    Description: The activity of hemolytically inactive C5b67, designated iC5b67, was evaluated as an agonist for functional responses of human polymorphonuclear leukocytes (PMN). C5b67 was formed from purified human complement components and decayed in phosphate-buffered saline (PBS) until it had no lytic activity for sheep erythrocytes in a standard assay. iC5b67, at nanomolar concentrations, stimulated PMN chemotaxis and Ca2+ fluxes, but inhibited superoxide production and failed to upregulate CR1 and CR3. There was no significant contamination of the iC5b67 with C5a to explain these results. Neither isolated C5b6 nor C7 alone exhibited the activities of iC5b67, while insolubilized anti-C7 could remove the PMN agonist activity from the iC5b67 preparation. Binding studies to define a specific receptor for iC5b67 on PMN were hampered by the very hydrophobic nature of the ligand. 125I-iC5b67, by contrast to hemolytically active 125I-C5b67, was unable to insert in erythrocytes, suggesting that iC5b67 need not insert in the PMN membrane to induce signaling. Two lines of evidence suggest that iC5b67 and C5a and FMLP share common steps in intracellular signaling (1) pretreatment of PMN with iC5b67 deactivates PMN for C5a- and FMLP-induced chemotaxis; and (2) pretreatment of PMN with pertussis toxin inhibits iC5b67-induced chemotaxis. Thus, iC5b67 has important effects on the activity of PMN and G-proteins and Ca2+ are involved in the signaling.

    Notes: Thermally activated magnetization reversal processes become manifest in the dependence of the remanent coercivity on the time during which a magnetic field is applied opposite to the initial magnetization direction. They have important consequences for the long term stability and short time writeability of future high density recording media. In this paper, we report on a new experiment using a contact write/read tester to study the time dependence of the remanent coercivity over more than 10 orders of magnitude (from 6 ns to 〉60 s). Remanence coercivity and signal decay measurements of a CoPtCr recording medium with 5.5 nm thickness are presented. © 1999 American Institute of Physics.

    Notes: A frequently used method for determination of impurity transport coefficients is the analysis of the radial and temporal time evolution of the impurity density of injected tracer impurities. In cases when the impurity density cannot be measured directly, e.g., by active charge exchange recombination spectroscopy, the impurity density has to be reconstructed from impurity radiation as measured, e.g., by the soft-x camera. Due to the energy integrated information, assumption about the spatial ionization state distribution, e.g., coronal equilibrium, is necessary. This might not be valid in cases of large transport. Consequently, the transport quantities, derived under the preliminary assumption of radial coronal distribution, are used again in an iterative way to improve the density reconstruction in order to approach more realistic transport quantities. The procedure was applied to plasma discharges in the stellarator Wendelstein 7-AS. © 1999 American Institute of Physics.

    Notes: The Wendelstein 7-X experiment is a concept test for properties of reactor relevant plasmas in advanced stellarators. Prominent features include a modular superconducting coil assembly, a fivefold toroidal symmetry, and a helical magnetic axis. Due to the optimization process, W7-X is characterized by a vacuum magnetic field configuration with smooth magnetic surfaces, improved equilibrium properties with a weak dependence of rotational transform and shear on the plasma pressure β, good magneto-hydrodynamic stability properties due to magnetic well stabilization, reduced neoclassical transport losses and negligible bootstrap current in the long mean-free-path regime, good collisionless α-particle confinement in an equivalent reactor, and, as a technical aspect, good feasibility of the superconducting modular coils. W7-X will be heated by continuous electron cyclotron resonance heating and pulsed neutral beam injection and ion cyclotron resonance heating. The envisaged parameters are Te≤10 keV, Ti≤6 keV central densities ≤3×1020 m−3 with an averaged 〈β〉≤5%. Despite the complicated geometrical structure, all basic diagnostics are compatible with W7-X. Generally, diagnostic methods and applications in a stellarator are not different from those in tokamaks. However, special efforts are being made to equip the experiment with those diagnostics necessary to measure the quantities directly related with the optimization of the machine: the verification of the predicted magnetic topology and characterization of the configuration throughout the entire parameter range, the identification of equilibrium and stability, and the determination of the confinement properties. The article describes the strategy developed which assures that the detailed measurement needs of the W7-X experimental program can be met. © 1997 American Institute of Physics.

    Notes: The spatially and temporally resolved x-ray emission in the W7-AS stellarator is analyzed by three diagnostic systems. The first consists of two cameras using arrays of surface barrier diodes. A variety of absorber-foils can be used to change the spectral response of the system. This flexibility allows one to analyze impurities and magnetohydrodynamic (MHD) activity. The measurements are also used for the reconstruction of equilibrium parameters (e.g., Shafranov shift) and of the topology of MHD modes. The second system serves to monitor the electron temperature profile by using the two-foil method. Pairs of large ion-implanted Si diodes in combination with different foil combinations are used. In addition, the measurements provide routine informations about Zeff, plasma position and profile width. Finally, a compact camera system (ten cameras with planar pin-diode arrays and curved Be-foils, 320 viewing chords) inside the vacuum vessel allows one to obtain proper two-dimensional tomograms of the equilibrium flux surfaces and of mode structures without prior information. In addition to the camera systems spectral information is obtained from x-ray pulse height measurements with two Si(Li) detectors. These data are correlated with the x-ray profile measurements by an impurity radiation code. The contribution of x-ray measurements to the investigation of various physics issues is illustrated by selected examples. © 1999 American Institute of Physics.

    Notes: A laser vibrometry based dynamic technique for biaxial residual stress determination in micromembranes is reported. Layered, low pressure chemical vapor deposition grown, rectangular shaped micromembranes are ultrasonically excited. The resulting micromembrane displacement, as a function of frequency and position, is recorded to yield resonance frequencies and associated vibrational mode numbers. The stress is determined from this information. Vibrations forced from ultrasonic pressure (∼25 Pa) resulted in peak displacements of tens of nanometers at resonance. For the sample set studied, the (3,1) mode resonance peak displayed the least damping in atmospheric testing and was used to establish stress levels ∼108 N/m2. A trend for this stress showing an overall decrease by ∼40% as a result of thermomechanical cycling was identified. © 1996 American Institute of Physics.

    Notes: Two groups of waterbirds have been chosen to assist in measuring the success of restoration of the traditional channel configuration, hydrologic regimes, and floodplain wetlands of the Kissimmee River: waterfowl (Anseriformes) and waders (Ciconiiformes). Waterfowl are dominant, swimming omnivores that use seeds, foliage, and invertebrates; waders are mainly walking predators that eat fish of various sizes. Both can be censused by well-established air and ground techniques, and both can be used to compare post-restoration with channelized or pre-channelization population data (waterfowl) or bird use of channelized versus restored wetlands (waders). In addition to use of population data, species richness and regularity of occurrence should provide a basis for assessing restoration of biological integrity. Conceptual models of avian habitat use for nesting and feeding demonstrate patterns of segregation that will aid assessments for some species. Other species show high overlap in foods and habitats and will require additional measures of response. To understand these patterns and reasons underlying waterbird use, measurements of habitat type, vegetation structure, and food resources will be essential. Integration of these high trophic-level guilds with evaluations of other system components will ensure an ecosystem perspective. Predicted responses to restoration suggest an increase in species richness and number of individuals of many species.
