    Notes: Abstract Macroscopic instantaneous and time-dependent currents have been measured in the vacuolar membrane of Beta vulgaris using a patch clamp configuration analogous to whole cell mode. At low cytosolic Ca2+ and in the absence of Mg2+, only an instantaneous current was observed. This current is carried predominantly by cations (PK∶PCl 7∶1, pna∶pcl 4∶1 and arginine is also conducted). The instantaneous current can be activated by ATP4− (e.g., ATP-activated mean K+ current density was −20 mA.m−2 at a membrane voltage of −20 mV) and by increasing cytosolic pH and Mg2+ (raising Mg2+ from 0 to 0.4 mm induced a mean current density increase of −7 mA.m−2 at −20 mV). Such current can be activated by simultaneous addition of putative in vivo concentrations of ATP4−/MgATP/Mg free 2+ (in the presence of bafilomycin to inhibit the vacuolar ATPase) and further modulated by cytosolic pH. With vacuolar K+ concentration greater than that of the cytosol, activation of the instantaneous current would mediate vacuolar K+ release over the range of physiological membrane voltage. It is argued that the ATP4−-activated current, in addition to acting as a K+ mobilization pathway, could provide a counter-ion (shunt) conductance, allowing the two electrogenic H+ pumps which reside in the vacuolar membrane to acidify the vacuolar lumen. A separate time-dependent current, which was not observed at low Ca2+ concentrations (less than 500 nm) could also be elicited by addition of Mg2+ at the cytoplasmic membrane face. This current was stimulated by increasing cytoplasmic pH.

    Notes: Abstract  The persistence of metapopulations is likely to be highly dependent on whether population dynamics are correlated among habitat patches as a result of migration between patches and spatially-correlated environmental stochasticity (weather effects). We examined whether population dynamics of the ringlet butterfly, Aphantopus hyperantus, were synchronous in an area of approximately 0.5 km2, with respect to extinction, colonization and population fluctuations. Monks Wood Butterfly Monitoring Scheme transect count data from 1973 to 1995, revealed (A) a major environmental perturbation, the drought of 1976, which caused synchronized extinctions of A. hyperantus in subsequent years, (B) synchronized recolonization in years following the large number of apparent extinctions, and (C) population changes by A. hyperantus were highly correlated in many of the 14 sections of the transect, presumably reflecting similar responses to environmental stochasticity, and the exchange of individuals among sections. However, extinction and population synchrony depended on habitat type. Following the 1976 drought, A. hyperantus apparently became extinct from the most open and most shady habitats it occupied, with some persistence in habitats of intermediate shading, thus showing retraction to core populations in central parts of an environmental gradient, albeit with an average shift to relatively open habitat. Populations at extreme ends of the environmental gradient occupied by A. hyperantus fluctuated least synchronously, suggesting a potential buffering effect of habitat heterogeneity, but this was not crucial to survival after the 1976 drought. Thus, not all habitats are equally important to persistence. Correlated temporal dynamics, variation in habitat quality and the interaction between habitat quality and temporal environmental stochasticity are important determinants of metapopulation persistence and should be incorporated in metapopulation models.

    Notes: Abstract. The treatment of infectious disease is compromised by the development of antibiotic-resistant strains of microbial pathogens. A variety of biochemical processes are involved that may keep antibiotics out of the cell, alter the target of the drug, or disable the antibiotic. Studies have shown that resistance determinants arise by either of two genetic mechanisms: mutation and acquisition. Antibiotic resistance genes can be disseminated among bacterial populations by several processes, but principally by conjugation. Thus the overall problem of antibiotic resistance is one of genetic ecology and a better understanding of the contributing parameters is necessary to devise rational approaches to reduce the development and spread of antibiotic resistance and so avoid a critical situation in therapy—a return to a pre-antibiotic era.

    Notes: Abstract Results of a statistical survey of F-region ion frictional heating are presented, a survey which is based on over 4000 h of common programme observations taken by the European incoherent scatter (EISCAT) UHF radar facility. The criterion adopted in this study for the identification of ion frictional heating was that defined by McCrea et al., requiring an enhancement in the F-region field-parallel ion temperature exceeding 100 K over two consecutive integration periods, which was itself based on a selection criterion for frictional heating derived for the study of high-latitude F-region ion temperature observations from the Atmospheric Explorer-C satellite. In the present study, the diurnal distribution of ion frictional heating observed by EISCAT is established and, furthermore, its dependence on geomagnetic activity and the orientation of the interplanetary magnetic field (IMF) is investigated; results are interpreted with reference to corresponding distributions of enhanced ion velocity, again derived from the extended set of EISCAT UHF common programme observations. The radar, due to its location relative to the large-scale convection pattern, observes ion frictional heating principally during the night, although preferentially during the post-midnight hours where there is reduced coupling between the ion and neutral populations. There is an increased preponderance of frictional heating during intervals of high geomagnetic activity and for a southward z component of the IMF and, moreover, evidence of asymmetries introduced by the y component of the IMF.

    Notes: Abstract In this study over 100 Pc5 events observed on the SAMNET magnetometer array have been catalogued. The ground horizontal polarization of these waves has been investigated and it has been shown that there are large differences between the polarization azimuth distributions on either side of noon. In the morning the azimuth distributions are spread evenly across all possible orientations whereas in the afternoon they are predominantly in a North-South orientation. The difference appears to be the result of a specific polarization pattern that occurred across the SAMNET array for 31 of the Pc5 events. These 31 Pc5 events all occurred in the morning sector, close to the dawn terminator. It is suggested that this special polarization pattern is a consequence of ionospheric conductivity gradients that occur at around dawn. This theory is reinforced by the comparison of the local times of these polarization variations with the local time of sunrise. By employing ionospheric conductivity data from the EISCAT radar for two of these Pc5 events, this connection has been studied in more detail.

    Notes: Abstract A statistical analysis of F-region and topside auroral ion upflow events is presented. The study is based on observations from EISCAT Common Programmes (CP) 1 and 2 made between 1984 and 1996, and Common Programme 7 observations taken between 1990 and 1995. The occurrence frequency of ion upflow events (IUEs) is examined over the altitude range 200 to 500 km, using field-aligned observations from CP-1 and CP-2. The study is extended in altitude with vertical measurements from CP-7. Ion upflow events were identified by consideration of both velocity and flux, with threshold values of 100 m s−1 and 1013 m−2 s−1, respectively. The frequency of occurrence of IUEs is seen to increase with increasing altitude. Further analysis of the field-aligned observations reveals that the number and nature of ion upflow events vary diurnally and with season and solar activity. In particular, the diurnal distribution of upflows is strongly dependent on solar cycle. Furthermore, events identified by the velocity selection criterion dominate at solar minimum, whilst events identified by the upward field-aligned flux criterion dominated at solar maximum. The study also provides a quantitative estimate of the proportion of upflows that are associated with enhanced plasma temperature. Between 50 and 60% of upflows are simultaneous with enhanced ion temperature, and approximately 80% of events are associated with either increased F-region ion or electron temperatures.

    Notes: Abstract The relationship between electric fields, height-integrated conductivities and electric currents in the high-latitude nightside electrojet region is known to be complex. The tristatic nature of the EISCAT UHF radar facility provides an excellent means of exploring this interrelationship as it enables simultaneous estimates to be made of the full electric field vector and the ionospheric Hall and Pedersen conductances, further allowing the determination of both field-perpendicular electric current components. Over 1300 h of common programme observations by the UHF radar system provide the basis of a statistical study of electric fields, conductances and currents in the high-latitude ionosphere, from which preliminary results are presented. Times at which there is significant solar contribution to the ionospheric conductances have been excluded by limiting the observations according to solar zenith angle. Initial results indicate that, in general, the times of peak conductance, identified from the entire set of EISCAT observations, do not correspond to the times of the largest electric field values; the relative contribution of ionospheric conductance and electric field to the electrojet currents therefore depends critically on local time, a conclusion which corroborates work by previous authors. Simultaneous measurements confirm a tendency for a decrease in both Hall and Pedersen conductances to be accompanied by an increase in the electric field, at least for moderate and large electric field value, a tendency which is also identified to some extent in the ratio of the conductances, which acts as an indicator of the energy of precipitating particles.
