    Notes: Abstract This paper investigates the proposal that the frequency of the swimming central pattern generator in young Xenopus tadpoles is partly determined by the population of glutamatergic premotor interneurons active on each cycle. During fictive swimming spinal neurons also receive cholinergic and electrotonic excitation from motoneurons. As frequency changes during swimming we make two predictions: first, since most motoneurons fire very reliably at all frequencies, the electrotonic and nicotinic drive from motoneurons should remain constant, and second, when swimming frequency decreases, the glutamatergic drive should decrease as the number of active premotor excitatory interneurons decreases. We have tested these predictions by measuring the excitatory synaptic drive to motoneurons as frequency changes during fictive swimming. The components of synaptic drive were revealed by the local microperfusion of strychnine together with different excitatory antagonists. After blocking the nicotinic acetylcholine receptor, the mainly glutmatergic excitatory synaptic drive still changed with frequency. However, when glutamate receptors or all chemical transmission was blocked, excitation did not change with frequency. Our predictions are confirmed, suggesting that premotor excitatory interneurons are a major factor in frequency control in the tadpole central pattern generator and that motoneurons provide a stable background excitation.

    Notes: Abstract Sulphur deficiency has become increasingly widespread in wheat in the U.K. Growth, nutrient content and biochemical responses to S and N supply of a breadmaking wheat variety (Hereward) and a non-breadmaking variety (Riband) were investigated in a pot experiment. Shoot dry matter (DM) at stem extension (Zadok's GS 37) and at maturity was increased markedly by S. Grain production of the Riband variety was more susceptible to the imbalance of N to S than the Hereward variety. At GS 37, the concentrations of total S and sulphate-S of shoots, chlorophyll meter readings and the concentrations of glutathione of the uppermost fully expanded leaves were increased significantly by increasing S supply, whereas the concentrations of nitrate and amides were decreased by S. The greatest relative changes in response to S supply were those of the glutathione and asparagine concentrations. Riband also showed greater response to S than Hereward. Critical values of various diagnostic indices at GS 37 were derived from the relationships between DM yield and different indices. The two varieties showed similar diagnostic curves except that for the ratio of total N to total S (N:S) in shoots. Either total S or sulphate-S can be used alone as a good indicator of deficiency, and with values of 1500 and 190 mg kg-1 DM in shoots for the two indices respectively. There was also a well defined relationship between DM yield and the glutathione concentration, with a critical value of 240 nmol g-1 FW. There were no advantages of using % of total S as sulphate-S. Shoot N:S ratio was found to be less accurate in predicting S deficiency than total S or sulphate-S. For prognostic purposes, a much higher S status at GS 37 was required to ensure no losses of DM yield due to S deficiency at maturity.

    Notes: Abstract A S-deficient soil was used in pot experiments to investigate the effects of S addition on growth and N2-fixation in pea (Pisum sativum L.). Addition of 100 mg S pot−1 increased seed yield by more than 2-fold. Numbers of pods formed were the most sensitive yield component affected by S deficiency. Sulphur addition also increased the concentration of N in leaves and stems, and the total content of N in the shoots. The amounts of N fixed by pea were determined at four growth stages from stem elongation to maturity, using the 15N dilution technique. Sulphur addition doubled the amount of N fixed at all growth stages. In contrast, leaf chlorophyll content and shoot dry weight were increased significantly by S addition only after the flowering and pod fill stage, respectively. Pea roots were found to have high concentrations of S, reaching approximately 10 mg g−1 dry weight and being 2.6–4.4 times the S concentration in the shoots under S-sufficient conditions. These results suggest that roots/nodules of pea have a high demand for S, and that N2-fixation is very sensitive to S deficiency. The effects of S deficiency on pea growth were likely to be caused by the shortage of N, due to decreased N2-fixation.

    Notes: Abstract. The centromeric region of swine chromosomes is comprised of tandemly repeated, divergent DNA monomer units. Here we report that these divergent DNA monomer sequences are organized into higher-order repeats, analogous to the hierarchical organization of α-satellite monomers in human centromeres. In this study, a centromeric cosmid clone was shown to be comprised entirely of a 3.3-kb higher-order repeat, with independent copies of this higher-order repeat more than 99% identical to each other. This higher-order repeat is composed of ten divergent monomer units of approximately 340 bp. The ten monomers are on average 79% identical, and all ten monomers are arranged in the same 5′ to 3′ orientation. In FISH analysis, a cloned 3.3-kb higher-order repeat hybridized to the centromere of Chromosome (Chr) 9 in metaphase spreads and detected two discrete foci in interphase nuclei, demonstrating that this swine higher-order repeat is chromosome-specific. The Chr 9 centromeric array spanned approximately 2.2 Mb as determined by pulsed-field gel electrophoresis. Moreover, the swine Chr 9 centromere is highly polymorphic, because an EcoRI restriction site polymorphism was detected. Thus, the assembly of divergent satellite sequences into chromosome-specific higher-order repeats appears to be a common organizational feature of both the human and swine centromere and suggests that the evolutionary mechanism(s) that create and maintain higher-order repeats is conserved between their genomes.

    Description: SUMMARYField experiments were conducted to test the seed yield responses of winter oilseed rape (Brassica napus L., cvs Libravo or Falcon) to the addition of different rates of S fertilizer, at three N application rates, on a sandy loam at Woburn, Bedfordshire, in 1990/91, 1991/92 and 1993/94. Large increases in seed yields, ranging from 0·7 to 1·6 t/ha, or 42–267% on a relative scale, were obtained in response to the application of 40 kg S/ha with 180 and 230 kg N/ha treatments. The effects of S were highly significant in 1991/92 (P 〈 0·01) and 1993/94 (P 〈 0·001) and close to significant (P = 0·053) in 1990/91. The yield benefits were obtained mainly from the application of the first 10 kg S/ha and further yield increases were unlikely above 40 kg S/ha. Increasing N application from 180 to 230 kg/ha decreased seed yield in 1990/91 and 1993/94, when no S was applied. In contrast, seed yield was not increased by S at zero or low (50 or 100 kg/ha) N rates. The interactions between N and S on seed yield were significant (P 〈 0·05) in 1990/91 but not in the other two seasons. Application of S also increased seed oil content in 1993/94, when the degree of S deficiency was particularly severe. With an application of 230 kg N/ha, the crops took up 5–22 kg S/ha at maturity when no S was applied and 26–51 kg S/ha when 40 kg S/ha was applied. The utilization efficiency of the fertilizer S ranged from 50 to 73% in the three seasons. Although the concentrations of total N in plants were largely unaffected by S treatments, large amounts of NO3-N accumulated in the leaves of S-deficient plants in 1993/94. This indicates that N metabolism was disrupted by S deficiency. The concentrations of S and the N: S ratios in different tissues and the whole plant changed considerably with time. The concentration of S in leaves at early flowering was found to be the best index in predicting S deficiency in terms of seed yield, and a critical value of 3·8 mg/g was obtained. In comparison, the N: S ratio in leaves at early flowering was a much poorer predictor of S deficiency.
