Notizen: The usefulness of artificial neural networks (ANN) for milk shelf-life prediction by multivariate interpretation of gas chromatographic profiles and flavor-related shelf-life was evaluated and compared to principal components regression (PCR). The training set consisted of dynamic headspace gas chromatographic data collected during storage of pasteurized milk (input information for the neural network used to make a decision) and its corresponding shelflife (prediction or response). ANN had better predictability than PCR. A standard error of the estimate of 2 days in shelf-life resulting from regression analysis of experimental vs predicted values indicated a high predictability of ANN.

Notizen: Neural networks (NN) provide a simple means of predicting outcomes that depend upon complex, possibly nonlinear, relationships between many variables. A trained neural network was created and used to predict loaf volume of breads made from different wheat cultivars. Although creating the NN required specialized skills and considerable computational time, using the “trained” NN to estimate remix loaf volume, was very rapid and required only basic computer skills. Random Centroid Optimization (RCO) was also employed to choose the best training parameters: learning rate = 0.820, smoothing factor = 0.123, noise = 0.056, number of hidden neurons = 5. NN was more accurate, faster and easier than Principal Component Regression Analysis.

Notizen: A water-soluble fraction (WSF) was prepared by filtering 10 times diluted yolk (pH 5.0) stored at 4–6°C through Whatman No. 52 or No.1 filter paper for delipidation. However, solid-phase delipidation using an octadecyl column after filtration through a cellulose powder column was more practical for industrial application. After ultrafiltration (UF) of the WSF (pH 9.0,1.5M NaCl), results showed: Amicon 80–85% recovery, 74–99% purity; Harp 72–74% recovery, 81–84% purity; A/G 75% recovery, 89% purity; Koch 88% purity.

Notizen: A serial filtration system was developed for separating immunoglobulin from egg yolk (IgY). Egg yolk was diluted 10 times with water, adjusted to pH 5.0, frozen, then thawed. The diluted yolk was diafiltered through a hard filter paper at 4-6°C to produce a clear water-soluble fraction (WSF). The WSF was then delipidated by filtering through a hydrophobic membrane filter. Ultrafiltration of delipidated WSF at pH 9.0, at 4-6°C, in the presence of 1.5M NaCl resulted in electrophoretically pure (〉90%) IgY with a recovery of about 92%. Advantages of this method are the use of only a few mild chemicals and the potential of developing a continuous process.

Notizen: An economical, small scale heat processing device was studied for rapid and efficient pasteurization of human milk while preserving immunoglobulins A,G and M. Using a continuous flow HTST milk pasteurizer, human milk inoculated with E. coli (106 CFU/mL) or S. aureus (107 CFU/mL) was heated at 71°C at flow rates of 5.9, 12.3 and 18.9 mL/ min. All conditions completely inactivated both microorganisms and resulted in negative alkaline phosphatase activity indicating complete pasteurization. Heat processing of bovine milk at 71°C, at 5.9 mL/min resulted in retention of 30% of the γ-glutamyl transpeptidase (GGTP) activity. Lower residual activity could therefore indicate overpasteurization. Pasteurization at 71°C for 9.0 sec (12.3 mL/min) resulted in retention of 74% of IgA, 75% IgG, and 68% IgM.

Notizen: Abstract  Global warming is increasingly recognized as a threat to the survival of human beings, because it could cause a serious increase in the occurrence of diseases due to environmental heat during intermittent hot weather. To assess the direct impact of extremely hot weather on human health, we investigated heat-related deaths in Japan from 1968 through 1994, analyzing the data to determine the distribution of the deaths by age and their correlation to the incidence of hot days in summer. Vital Statistics of Japan, published by the Ministry of Health and Welfare of Japan, was the source of the heat-related mortality data employed in this study. Meteorological data were obtained from the District Meteorological Observatories in Tokyo and Osaka, the two largest cities in Japan. Heat-related deaths were most prone to occur on days with a peak daily temperature above 38°C, and the incidence of these deaths showed an exponential dependence on the number of hot days. Thus, even a small rise in atmospheric temperature may lead to a considerable increase in heat-related mortality, indicating the importance of combating global warming. Furthermore, half (50.1%) of the above-noted deaths occurred in children (4 years and under) and the elderly (70 years and over) irrespective of gender, indicating the vulnerability of these specific age groups to heat. Since a warmer climate is predicted in the future, the incidence of heat waves will increase, and more comprehensive measures, both medical and social, should be adopted for children of 4 years and younger the elderly to prevent heat-related deaths in these age groups.

Notizen: Abstract Autoimmune-prone lpr mice develop lymphoproliferative disorders, whereas their lymphocytes show accelerated apoptosis in culture. To elucidate whether the bcl-2 protein, a repressor of apoptosis, is critical to the discrepancy between in vivo and in vitro survival, we examined bcl-2 expression in T cells from +/+ and lpr mice during culture. The expression levels of bcl-2 in cultured T cells from lpr mice were significantly down-modulated compared to those from +/+ mice and freshly obtained T cells. Besides, the reduction of bcl-2 protein levels was inhibited in T cells cultured in the presence of T cell receptor (TCR) signalling. These results suggest that lpr T cells might be susceptible to apoptosis in vitro due to down-modulation of bcl-2 by withdrawal of TCR signalling.

Notizen: A finite element method (FEM) system is complex in nature and still faces bottleneck problems in its maintenance, extension, etc., and it is yet necessary to be dealt with using some new methodologies. An object-orientation is a promising paradigm for treating complexities. In this paper, first an object-oriented FEM knowledge base system architecture is proposed. Then, an object model in the FEM domain is established through entity abstractions, action abstractions, category abstractions, agent abstractions, etc. Finally, a semantic network results from a semantic analysis of the FEM object model to represent the generative relationships among the FEM objects. Through an illustrative example, it is shown that a control task of finite element elasto-static analysis can be represented by a traveling path in the semantic network.
