    Notizen: Abstract This paper presents the reflections of the Working Group of which the tasks were to examine the non-rigid Earth nutation theory. To this aim, six different levels have been identified: Level 1 concerns the input model (giving profiles of the Earth's density and theological properties) for the calculation of the Earth's transfer function of Level 2; Level 2 concerns the integration inside the Earth in order to obtain the Earth's transfer function for the nutations at different frequencies; Level 3 concerns the rigid Earth nutations; Level 4 examines the convolution (products in the frequency domain) between the Earth's nutation transfer function obtained in Level 2, and the rigid Earth nutation (obtained in Level 3). This is for an Earth without ocean and atmosphere; Level 5 concerns the effects of the atmosphere and the oceans on the precession, obliquity rate, and nutations; Level 6 concerns the comparison with the VLBI observations, of the theoretical results obtained in Level 4, corrected for the effects obtained in Level 5. Each level is discussed at the state of the art of the developments.

    Notizen: Abstract. Environmental concentrations and biological effects of certain metals and organic compounds found in wood preservatives were examined. The study focused on leachates from private residential docks in South Carolina tidal creeks. Copper, chromium, arsenic, and polynuclear aromatic hydrocarbons (PAHs) were measured in composite samples of surficial sediments and naturally occurring oyster populations (Crassostrea virginica) from creeks with high densities of docks, and from nearby reference creeks with no docks. In some cases, metal concentrations in sediments and oysters were higher immediately adjacent to dock pilings than they were elsewhere in the same creek. Sediments from most sites had concentrations of metals and total PAHs which were below levels reported to cause biological effects, however. Solid-phase Microtox® bioassays using whole sediments and rotifer bioassays using sediment pore water showed no significant differences in acute toxicity between creeks with and without docks. Oysters growing directly on dock pilings had significantly higher concentrations of copper than oysters growing at least 10 m away; however, there was no significant difference in the physiological condition of these oysters. Four-day field bioassays measuring percent survival of mummichogs (Fundulus heteroclitus), mud snails (Ilyanassa obsoleta), juvenile red drum (Sciaenops ocellatus), and juvenile white shrimp (Penaeus setiferus) showed no significant differences between sites near to and distant from newly constructed docks. Hatchery-reared oysters showed no significant differences between dock and reference sites in percent survival, growth, or bioaccumulation of metals after six weeks of exposure. The results suggest that, in estuarine environments with a moderate tidal range (1.5–2.0 m), wood preservative leachates from dock pilings have no acutely toxic effects on four common estuarine species, nor do they affect the short-term survival or growth of juvenile oysters.

    Notizen: Abstract We have developed an efficient transformation system for red raspberry (Rubus ideaus L.) using Agrobacterium mediated gene transfer. Using this system we have successfully introduced a gene that encodes an enzyme, S-adenosylmethionine hydrolase (SAMase), in raspberry cultivars Meeker (MK), Chilliwack (CH) and Canby (CY). Leaf and petiole expiants were inoculated with disarmed Agrobacterium tumefaciens strain EHA 105 carrying either of two binary vectors, pAG1452 or pAG1552, encoding gene sequences for SAMase under the control of the wound and fruit specific tomato E4 promoter. Primary shoot regenerants on selection medium were chimeral containing both transformed and non-transformed cells. Non-chimeral transgenic clones were developed by iterative culture of petiole, node and leaf explants, on selection medium, from successive generations of shoots derived from the primary regenerants. Percent recovery of transformants was higher with the selection marker gene hygromycin phosphotransferase (hpt), than with neomycin phosphotransferase (nptII). Transformation frequencies of 49.6%, 0.9% and 8.1% were obtained in cultivars Meeker, Chilliwack and Canby respectively from petiole expiants using hygromycin selection. Genomic integration of transgenes was confirmed by Southern hybridization. Transgenic plants from a total of 218 independent transformation events (161 MK, 4 CH, 53 CY) have been successfully established in soil.

    Notizen: Abstract Previous research has demonstrated a west-to-east decrease in zooplankton δ13C along the Alaskan coast. We extended the transect into the Canadian Arctic archipelago through the Northwest Passage in order to investigate whether this longitudinal gradient persists eastward. Contrary to the previous observations, zooplankton δ13C was found to be essentially invariate, despite a sampling survey length double that of the earlier studies. This resulted from zooplankton at the coldest and ice-bound stations being found to be enriched in 13C relative to what would be expected from the ambient water temperatures and calculated [CO2aq]. We suggest that this was due to the assimilation of ice POM which, as has been demonstrated for other forms of benthic algae, is enriched in 13C due to restraints on boundary layer diffusion. The present results therefore support other traditional, non-isotope research in suggesting that ice-bound polar foodwebs may be examples of aquatic systems where there is a functional energy connectivity between the attached and openwater environments.

    Notizen: Abstract A full-length cDNA encoding the 206 amino acid open reading frame of a trypsin/chymotrypsin inhibitor abundant in the corms of giant taro (Alocasia macrorrhiza) was isolated. An internal fragment was cloned using degenerate primers corresponding to a region of the mature protein sequence and the ‘rapid amplification of cDNA ends’ (RACE) method used to generate a composite cDNA sequence. The length of the cDNA was close to the predicted size of the corresponding transcript deduced from northern blot analysis of corm mRNA. The inhibitor was expressed strongly in the mature corm, at low levels in leaf blades and petioles but not in roots. Southern blot analysis of the giant taro DNA indicated that this inhibitor is encoded by a small multigene family and this was further supported by the isolation of two different sequence classes from corm cDNA using primers to the ends of the composite sequence.

The gating cycle of CFTR (Cystic Fibrosis Transmembrane conductance Regulator) chloride channels requires ATP hydrolysis and can be interrupted by exposure to the nonhydrolyzable nucleotide AMP-PNP. To further characterize nucleotide interactions and channel gating, we have studied the effects of AMP-PNP, protein kinase C (PKC) phosphorylation, and temperature on gating kinetics. The rate of channel locking increased from 1.05 × 10−3 sec−1 to 58.7 × 10−3 sec−1 when AMP-PNP concentration was raised from 0.5 to 5 mm in the presence of 1 mm MgATP and 180 nm protein kinase A catalytic subunit (PKA). Although rapid locking precluded estimation of P o or opening rate immediately after the addition of AMP-PNP to wild-type channels, analysis of locking rates in the presence of high AMP-PNP concentrations revealed two components. The appearance of a distinct, slow component at high [AMP-PNP] is evidence for AMP-PNP interactions at a second site, where competition with ATP would reduce P o and thereby delay locking. All channels exhibited locking when they were strongly phosphorylated by PKA, but not when exposed to PKC alone. AMP-PNP increased P o at temperatures above 30°C but did not cause locking, evidence that the stabilizing interactions between domains, which have been proposed to maintain CFTR in the open burst state, are relatively weak. The temperature dependence of normal CFTR gating by ATP was strongly asymmetric, with the opening rate being much more temperature sensitive (Q 10= 9.6) than the closing rate (Q 10= 3.6). These results are consistent with a cyclic model for gating of phosphorylated CFTR.

    Notizen: Abstract Transgenic plants of strawberry cultivar Totem were developed by Agrobacterium-mediated transformation using a plasmid vector containing gus and nptII genes. Parallel experiments were carried out with and without repeated subculturing (iterative cultures) for generation of transgenic shoots on selection medium. The selection levels in the non-iterative pathway were kept constant, while in the iterative protocol, stepwise increase of selection pressure was applied at different stages of tissue growth. Rooted transgenic plants obtained via both protocols were outplanted in soil. Random leaf samples of greenhouse-grown transgenics were analysed for the presence of gus gene sequences by Southern hybridization as well as gus expression on leaf and petiole tissues by X-Gluc histological assay. Random leaf samples analysed from individual transgenic events developed under iterative culture were positive for the gus insert as verified by Southern analysis confirming the presence of transgenes and lack of chimaeras. Leaf samples of the transgenic events from the non-iterative protocol were either positive or negative on Southern analysis indicating the chimaeric nature of the transgenic plants. The absence of gus sequences in the transgenic plants grown under the non-iterative protocol reinforced the necessity of iterative cultures along with stepwise increase in selection levels for generating non-chimaeric transgenics in strawberry. The gus expression was highly variable, irrespective of the iterative or non-iterative protocol used for transformation. We conclude that strawberry is highly prone to develop chimaeric transgenics if derived from primary regenerants and that the iterative culture technique effectively converts chimaeras to pure line transgenic plants
