    Notizen: A Biofilm Airlift Suspension (BAS) reactor was operated with nitrifying biofilm growth and heterotrophic suspended growth, simultaneously converting ammonium and acetate. Growth of heterotrophs in suspension decreases the diffusion limitation for the nitrifiers, and enlarges the nitrifying capacity of a biofilm reactor. Neither nitrifiers nor heterotrophs suffer from additional oxygen diffusion limitation when the heterotrophs grow in suspension. Control of the location of heterotrophic growth, either in suspension or in biofilms over the nitrifying biofilms, was possible by manipulation of the hydraulic retention time. A time delay for formation and disappearance of the heterotrophic biofilms of 10 to 15 days was observed. Surprisingly, it was found that in the presence of the heterotrophic layers the maximum specific activity on ammonia of the nitrifying biofilms increased. The reason for the increase in activity is unknown. The effect of heterotrophic biofilm formation on oxygen diffusion limitation for the nitrifiers is discussed. Some phenomena compensating the increased mass transfer resistance due to the growth of a heterotrophic layer are also presented. © 1997 John Wiley & Sons, Inc. Biotechnol Bioeng 53: 397-405, 1997.

    Notizen: A method for the control of dissolved oxygen tension in growing microbial cultures is described. The apparatus consists of a motor-driven air sparge pipe which may be lowered or raised to give a variable point of entry of the air stream into the culture liquid and hence a variable gas dispersion and gas-liquid contact time. Control of the sparge pipe position is by means of a feedback control loop consisting of a dissolved oxygen probe, an on/off controller, and a reversing electric motor which drives the sparge pipe. The difficulty presented by the relatively slow response of the oxygen probe has been overcome by incorporating an adjustable rate of control action.

    Notizen: The intracellular levels of seven enzymes in mouse LS cells growing in suspension culture at controlled dissolved oxygen partial pressures (pO2) have been measured. During the growth of each culture large fluctuations were observed in the levels of some enzymes, particularly aldolase and cytochrome oxidase. Mean values for the concentration of each enzyme during the growth phase have been calculated. These results are discussed in relation to previous observations made on the growth of mouse LS cells.

    Notizen: A laboratory process for the isolation of acid soluble nucleotides from bacteria has been scaled-up. Escherichia coli was grown in 220-l. batches, harvested by centrifugation, and extracted with cold trichloracetic acid. The nucleotides in the extract were separated by adsorption onto a Dowex-1 ion exchange resin column followed by elution with dilute hydrochloric acid and lithium chloride. After concentration of the effluent, the lithium chloride was removed by extraction with organic solvents. The overall yield of nucleotides was 75-80%. This elution system has also been used for the separation of nucleotides from bakers' yeast extracts.

    Notizen: Dried vaccinia virus was found to be very stable in aerosol form in our chamber at a temperature of 75°F and a relative humidity (RH) of 85% when the virus was protected with 1.5% lysine, 1.5% sodium glutamate, 0.5% isoniazid, 0.5% thiourea, and either 2.5% heart infusion broth (HIB) or a combination of 3.75% lactose plus 1.25% raffinose prior to freeze-drying. Utilization of the Environmental chamber technique resulted in (a) selection of two diluents which protected vaccinia virus against the effects of high relative humidity, (b) measurement of the effect of various chemical additives in the diluents, and (c) quantitative measurement of the moisture absorbed by the various dried products.

    Notizen: A strain of Klebsiella aerogenes was selected which gave marked diauxic growth in a batch system on a mixture of glucose and lactose in a simple salts medium; the diauxic lag was 15-20 hr. at 30°C. The growth of this organism on glucose and lactose was studied in a single-stream two-stage continuous-stirred fermentor system over a wide range of flow rates. Glucose was metabolized instantaneously to give very low reactor concentrations at all flow rates, but the time lag before lactose was attacked, when present for the first time, was never less than 40 hr. at low feed rates, rising to 60 hr. at higher rates. The adaptation to lactose of cells in the first vessel lagged behind that in the second vessel but eventually both sugars were completely utilized in the first vessel except at very high dilution rates. At these feed rates, lactose utilization was not only prevented completely in the first vessel but also could be delayed almost indefinitely in the second vessel at the highest dilution rates; thus the lactose passed unchanged through both vessels. Once the enzymes required for lactose utilization had been induced, this ability to use lactose was retained, even in the absence of lactose, for very long periods of time under continuous conditions. Thus on presenting lactose for the second and subsequent occasions it was immediately metabolized. The significance of these results is discussed.

    Notizen: The broth rheologies and morphologies of three actinomycetes (Saccharopolyspora erythraea, Actinomadura roseorufa, and Streptomyces rimosus) in submerged culture have been examined. The rheology of all the broths became pseudoplastic as soon as significant growth occurred with the power law index, n, falling to 0.20 to 0.25. The consistency index, K, rose with biomass concentration although in some instances it fell later in the fermentation. The mean main hyphal lengths of all cultures were in the range, 15 to 25 μm, and did not alter greatly even when large changes in K were occurring. © 1995 John Wiley & Sons, Inc.
