    Notes: Particulate ceramic composites that were composed of a combustion-synthesized β′;-SiAlON matrix and dispersed MoSi2 particles were hot pressed at 1600°C in a nitrogen atmosphere. The physical and mechanical properties of the composites that contained 15, 30, and 45 vol% MoSi2 were evaluated. The average four-point bend strength, fracture toughness, and Vickers hardness of the composites were in the ranges of 500-600 MPa, 3-4 MP·am1/2, and 11-13 GPa, respectively. The measured mechanical strength and hardness were very similar to the values that were predicted from the rule of mixtures. The fracture toughness of the combustion-synthesized β′;-SiAlON (2.5 MPa·m1/2) was apparently enhanced by the MoSi2 particles that were added. The increase in the fracture toughness was predominately attributed to the residual thermal stress that was induced by the thermal expansion mismatch between the MoSi2 particles and the β′;-SiAlON matrix. The composites showed improved electrical conductivity and oxidation resistance over monolithic β′;-SiAlON. High-resolution transmission electron microscopy examination of the composites indicated that the MoSi2 was chemically well compatible with the β′;-SiAlON.

    Notes: Abstract Congenital adrenal hyperplasia (CAH) is a common autosomal recessive disorder mainly caused by defects in the steroid 21-hydroxylase (CYP21) gene. We have analyzed CYP21 gene sequences in 65 CAH families in Taiwan. All ten exons of the CYP21 gene were analyzed by differential polymerase chain reaction followed by single-strand conformation polymorphism electrophoresis and the amplification-created restriction site method. About 95% (123 chromosomes) contain mutations due to conversion of DNA sequences into its neighboring homologous pseudogene, CYP21P. Four novel mutations representing 5% of the total chromosomes have also been identified. The mutations were confirmed by sequencing an aberrant DNA fragment. These four mutations included a base change of the splicing donor site at intron 2 from GT to AT, a base substitution of C to T at codon 316, deletion of ten bases (TCCAGCTCCC) at codons 330–333 of exon 8, and duplication of 16 bases (CCTGGATGACACGGTC) at codons 393–397 of exon 9. The loss of the splicing donor site at intron 2 and the premature stop at codon 316 may result in aberrant splicing to reduce enzyme activity and a truncated protein with no enzyme activity, respectively. Likewise, both the duplication and the deletion forms create a frameshift and premature stop during translation. The resulting proteins lack the heme-binding domain and hence are expected to lose enzymatic activity. Since these mutations are not found in the neighboring CYP21P pseudogene, gene conversion should not be the cause of these novel mutations.

    Notes: Abstract  The tissue kallikrein-kinin system contributes to the regulation of local blood flow, vascular permeability, inflammatory responses, and ion transport. Tissue kallikrein is a serine proteinase which produces vasoactive kinin peptides. Kallistatin specifically binds to tissue kallikrein and inhibits its proteolytic activity. To investigate their anatomical relationship in the human pancreas and salivary glands, the expression and localization of kallistatin and tissue kallikrein were identified by immunoassays, immunohistochemistry, and in situ hybridization histochemistry. Human kallistatin and tissue kallikrein levels were measured by ELISA and radioimmunoassay, respectively, in pancreatic and salivary tissue extracts, and in pancreatic fluid and saliva. Immunoreactive kallistatin and kallikrein were colocalized in acinar cells of the human pancreas by immunohistochemistry. In situ hybridization histochemistry confirmed the presence of both mRNAs in pancreatic acini. In salivary glands, kallistatin and kallikrein mRNAs were also colocalized in serous acinar cells, and the kallikrein transcript was further localized to striated and interlobular ducts. Immunoreactive kallistatin was localized in serous acinar and demilune cells of salivary glands and kallikrein was localized to the epithelium of striated and interlobular ducts. The colocalization and/or coexpression of human tissue kallikrein and kallistatin in the pancreas and salivary glands suggest a role for kallistatin in the regulation of tissue kallikrein in these organs.

    Notes: Abstract Human tissue kallikrein gene polymorphisms were identified in the promoter region by polymerase chain reaction (PCR) and DNA sequencing. One polymorphic region was identified between nucleotides –121 and –133 with respect to the transcription initiation site of the tissue kallikrein gene. Ten alleles with length and nucleotide sequence variations were detected among 108 unrelated Caucasians, African-Americans, and Asians. The polymorphisms show Hardy-Weinberg equilibrium. Allele-specific amplification and PCR analyses were used to detect the various forms of polymorphism. The promoter activity was analyzed in human embryonic kidney 293 cells by transient transfection assays. Sequential 5′-deletion analysis of the tissue kallikrein gene promoter revealed that the region from –144 to –98 is crucial for its promotor activity, while alleles D and H had significantly lower promoter activities than the other alleles in the –940/+10 deletion constructs. The high variability and the proximity to the tissue kallikrein gene render it suitable for application as a new tool in genetic studies for evaluation of the tissue kallikrein gene in the pathogenesis of human essential hypertension.

    Notes: Abstract Samples of alluvial soil from mixed sandstone shale and slate and of Taiwan clay were collected from two sites, both managed under a similar crop rotation scheme. The fields were further divided into sections which were managed under either conventional farming or sustainable farming practices. When the soil samples were collected in April 1989, after 1 year of operation under conventional or sustainable practices, the nitrification activities of both soils managed under sustainable practices practices. The nitrifying activities in Taiwan clay samples collected in April 1993 which had been managed with chemical or with organic fertilizer were not significantly different. However, nitrifying activity in the alluvial soil was higher under sustainable than under conventional practices. Numbers of NH 4 + -oxidizing bacteria were not significantly different in any of the soil samples irrespective of the different management practices. In contrast, higher numbers of NO 2 - -oxidizing bacteria were detected in both soils managed sustainably. The results also indicated that the composition of NH 4 + -oxidizing bacteria differed in the alluvial soil when managed with different kinds of fertilizer.
