, suggest the involvement of the redox-couple cysteine/cystine in the structural organization of the active site of nitrous oxide reductase.

    Notes: Summary Copper-deficient cells ofPseudomonas stutzeri strain ZoBell synthesize catalytically inactive nitrous oxide (N2O) reductase which is activated by added Cu(II) in the absence of de novo protein synthesis. The apparentK m for the activation process is 0.13 μM. Activation is temperature-dependent and is inhibited by Cd(II)(K i 1.27 μM) and less strongly by Zn(II), Ni(II), and Co(II). The same metal ions at 20 μM have little or no effect on N2O reduction of intact cells. Apo-N2O reductase of transposon Tn5-inducednos − mutants with defective Cu-chromophore biosynthesis is not reactivated by Cu(II). N2O reductase of Cu-sufficient and Cu-deficient wild type, and ofnos − mutants is localized in the periplasm, the latter providing the likely site of metal incorporation into the apoenzyme.

    Notes: Abstract Mutants with defective respiratory nitrite utilization (Nir- phenotype) were obtained by transposon Tn5 insertion into genomic DNA of the ZoBell strain of Pseudomonas stutzeri. Three representative mutants were characterized with respect to their activities of nitrite and nitric oxide reduction, cytochrome cd 1 content, and pattern of soluble c-type cytochromes. Mutant strain MK201 over-produced cytochrome c 552 about fourfold by comparison with the wild type, but possessed an in vitro functional cytochrome cd 1. Mutant strain MK202 lacked cytochrome cd 1 and, simultaneously, had low amounts of cytochrome c 552 and the split α-peak c-type cytochrome. Strain MK203 synthesized nitrite reductase defective in the heme d 1 prosthetic group. Irrespective of these biochemically distinct Nir- phenotypes, all mutants preserved the nitric oxidereducing capability of the wild type. The mutant characteristics demonstrate that cytochrome cd 1 is essential for nitrite respiration of P. stutzeri and establish the presence of a nitric oxide-reducing system distinct from cytochrome cd 1. They also indicate the functional or regulatory interdependence of c-type cytochromes.

    Notes: Abstract The novel multicopper enzyme nitrous oxide reductase from Pseudomonas perfectomarina was purified to homogeneity to study its properties and distribution in various pseudomonads and other selected denitrifying genera by immunochemical techniques. Quantitation of immunochemical crossreactivity by micro-complement fixation within the denitrifying pseudomonads of Palleroni's ribosomal ribonucleic acid group I corresponded to the taxonomic positions established by nucleic acid hybridization. The assignment of P. perfectomarina to the stutzeri-group (as strain ZoBell) was consolidated by immunochemical crossreactivity based on nitrous oxide reductase. Crossreactivity of nitrite reductase (cytochrome cd 1) with a respective P. perfectomarina rabbit antiserum was limited to strain DSM 50227 of P. stutzeri; although it could not contribute information towards broader relationships within rRNA group I, it lent further prove to the unity of these two species.

    Notes: The classical potential formulation of inviscid transonic flows is modified to account for non-isentropic effects. The density is determined in terms of the speed as well as the pressure, which in turn is calculated from a second-order mixed-type equation derived via differentiating the momentum equations.The present model differs in general from the exact inviscid Euler equations since the flow is assumed irrotational. On the other hand, since the shocks are not isentropic, they are weaker and are placed further upstream compared to the classical potential solution. Furthermore, the streamline leaving the aerofoil does not necessarily bisect the trailing edge.Results for the present conservative calculations are presented for non-lifting and lifting aerofoils at subsonic and transonic speeds and compared to potential and Euler solutions.

    Notes: A velocity-vorticity formulation of the Navier-Stokes equations is presented as an alternative to the primitive variables approach. The velocity components and the vorticity are solved for in a fully coupled manner using a Newton method. No artificial viscosity is required in this formulation. The pressure is updated by a method allowing natural imposition of boundary conditions. Incompressible and subsonic results are presented for two-dimensional laminar internal flows up to high Reynolds numbers.

    Notes: A method is outlined for solving two-dimensional transonic viscous flow problems, in which the velocity vector is split into the gradient of a potential and a rotational component. The approach takes advantage of the fact that for high-Reynolds-number flows the viscous terms of the Navier-Stokes equations are important only in a thin shear layer and therefore solution of the full equations may not be needed everywhere. Most of the flow can be considered inviscid and, neglecting the entropy and vorticity effects, a potential model is a good approximation in the flow core. The rotational part of the flow can then be calculated by solution of the potential, streamfunction and vorticity transport equations. Implementation of the no-slip and no-penetration boundary conditions at the walls provides a simple mechanism for the interaction between the viscous and inviscid solutions and no extra coupling procedures are needed. Results are presented for turbulent transonic internal choked flows.

    Notes: Finite element solution methods for the incompressible Navier-Stokes equations in primitive variables form are presented. To provide the necessary coupling and enhance stability, a dissipation in the form of a pressure Laplacian is introduced into the continuity equation. The recasting of the problem in terms of pressure and an auxiliary velocity demonstrates how the error introduced by the pressure dissipation can be totally eliminated while retaining its stabilizing properties. The method can also be formally interpreted as a Helmholtz decomposition of the velocity vector.The governing equations are discretized by a Galerkin weighted residual method and, because of the modification to the continuity equation, equal interpolations for all the unknowns are permitted. Newton linearization is used and at each iteration the linear algebraic system is solved by a direct solver. Convergence of the algorithm is shown to be very rapid. Results are presented for two-dimensional flows in various geometries.

    Notes: The application of the Trefftz method for calculating wave forces on offshore structures is presented. Indirect and direct formulations using complete and non-singular systems of Trefftz functions for the Helmholtz equation are posed in this paper. An effective technique using different interpolation functions for the velocity potential and wave force are suggested to improve the computational accuracy of the wave force. The numerical examples show that the present method is highly efficient and accurate.
