    Notes: Reduction of a bis-radical precursor, 6-phenyl-1,1-bis[3-(phenylselanyl)propyl]-3a,4-dihydro-1H,3H-cyclopenta[c]furan-5-one (6), with 3 equiv. of Ph3SnH provides mixtures of cis,cis- or cis,trans-angular triquinane products (3aα,5aβ,8β,8aR*)- and (3aα,5aα,8β,8aR*)-hexahydro-3a-propyl-8-phenyl-5H-dicyclopenta[b,c]furan-7(8H)-one (cis,cis-12/cis,trans-12), in yields that vary from 50%/50% to 91%/6% depending on the reaction concentration. A mechanistic model for this process is proposed that involves a non-selective phenylselenium-group abstraction step followed by successive kinetic resolutions of diastereoisomeric radical intermediates. This reaction shows how yields in group-selective reactions can be compounded to levels above that ostensibly permitted by the level of the group-selective step.

    Notes: TADDOLs (α,α,α′,α′,-tetraaryl-1,3-dioxolane-4,5-dimethanols) with dendritic branches attached to the aryl groups and with 8 or 16 peripheral styryl double bonds have been synthesized (4 and 5). With these compounds, dendritic molecules were employed for the first time as cross-linkers in a polymerization. The resulting polystyrene was loaded with titanate (Ti(OCHMe2)4) to generate polymer beads incorporating Ti-TADDOLate centers (Figs. 1 and 2) for enantioselective catalysis (Et2Zn addition to PhCHO). Compared with conventionally polymer-attached, insoluble Ti-TADDOLates (cf. 3), the new materials have a much higher catalytic activity, rather close to that of soluble analogs (Fig. 3).

    Notes: A general approach to the synthesis of enantiomerically pure α-phenyl amino alcohols via the asymmetric reduction of α-phenyl haloalkyl ketones or α-phenyl aminoalkyl ketones with B-chlorodiisopinocampheylborane is described. Using this approach, an improved synthesis of a potential antipsychotic, α-(4-fluorophenyl)-4-(2-pyrimidinyl)-1-piperazinebutanol in ≥98% ee, and the broncholdilator 1-(2-methoxy-2-phenethyl)-4-(3-hydroxy-3-phenylpropyl)piperazine (eprozinol) in ≥99% ee is achieved. © 1995 Wiley-Liss, Inc.

    Notes: Intramolecular overcrowding in bistricyclic enes (1) requires out-of-plane deformations in order to accommodate the sterically demanding tricyclic moieties without prohibitively close contacts of nonbonded atoms. The nonplanarity of 1 introduces the notion of chirality to the arena of overcrowded aromatic enes. Deviation from coplanarity occurs by twisting around the double bond and out-of-plane bending (hence pyramidalization). The bending is realized by folding of the tricyclic moieties. The conformations of homomerous (1, × = Y) and heteromerous (1, × ≠ Y) bistricyclic enes and their mono- and disubstituted derivatives are analyzed along with their point group symmetries and chirality. The implications are illustrated using the structures of characteristic examples calculated with the semiempirical method PM3. © 1995 Wiley-Liss, Inc.

    Notes: Enantiopure levodropropizine can be synthesized in high yield by the direct nucleophilic addition of 1-phenylpiperazine to (R)-glycidol followed by recrystallization in absolute ethanol. A chiral HPLC method has been developed for the determination of its optical purity. The enantiomeric enrichment behavior of this compound in the recrystallization process is discussed according to its binary melting point phase diagram. Single crystal x-ray structural analysis shows the intermolecular hydrogen-bonding interactions between the hydroxyl and piperazinyl groups within the crystal lattice of levodropropizine. © 1996 Wiley-Liss, Inc.

    Notes: The enantiomeric resolution of a series of N-arylamides was examined on amylose tris[(S)-1-phenylethylcarbamate] coated onto aminopropylated 7 μm silica with 500 Å diameter pores and on naked silica 5 μm particle size with 500 Å diameter pores. The enantiomeric resolution obtained for this series was excellent on both columns. The enantioselectivity of cellulose and amylose tris (3,5-dimethylphenylcarbamate) coated onto APS-Hypersil (120 Å pore size, 5 μm particle size) was also investigated for this series of compounds.     Notes: Reboxetine, (RS)-2-[(RS)-α-(2-ethoxyphenoxy)benzyl]morpholine methanesulphonate, is a racemic compound and consists of a mixture of the (R,R)- and (S,S)-enantiomers. The pharmacokinetics of reboxetine enantiomers were determined in a crossover study in three male beagle dogs. Each animal received the following oral treatments, separated by 1-week washout period: 10 mg/kg reboxetine, 5 mg/kg (R,R)- and 5 mg/kg (S,S)-. Plasma and urinary levels of the reboxetine enantiomers were monitored up to 48 h post-dosing using an enantiospecific HPLC method with fluorimetric detection (LOQ: 1.1 ng/ml in plasma and 5 ng/ml in urine for each enantiomer). After reboxetine administration mean tmax was about 1 h for both enantiomers. Cmax and AUC were about 1.5 times higher for the (R,R)- than for the (S,S)-enantiomer, mean values ± SD being 704 ± 330 and 427 ± 175 ng/ml for Cmax and 2,876 ± 1,354 and 1,998 ± 848 ng.h/ml for AUC, respectively. No differences between the (R,R)- and (S,S)-enantiomers were observed in t½ (3.9 h). Total recovery of the two enantiomers in urine was similar, the Ae (0-48 h) being 1.3 ± 0.7 and 1.1 ± 0.7% of the enantiomer dose for the (R,R)- and the (S,S)-enantiomers, respectively. No marked differences in the main plasma pharmacokinetic parameters were found for either enantiomer on administration of the single enantiomers or reboxetine. No chiral inversion was observed after administration of the separate enantiomers, as already observed in humans. Chirality 9:303-306, 1997. © 1997 Wiley-Liss, Inc.
