Beschreibung: The textbook description of mitochondrial respiratory complexes (RCs) views them as free-moving entities linked by the mobile carriers coenzyme Q (CoQ) and cytochrome c (cyt c). This model (known as the fluid model) is challenged by the proposal that all RCs except complex II can associate in supercomplexes (SCs). The proposed SCs are the respirasome (complexes I, III, and IV), complexes I and III, and complexes III and IV. The role of SCs is unclear, and their existence is debated. By genetic modulation of interactions between complexes I and III and III and IV, we show that these associations define dedicated CoQ and cyt c pools and that SC assembly is dynamic and organizes electron flux to optimize the use of available substrates.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Lapuente-Brun, Esther -- Moreno-Loshuertos, Raquel -- Acin-Perez, Rebeca -- Latorre-Pellicer, Ana -- Colas, Carmen -- Balsa, Eduardo -- Perales-Clemente, Ester -- Quiros, Pedro M -- Calvo, Enrique -- Rodriguez-Hernandez, M A -- Navas, Placido -- Cruz, Raquel -- Carracedo, Angel -- Lopez-Otin, Carlos -- Perez-Martos, Acisclo -- Fernandez-Silva, Patricio -- Fernandez-Vizarra, Erika -- Enriquez, Jose Antonio -- New York, N.Y. -- Science. 2013 Jun 28;340(6140):1567-70. doi: 10.1126/science.1230381.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉Centro Nacional de Investigaciones Cardiovasculares Carlos III, Madrid, Spain.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/23812712" target="_blank"〉PubMed〈/a〉

Notizen: Resolution in chromatographic separations can be compromised by nonuniform flow distribution, heterogeneous packing, or flow instabilities such as viscous fingering. In this work, the displacement of solutes within a chromatographic column was monitored with an advanced magnetic-resonance technique, echo-planar spectroscopic imaging (EPSI). Unlike conventional magnetic-resonance imaging, this approach allows multiple solutes to be observed simultaneously, providing separate “chemical images” for each component. Qualitative chemical images were obtained in approximately 1 min with approximately 1 mm3 spatial resolution. EPSI allowed us to examine the multi-component viscous fingering problem for the first time. While many features of the instability are preserved, solute retention appears to slow the growth of solvent fingers.

Notizen: The idea of separating the free energy of mixing into two contributions (corresponding to the “physical” or “nonspecific” and to the “specific” interactions, respectively) proposed by Painter and Coleman has been extended to ternary polymer/polymer/polymer blends. In this paper experimental phase diagrams of three ternary blends have been reproduced by estimating both contributions for the interaction energy density (Bij) which characterizes each polymer pair. In contrast to the Painter-Coleman Miscibility Guide rules, we have found that, in some cases, “specific” contributions positive and unfavorable to the mixing, due to the self-association of one of the components, can describe more adequately the experimental trends.

Notizen: The phase behavior of polymer blends under simple shear flow has been studied using a custom-designed rheo-optical system consisting of a two-dimensional small-angle light scattering (SALS) device incorporated into a conventional rheometer. Two-dimensional SALS images were gathered for model polymer blend systems with different quiescent phase behavior: polystyrene/polyisobutylene (PS/PIB) that exhibits upper critical solution temperature phase behavior and polystyrene/poly(vinyl methyl ether) (PS/PVME) that shows lower critical solution temperature phase behavior. For the PS/PIB blend, shear-induced phase mixing occurred at a critical shear rate. Below that critical shear rate, the dispersed phase was highly elongated parallel to the flow direction. For PS/PVME blends, a streak scattering pattern was observed even though the sample became optically clear after shearing. We observed, apparently for the first time, the development of a bright-streak pattern from a transient dark-streak pattern for a polymer blend system under shear. Rheo-microscopy studies revealed an intriguing wave pattern that developed coincident with the observation of a streak pattern by SALS. The relationship between the two phenomena has not yet been established.

Notizen: The direct study of polymer Interfaces is very difficult. Small angle scattering methods have been most successfully applied to block copolymer Interfaces where the morphology is well known. Other techniques such as electron microscopy, Rutherford back scattering, and forward recoil spectroscopy generally only give resolution of the order of 10 nm. Here we describe two techniques which have only recently been applied to polymer interfaces, neutron reflection, and spectro-scopic ellipsometry. Both reflection techniques examine planar samples and give compositional information in the depth direction. Neutron reflection gives a resolution of 0.5 nm and spectroscopic ellipsometry may give better than 5 nm. We will describe experiments using neutron reflection which measure the inter-facial thickness between two immiscible polymers and experiments using both techniques which measure the interdiffusion of miscible polymers. Other applications and limitations of the techniques will also be discussed.

Notizen: A slit die viscometer (SDV) was built and evaluated extensively. A major advantage of the SDV is the ability to measure in-line rheological data in a continuous fashion, using a setup that most closely approximates the conditions encountered in a real extrusion process. Comparisons will be presented of viscosity data of the SDV to data from a capillary rheometer (CR) and a Rheometrics Mechanical Spectrometer (RMS). Viscosity values as measured on the SDV tend to be lower than those measured on the CR and RMS. Possible reasons for this disagreement will be discussed. The effect of temperature on viscosity, the effect of pressure on viscosity, and the effect of compressibility will be analyzed in detail. It will be shown that these effects can be substantial, particularly with certain types of polymers. Finally, the feasibility of using the slit die viscometer to determine first normal stress differences will be explored.
