    Notes: Abstract To examine the influence of the spectral characteristics of underwater light on spectral sensitivity of the ON and OFF visual pathways, compound action potential recordings were made from retinal ganglion cells of threespine stickleback from different photic regimes. In fish from a red-shifted photic regime (λP50 680 nm for downwelling light at 1m), peak sensitivity of both the ON and OFF pathways was limited to long wavelength light (λmax 600–620). In contrast, the ON pathway of fish from a comparatively blue-shifted (λP50 566 nm) photic regime exhibited sensitivity to medium (λmax 540–560) and long (λmax 600 nm) wavelengths, while the OFF pathway exhibited peak sensitivity to only medium (λmax 540 nm) wavelength light. In a third population, where the the ambient light is moderately red-shifted (λP50 629 nm), the ON pathway once again exhibited only a long wavelength sensitivity peak at 620 nm, while the OFF pathway exhibited sensitivity to both medium (λmax 560 nm) and long (λmax 600–620 nm) wavelength light. These findings suggest that the photic environment plays an integral role in shaping spectral sensitivity of the ON and OFF pathways.

    Notes: Abstract Chromosome 3 displayed the two largest yield QTLs in a previous study of 150 doubled haploid lines derived from a cross of Steptoe and Morex barley varieties. Low-copy number RFLP markers, detected using Southern analysis, are excellent tools for generating robust linkage maps as demonstrated by the Steptoe and Morex map produced by the North American Barley Genome Mapping Project (SM NABGMP). However, this technique can be cumbersome when applied to practically oriented plant breeding programs. In the present report, we demonstrate the conversion of RFLPs to more practically useful PCR-based markers that are co-dominant and allelic to the barley chromosome-3 RFLP markers from which they derive. We have used these sequence-tagged-site (STS) PCR markers to evaluate the putative yield QTL components of the Steptoe chromosome 3 in a Morex backcross population. Headshattering, plant lodging, and yield measurements are reported from five replicated field experiments conducted under diverse growing conditions in Montana. Our study detected significant effects for all three traits in a chromosomal region that evidently corresponds to the larger of the two previously reported chromosome-3 QTLs. However, we failed to detect any yield or other effects which might be coincidental to the second largest yield QTL. The genetic effects of the yield QTL identified in our first backcross breeding population show similar magnitude, environmental interactions, and association with lodging and headshattering QTLs observed in the SM NABGMP experiments. Our study elucidates complex environmental conditioning for headshattering and plant lodging which probably underlie the variable yield effects observed under different growing conditions.

    Notes: Abstract Introgression of chromosomal segments across large taxonomic distances has long been an objective of scientists interested in understanding the relationships between genes and their effect on phenotype. Barley and wheat represent cultivated members of the Triticeae with different zones of adaptation, different responses to pathogens, and different end-use characteristics. Introduction of small, well-characterized chromosomal segments among grass relatives presents an opportunity to both better understand how genes perform in novel genomic environments and to learn more about the evolutionary novelties which differentiate related species. Since the distribution of the wheat-barley addition lines, the potential power and value of a comprehensive series of wheat/barley translocation lines has been widely appreciated. A scarcity of easy-touse markers which unambiguously distinguish barley loci from their wheat homologues has limited the ability of scientists to identify the relatively rare inter-chromosomal recombination events which are the necessary antecedents of these lines. Since the single most critical pathogen affecting U.S. wheat producers is Karnal bunt (Tilletia indica) and since barley carries a gene conferring immunity, molecular markers may prove practically and immediately important. In this report we describe a series of 135 barley-specific markers amplified by 115 primer sets developed from sequences from previously mapped restriction fragment length polymorphism (RFLP) markers. These easily distinguish the cognate barley products from their wheat counterparts and should find ready use in the identification of lines which contain wheat/barley translocation events.

    Description: Current assays of human committed-stem cells are of limited value in predicting the rate of engraftment or in assessing the integrity of the stem cell pool after allogeneic bone marrow (BM) transplantation (BMT). We have used a limiting dilution assay of mafosfamide-resistant progenitors (pre-colony-forming units [CFU]), which are ancestral to committed progenitors such as CFU-granulocyte-macrophage (GM) to analyze the kinetics of myeloid engraftment after BMT and to assess the size of the stem cell pool at intervals up to 66 months thereafter. In 24 patients transplanted for chronic myeloid leukemia in chronic phase (eight with matched unrelated donors and 16 with sibling donors), the rate of neutrophil engraftment correlated strongly with the number of pre-CFU transfused per kilogram recipient body weight (r = .7, P 〈 .005) but not with CFU-GM per kilogram or nucleated cells per kilogram. In 25 patients studied 6 to 66 months after allogeneic BMT, the mean number of pre-CFU in the marrow was 3.1/10(5) mononuclear cells (MNC) (median, 3.47; range, 0.4 to 23.3), compared with 24.7/10(5) MNC (median, 27.3; range, 4.2 to 180) in 25 normal subjects. CFU-GM were also reduced in these patients, but with considerable overlap into the normal range (mean +/- SD: 54 +/- 45.6 per 10(5) MNC; normal, 129 +/- 61.6). Low pre-CFU but not low CFU-GM levels were associated with reduced peripheral blood white blood cell counts in post-BMT patients. Pre-CFU and CFU-GM levels were not related to the interval posttransplant and remained low for up to 66 months. We conclude that the pre-CFU assay measures a population of stem/progenitor cells that are important in the kinetics of engraftment after allogeneic BMT. Our data suggest that pre-CFU levels may remain low for some years after BMT in humans.

    Description: Two 214.5 cm. long high performance periodic (26 cm period) permanent magnet half-assemblies were designed and constructed for use as a wiggler using Nd-B-Fe and vanadium permendur as hard and soft magnetic materials by Field Effects, a division of Intermagnetics General Corporation. Placing these assemblies in a supporting structure with a 2.1 cm pole to pole separation resulted in a periodic field with a maximum value of 2.04 T. This is believed to be the highest field ever achieved by this type of device. The attractive force between the two 602 kg magnet assemblies is 228 kN, providing enough force for suspension of a 45,500 kg vehicle. If used in an attractive maglev system with an appropriate flat iron rail, one assembly will generate the same force with a gap of 1.05 cm leading to a lift to weight ratio of 38.6, not including the vehicle attachment structure. This permanent magnet compares well with superconducting systems which have lift to weight ratios in the range of 5 to 10. This paper describes the magnet assemblies and their measured magnetic performance. The measured magnetic field and resulting attractive magnetic force have a negative spring characteristic. Appropriate control coils are necessary to provide stable operation. The estimated performance of the assemblies in a stable repulsive mode, with eddy currents in a conducting guideway, is also discussed.

    Description: The purpose of this study was to generate design data and complete dynamic performance estimates for a high performance permanent magnet actuator. The basic configuration selected for analysis is an axisymmetric Nd-B-Fe permanent magnet actuator capable of providing force in one direction along its major axis. The actuator consisted of two main axisymmetric components separated by an air gap. The design was optimized for each value of force, gap and magnetic field to yield minimum weight and maximum lift to weight ratio. The basic conclusion is that, within parameters considered, the 10,000 lb. and 50,000 lb. actuators are lightweight and compact. As expected for most permanent magnet devices, the smaller ones have higher lift to eight ratios.
