    Notes: Abstract A Nafion® matrix was used to prepare a small and simple oxygen sensor whose sensing electrode and counter electrode were made by r.f. sputtering of platinum deposits on both sides of the Nafion® membrane. The sensing current varied linearly with oxygen partial pressure. This sensor enabled an estimation of oxygen permeation in coated films at working conditions. The present method clarifies the correspondence between the permeation rate through the clear layer and the degree of deterioration of underlayered paint film.[/p]

    Notes: Summary The activity of membrane-bound dipeptidyl aminopeptidase IV (EC 3.4.14.5) was found to be markedly reduced in the Fischer 344 rat strain compared with that in the Wistar strain. Analysis of membrane proteins by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and Ouchterlony immunodiffusion revealed the specific absence of this enzyme molecule in the Fischer 344 strain.

    Notes: Summary Lysosomal acid hydrolases were determined in established lymphoblastoid cell lines, transformed in vitro by Epstein-Barr virus (EBV) from lymphocyte-rich cell populations isolated from the peripheral blood of patients with genetic lysosomal storage diseases—Hurler syndrome, Scheie syndrome, GM1-gangliosidosis type 1 and type 2, Tay-Sachs disease, and I-cell disease—and from obligate heterozygotes for these diseases. The respective enzyme activity was undectectable in lymphoblastoid cells from the patients, but not from controls. Obligate heterozygotes could not always be distinguished from controls in lymphoblastoid cells as well as in leukocytes. These results suggest that established lymphoblastoid cell lines are useful material for the enzymatic study of genetic lysosomal storage diseases.

    Notes: Summary A computerized system was devised to trace the attachment, growth and disappearance of peritrich ciliates in an urban river. By tracing the development of each Carchesium polypinum colony in the river, it was possible to estimate the actual growth rate of this species without considering the effect of immigration. The survival and colonization rates of the colonies and individuals of solitary species could also be estimated. C. polypinum showed high growth rates (r=1.370 day-1, doubling time 12.14 h). The number of daily colonizers also increased at a high rate, and the combination of growth and colonization caused very high population increase rates at the area level. Because of its low survivorship, the rate of Vorticella microstoma increase was much lower than previously reported. The survival of solitary species and small colonies was lower than with large colonies, and fewer new colonizers survived with the development of the attached microbial community.

    Notes: Abstract Temporal dynamics of a lentic microphagotroph community were studied during leaf litter decomposition from December to May. Small plastic vessels containing leaf litter were placed on a pond bottom. They were sampled periodically to collect microphagotrophs. Three abiotic factors and abundance of two food items were also measured to analyze the autogenic and allogenic phenomena during a microphagotroph succession. Three behavior types were recognized in dominant taxa: a free-swimming type, a vagile (creeps on substratum, sometimes swims) type, and a voluntarily fixed type. Dominant taxa changed from the free-swimming to the vagile type up to mid-March, and the reverse change occurred from mid-April. Principal component analysis (PCA) indicated four factors affecting the dynamics of the community: water temperature as a seasonal factor, detritus volume on the litter surface as a habitat factor, and densities of bacteria and small flagellates as food factors. Taxa replacement appeared to occur through two mechanisms. (1) Dominance of small holotrichs, a free-swimming type, was brought about by a high bacterial density caused by seasonal events, i.e., leaf fall in December and detritus formation by litter feeders in mid-April. This is an allogenic aspect of community dynamics. (2) The free-swimming type was replaced by the vagile one during the period with high taxa diversity. This replacement occurred through intertaxa competition for scarce food and/or selective predation by larger microphagotrophs. It is an autogenic process within the community.

    Notes: Abstract The growth kinetics and martensitic transformation induced by simple cooling of large Fe particles in Cu-1.5 mass % Fe single crystals aged at 973 K for up to 60 days are examined by transmission electron microscopy and magnetic measurement. The growth kinetics of large Fe particles can be described by the Lifshitz-Slyozov-Wagner theory and the interfacial energy between the Fe particles and the Cu matrix is estimated as 0.24 to 0.40 J m−2. The volume fraction of α-Fe particles increases with the mean particle size and with lowering of the cooling temperature. The critical sizes below which the transformation by simple cooling does not occur, are found to be 290 and 350 nm for specimens cooled to 77 K and room temperature, respectively. The saturation magnetization of the Cu-Fe alloy containing large α-Fe particles increases by further annealing of the cooled specimens. These facts imply that α-Fe particles are not produced during the prolonged ageing but are formed martensitically during the cooling process.

    Notes: Summary In order to investigate the mechanism of synthesis and secretion of lysozyme (LZ) by human mononuclear phagocytes, the ultrastructural localization of LZ was studied by a pre-embedding direct immunoperoxidase method. Blood monocytes showed a reaction product for LZ in cytoplasmic granules, whereas cultured monocytes showed the reaction product in phagosomes as well as granules at 5 h of culture and in numerous large granules at 3 days of culture. In Kupffer cells, LZ was present in cytoplasmic granules, vacuoles and phagosomes. Some Kupffer cells showed a positive reaction for LZ in the rough endoplasmic reticulum, perinuclear cisterna and Golgi apparatus. Macrophages in the lymph nodes contained LZ in cytoplasmic granules. Bone marrow macrophages contained numerous phagosomes with electron-dense degradation products of erythrocytes, but the reaction product for LZ could not be clearly identified. The present study demonstrated that LZ is present in the granules of human mononuclear phagocytes and released into phagosomes. An in-vitro culture study, furthermore, demonstrated that macrophages produce LZ-containing large granules distinct from those of monocytes. However, findings that indicate the synthesis and secretion of LZ by cultured monocytes, as suggested previously by other investigators, were not observed in this study.

    Notes: Summary Cellular proliferation in the anterior pituitary of 2-, 8-, 15- and 30-day-old rats was examined by injection of bromodeoxyuridine 1 h before autopsy. Bromodeoxyuridine incorporated into DNA was detected immunohistochemically by use of a monoclonal antibody. The highest rate of cell proliferation was found in 2-day-old animals; it decreased thereafter during the postnatal period. Possible toxic effects of colchicine on cellular proliferation were examined. Colchicine treatment (10 mg/kg in 8- and 30-day-old animals) significantly decreased the number of bromodeoxyuridine-labelled cells/mm2 in 8-day-old rats. Some sections were doubly immunostained for bromodeoxyuridine and various pituitary hormones. The proportion of doubly-immunostained cells to all proliferating cells was generally low, ranging from 23% at 2 days to 32% at 30 days of age.
