    Notes: The configurational and conformational properties of six 4-trimethylammoniummethyl 2,2-disubstituted 1,3-dioxolanes were studied using NOE experiments and vicinal coupling constants. Selective 1D NOE experiments proved to be effective tools in the configurational assignment of C-2 relative to C-4. The preferred conformation of the dioxolane ring and the exocyclic group at C-4 was obtained by employing vicinal coupling constants and NOE results. The 1H and 13C NMR chemical shifts show stereochemically dependent trends. Quantitative analysis of conformer populations was performed using Haasnoot et al's equation. The —N+(CH3)3 group was found to be synclinal with respect to the heterocyclic O-3 atom and points outside the ring. When a phenyl group is present at C-2, the 4-CH2N+ -  group in a trans relationship to the 2-phenyl ring was found to occur prevalently in a pseudo-axial orientation, whereas it was established to be prevalently pseudo-equatorial when cis with respect to the phenyl ring.

    Notes: Two pairs of enantiomerically pure cis-fused cyclopenteno-1,2,4-trioxanes (7, ent-7 and 8, ent-8) are prepared (Schemes 1-3). Their identities are established by dye-sensitized photo-oxygenation of ent-7 and 8, ent-8 to the allylichydroperxides, reduction to the corresponding alcohols, and conversion to the (1S)-camphanates (Scheme 4), the structures of which are determined by X-ray analysis. The dynamic properties of ent-7 are investigated by NMR spectroscopy and PM3 calculations. Evidence for an easily accessible twist-boat conformation is obtained. The in vitro and in vivo antimalarial activities of 7, ent-7,8, and ent-8 as well as those of the racemic mixtures are evaluated against Plasmodium falciparum, P. berghei, and P. yoelii. No correlation is observed between configuration and activity. Racemates and pure enantiomers have commensurate activities. The mode of action on the intraerythrocytic parasite is rationalized in terms of close docking by the twist-boat conformer of the trioxane on the surface of a molecule of heme, single-electron transfer to the O—O σ* orbital, and scission to the acetal radical which then irreversibly isomerizes to a C-centered radical, the ultimate lethal agent (Scheme 5).

    Notes: The reaction of palladium acetate with two equivalents of di- and triethanolamines RN(CH2CH2OH)2 in the presence of a base affords the new chelate-stabilized alkoxo Pd(II) complexes [Pd(OCH2CH2N(R)CH2CH2OH)2] [R = Me (1), Et (2), n-Bu (3), benzyl (4) or CH2CH2OH (5)]. These N,O-ligated complexes are isolated in high yield as yellow, crystalline solids and are thermally stable despite the presence of several β-hydrogen atoms in the ligand system. Both complexes possess a square-planar palladium coordination geometry with the two oxygen atoms positioned mutually trans. The most notable difference in the molecular structures is that 1 forms a two dimensional network of intermolecular O-H≡O hydrogen bonds, whereas 5 forms intramolecular O-H⃛O hydrogen bonds, which cage the palladium center. In solution 1-4 exist as a diastereoisomeric mixture (a racemic enantiomeric pair SNSN, RNRN and a mesomeric form RNSN) in a 1:1 molar ratio, and this ratio is independent of temperature in nonalcoholic solvents, When complexes 1-4 are dissolved in protic solvents (e.g. MeOH) the diastereomeric excess is temperature-dependent due to an exchange process between the meso diastereoisomer and the (racemic) enantiomeric pair. Thermodynamic parameters for this process in a mixture of MeOH-toluene have been determined with NMR and show this process to be influenced by the steric nature of the alkyl substituent (R) on nitrogen. A conformational analysis based on 1H-NMR coupling constants within the N,O-chelate ring of complexes 1-4 provides details on the solution structure of the ring in both diastereoisomers.

    Notes: A high-performance liquid chromatographic method has been developed for the analysis of the intermediate imines and end products in an asymmetric isomerization route toward optically active amines. Separation of the imine enantiomers was performed on commercially available Chiralcel OD-H, Chiralcel OJ, and Chiralpak AD chiral stationary phases. All substituted imine enantiomers could be readily resolved with selectivities (α) higher than 1.10 using the Chiralpak AD column. By derivatization with ring-substituted benzaldehydes, aromatic amines were converted into Schiff base derivatives and the enantiopurity of these amines was determined.     Notes: A high-performance liquid chromatography and molecular modelling study is presented for the diastereomeric adducts of a series of chiral thiol compounds and chiral amine compounds after reaction with o-phthalaldehyde (OPA). It is shown that the possibility of hydrogen bonds forming between the thiol and amino residues in the diastereomeric adducts is an important factor for obtaining good selectivity. The experimental elution orders were explained on the basis of the strength of the interaction between the polar groups of the thiol and amine constituents; it was found that the diastereomeric form having the largest distance between these groups always eluted first. Moreover, within a series of OPA derivatives, the differences between the Boltzmann weighted averages of the distances between the polar groups of the two diastereomers show a good correlation with the experimental selectivities. © 1995 Wiley-Liss, Inc.

    Notes: Matrix-assisted laser desorption/ionization (MALDI) was used for several small proteins (such as insulin) and for peptides. It was found that the detection efficiencies of MALDI for the insulin B chain and the insulin A chain are drastically different. Similar phenomena were also observed for various types of peptides. The positive-ion signal of MALDI in detecting proteins or peptides was found to be greatly enhanced by the presence of a basic amino acid in their chains. The experimental results indicate that this enhancement may arise from proton transfer in solution by an acid-base reaction between the protein/peptide and matrix molecule. This pre-protonated mechanism provides a low energy barrier for the ionization of peptides in a MALDI process, and greatly reduces the energy threshold of MALDI. Matrix effects on the ionization mechanism are discussed.

    Notes: High nitrile content cyano-substituted polymethylsiloxane encapsulated particles were prepared for double bond selectivity in packed capillary column supercritical fluid chromatography (SFC). Silica particles were deactivated with 50% cyanopropyl-substituted polymethylhydrosiloxane, and coated with 25% o,p-dicyano-p-biphenyl-substituted polymethylsiloxane (o,p,p-DCBP), and 25% m,m-dicyano-p-phenyl-substituted polymethylsiloxane (m,m,p-DCBP) stationary phases. Argentation of the particles was carried out by complex formation between silver ions and the cyano groups on the particles. Unsaturated fatty acid methylesters (FAMEs) were used to investigate the selectivity of these packing materials to double bonds using packed capillary column SFC. Separations were performed according to the degree, position, and geometrical configuration of the double bonds. The effect of temperature on selectivity was also investigated. The stability of the packing materials was evaluated by monitoring the loss in selectivity of the oleate/elaidate isomers after washing the column with supercritical CO2 at 300 atm and 85°C. The separation of FAMEs in a commercial fish oil was compared using capillary columns packed with DCBP coated and silver-complexed particles.
