    Notes: The influence of the pH of the heating medium (which included several foods and buffers) on the thermal resistance (D and z-values) of spores of three Bacillus cereus strains was studied. Acidification from pH 7.0 to 4.0 produced a 5-fold decrease in D-values. Plots of log D vs pH gave straight lines, which made it possible to develop an equation to approximately predict the changes in heat sensitivity of B. cereus spores which occurred with changing pH. z-Values for two of the strains studied were not affected by acidification. On the other hand, with the strain ATCC 9818, a clear and statistically significant increase in z-value was observed as the pH decreased.

    Notes: The effects of the addition of starch, glucose, sodium chloride, sodium citrate, monopotassium phosphate and disodium phosphate to the recovery medium on apparent heat resistance of Bacillus cereus spores (ATCC 4342, 7004 and 9818) were investigated. Sodium citrate, monopotassium and disodium phosphate at concentrations of 0.1% were effective inhibitory agents for heat injured B. cereus spores especially for strain 9818, although only monopotassium and disodium phosphate caused a significant reduction (p 〈 0.05) in D-values obtained for strain 9818. Sodium chloride also had a marked effect on the recovery of heat injured spores. Concentration as low as 0.5% caused a significant reduction in the recovery rates for strains 9818 and 7004. In all cases, increasing the salt levels from 0.5 to 4% resulted in a progressive decrease in spore recovery. D-values gradually decreased as the salt content increased, although the concentrations which produced statistically significant differences (p 〈 0.05) varied among strains. The addition of starch at 0.1% resulted in a significant increase in the counts for strains 9818 and 7004. In contrast, glucose (0.1%), did not significantly modify the counts obtained Neither of these compounds affected decimal reduction times. No statistical significance (p〉0.05) differences were detected among z-values for the spores of the three strains recovered in the presence of different additives assayed. z-Values ranged from 6.67 to 8.32, with a mean value of 7.56 ± 0.46C.

    Notes: . The in vitro cytopathic effect of Trichomonas vaginalis on epithelial cells was explored through the interaction of trophozoites of the virulent strain GT-10 with MDCK monolayers. The interaction was analyzed through electrophysiology, video microscopy, and transmission and scanning electron microscopy. Electrical measurements revealed that living parasites produced severe damage to the cell monolayers within 30 min, manifested as a rapid decrease in transepithelial resistance. Microscopic observations demonstrated that when placed in contact with epithelial cells, trichomonas formed clumps through interdigitations and transient plasma membrane junctions between adjacent parasites. Also, attached trophozoites adopted an ameboid shape. The in vitro cytopathic action of T. vaginalis on MDCK cells was initially evident by modifications of the plasma membrane, resulting in opening of tight junctions, membrane blebbing, and monolayer disruption. After 15 min of interaction the damage was focal, concentrating at sites where parasite clumps adhered to the monolayer. At 30 min practically all MDCK cells were dead, whether or not trichomonas were attached to them. These events were followed by detachment of lysed cells and complete disruption of the monolayer at 60 min. Electron microscopy demonstrated a peculiar form of adhesion that appears to be specific for trichomonas, in which the basal surface of T. vaginalis formed slender channels through which microvilli and cytoplasmic fragments of epithelial cells were internalized. The same sequence of lytic events was found with the less virulent GT-3 strain. However, the time course of cytolysis with GT-3 parasites was much slower, and lysis was limited to areas of attachment of T. vaginalis.

    Notes: The effect of cow slurry N on productivity and white clover content was compared with that of mineral fertilization in a natural grassland sward. Fertilizer was applied at two different N rates as split applications of ammonium nitrate, or as cow slurry (145 or 290 kg N ha−1 in the first year and 120 or 240 kg N ha−1 In the second year)- In the first year, fertilizer N produced higher dry matter (DM) yields than the slurry treatments. Nitrogen efficiency and clover content reduction were higher for the mineral treatments, the efficiency being lower for the higher rate of N application. In the second year, N supplied by the slurry gave the same productivity, N efficiency and white clover content reduction as that from the mineral N fertilization; higher efficiency in the second year being due to favourable weather conditions and low white clover contents. The high N efficiencies of the slurry compared with the fertilizer N treatments were attributed to the dilution of the slurry and to split applications over the year. Yield differences between treatments were due mainly to the grass component of the sward. Competition between grasses and other species, and white clover for mineral N resulted in a reduction in the white clover content in those treatments in which more N was supplied, whether it came from the mineralization of slurry or from the fertilizer N applied.

    Notes: Wine vinegar is a product obtained from wine acidification which contains at least 5% by wt. of acetic acid, in general without any additives or colorings.Aspects studied in this work include: the determination of the taste group thresholds (geometric mean of the individual best-estimate thresholds “BET”) of two different acids (citric and acetic acids) in aqueous solution and spanish vinegars produced from table and sherry wines. The results obtained suggest that wine vinegar can be considered something more than just an acidulant agent.In order to evaluate differences among wine vinegars, discriminant tests for twenty-five spanish vinegars (sherry, table and flavored vinegars) were applied. Six of the twelve attributes freely chosen by assessors allowed grouping of the spanish wine vinegars according to their sensory aspects.

    Notes: A continuous extrusion process to provide instant corn flour for tortillas was evaluated. Variables investigated included two types of mill (knives and hammer) with screens with two diameters (0.5 and 0.8 mm), two types of corn (hard endosperm or normal and soft endosperm or cacahuazintle), lime concentration (0.15 and 0.25% w/w), processing moisture (45 and 48% w/w) and temperature (70,80 and 90°C). The water absorption capacity, water solubility index, color of instant corn flour, adhesiveness of masa, tensile strength, cutting force, rollability and puffing of tortillas, were compared. Based on textural data the hammer mill with 0.8 mm sieve, normal corn type, 0.15% lime, 48% moisture and 90°C processing temperature, produced the highest quality tortillas.

    Notes: Samples of skin tissue were collected by biopsy darting from humpback whales (Megaptera novaeangliae) in six seasonal habitats representing three stocks and four regions: Groups IV (western Australia), V western component (eastern Australia), V eastern component (New Zealand and Tonga) and VI (the Antarctic Peninsula and Gorgona Island, Colombia, South America) of the Southern Hemisphere. A variable section of the mitochondrial DNA control region was amplified and sequenced from 84 of these individuals, distinguishing a total of 48 unique sequences (i. e., mtDNA nucleotypes). Phylogenetic reconstructions suggested that these nucleotypes form three clades, corresponding to those previously described in a world-wide survey of humpback whale mtDNA variation, although bootstrap support for two of the clades was relatively low (〈50%). An analysis of variance adapted for molecular information showed significant differentiation of nucleotypes among the three Groups (Stocks) and heterogeneity of haplotype diversity among the four regions. A pattern of interchange within and between oceanic basins was demonstrated by the presence of shared identical nucleotypes among humpback whales in regions of the Southern and Northern Hemispheres.

    Notes: Development of multiple insect resistance in tropical maize represents a major effort of the maize breeding programme at CIMMYT. Resistance to the southwestern corn borer (SWCB) is polygenically controlled with primarily additive gene action. Our main objective was to identify quantitative trait loci (QTL) involved in resistance to SWCB. Other objectives were to detect QTL in the same population for plant height, female flowering, and the anthesis-silking interval (ASI). A population of 472 F2 individuals derived from a cross between the susceptible line Ki3 and the resistant inbred CML139, was restriction fragment length polymorphism (RFLP) genotyped using 110 maize probes. F3 families were rated for leaf-feeding damage after artificial infestation at one location in three consecutive years. Height and flowering were measured in protected trials in two locations. QTL analyses were conducted using joint composite interval mapping. Seven QTL on chromosomes 3, 5, 6, 8, and 9 explained 30% of the phenotypic variance (σ2p) for SWCB damage. Most QTL alleles conferring resistance were contributed from CML139. QTL showed dominance, partial dominance and additive gene action. Eleven QTL dispersed across the genome were determined to affect plant height and explained 43% of σ2p. Four of these were in close proximity to loci with qualitative effects on plant height. Thirteen QTL (50% of sigma;2p) were identified for days to female flowering and nine (30% of σ2p) for ASI. Our results, along with those from other mapping studies at CIMMYT, are allowing us to formulate marker-assisted selection schemes to complement the breeding efforts for such complex traits as borer resistance.
