    Notes: Approximate methods are developed and evaluated for treating the rate of binding ligands that cover several contiguous sites to a homogeneous one-dimensional lattice, which represents a nucleic acid or other linear biopolymer. The model requires as input only the number of lattice sites necessary for binding, the total number (possibly infinite) of lattice sites, and elementary rate constants for the cooperative and noncooperative association and dissociation of the ligand on the lattice. The computational methods employed are an extension of the triplet closure approximation from the helix-coil (single-site ligand) problem to the large ligand binding problem. It is found that consideration of clusters of n + 2 lattice sites, where each ligand covers n sites, gives a surprisingly accurate description of the kinetics. The approximation is implemented by an extension of the matrix-iteration approach proposed by Craig and Crothers. The effects of the finite lattice length, as well as the capability to treat ligand motion along the lattice, are incorporated. When all symmetries are taken into consideration, the time required for the matrix iteration calculation rises only linearly with the ligand length n and is considerably less than that of the Monte Carlo method, which is used as a standard for comparison.

    Notes: We consider the irreversible dissociation kinetics of proteins that bind cooperatively and nonspecifically to DNA. Our model consists of an infinitely long one-dimensional nucleic acid lattice on which are bound protein ligands. A set of adjacent bound proteins forms a cluster of length n. A protein molecule may dissociate from any site within the bound cluster, not only from the ends, as was assumed in a previous model of this process due to Lohman [(1983) Biopolymers 22, 1697-1713]. By considering this additional pathway, we present a more general treatment of the dissociation kinetics of cooperatively bound ligands. We show that dissociation from the (n-2) internal positions of an n-cluster is an important pathway when the initial fractional saturation of the lattice is close to unity and the co operatively is low. When the fractional saturation is initially equal to 1 and the co operatively is low, our model does not give the zero-order dissociation kinetics predicted by the Lohman model.

    Notes: Syntheses of parent polyaniline and methyl, methoxy, and ethoxy ortho-substituted polyanilines were performed using the conventional chemical methodology and monitored using the new open-circuit-potential (Voc) profile technique. The intermediate pernigraniline oxidation state was identified and isolated at the Voc maximum (A) during the conventional chemical synthesis of poly(o-methoxyaniline) in the emeraldine oxidation state. The introduction of the substituent on the aniline ring leads to longer polymerization times and lower Voc values. Syntheses in the presence of two different monomers in solution were also investigated and showed preferential polymerization of the monomer with the lowest Voc potential. All polymers produced were characterized by elemental analysis, gel permeation chromatography, UV-VIS spectroscopy, and cyclic voltammetry. The influence of the substituent on the Voc profile and on the polymer characteristics are rationalized in terms of steric and electronic effects. © 1995 John Wiley & Sons, Inc.

    Notes: Summary A correlative radioimmunoassay (RIA) and immunocytochemical (ICC) study was carried out on vasopressin (VP) distribution and content in brains of normal and 3-day water-deprived rats. By RIA there were statistically significant differences in brain VP per pg/mg between normal and osmotically stressed specimens in hypothalamus (338.4 versus 134.4), thalamus (4.8 versus 0.9), septum (18.0 versus 3.4), striatum (1.6 versus 0.7) and amygdala (17.3 versus 1.3), but not in other brain regions measured. Pituitary VP decreased from 71.1 to 8.7 ng/mg, and plasma VP rose from 3.6 to 19.3 pg/ml during water deprivation. Application of the peroxidase-anti-peroxidase ICC method of Sternberger to vibratome sections showed that VP-immunoreactivity in dehydrated specimens decreased in perikarya of paraventricular nucleus and suprachiasmatic nucleus, while intrahypothalamic immunoreactive magnocellular fibers appeared more conspicuous due to proliferation of large Herring bodies. In extrahypothalamic sites VP-immunoreactivity in water-deprived rats was visibly reduced in periventricular thalamus and septum. Thus it is apparent that both intra- and extrahypothalamic VP are affected by osmotic stress, and these results are discussed within the context of current ideas relating to co-activation of neurosecretory cells that project to different sites.

    Notes: Summary Subcellular distribution of a thiol-proteinase inhibitor protein was determined in the epidermis of the newborn rat by light and electron microscopy. This protein was highly soluble in basal cells and concentrated on ribosomes in the perinuclear region. Solubility in Tris buffer decreased in granular and cornified cells in which the protein appeared on polysomes which were attached on other cellular structures such as dense homogenous deposits and tonofilaments. The protein also appeared to be deposited on the plasma membrane and became insoluble in Tris buffer at 37° C, but solubilized in 1 M phosphate buffer. Location of the protein around keratohyalin granules or by the plasma membrane suggested that the inhibitor protein bound to cysteinerich protein of the epidermis with or without forming a thiol-proteinase inhibitor complex. The thiol-proteinase inhibitor protein seems to contribute to epidermal cell differentiation at multiple points through changes in its solubility and subcellular localization from basal cells to cornified cells.

    Notes: The dilute solution properties of copolymers are briefly discussed in relation to those of the parent homopolymers. It is shown that copolymer molecules are usually more expanded in solution than would be expected from the averaged behavior of the pure polymers, because of repulsive interactions between the unlike monomer units. A thermodynamic parameter χAB characterizing these interactions can be derived from measurements of the dilute solution properties of copolymers. In favorable cases this parameter can be independently evaluated from studies of ternary systems composed of the two parent homopolymers and a solvent, thus allowing prediction of the behavior of the copolymer. Light scattering and viscosity measurements on fractions of approximately equimolal copolymers of styrene and methyl methacrylate are presented and analyzed. The values of χAB deduced from the results in two solvents agree satisfactorily with each other, but are somewhat larger than those earlier obtained from measurements on ternary systems.

    Notes: The influence of fiber type and fiber-surface properties on matrix flow behavior was investigated using structural reaction injection-molding (SRIM). The influence of fiber type, fiber-surface properties, and matrix type on strength properties in elastomeric composites reinforced with nonwoven fibrous structures was investigated using tensile tests on elastomer composite samples from SRIM and latex coagulation (LC) fabrication methods and the microbond strength method on individual fibers. The fibers used were PET, LLDPE, and p-aramid. Fibers were treated with epoxy, styrene, and isocyanate derivatives, which make the surface chemically reactive. Treatments were also made with NaOH and a copolymer of polyester and polyol ether, causing a change in the fiber surface energy. The matrix types were polyurethane elastomer and natural rubber. The results show that the surface treatments which produced a change in the surface energy influenced the flow rate of the matrix polymer during the composite fabrication process. The treatments resulted in chemically reactive fiber surfaces which improved the fiber-matrix bond strength without affecting the Young's modulus of the composite material. Good correlation was found between bond strength and surface energy including the dispersive component of surface energy in the case of polyurethane elastomer and surface-modified PET fibers. The age of the polyurethane matrix has a marked influence on the bond strength. The fiber volume fraction in composites has a strong influence on the Young's modulus of the elastomer composite. © 1995 John Wiley & Sons, Inc.

    Notes: A process has been developed by which very high-modulus semicrystalline polymer films can be extruded continuously from a melt. This is accomplished by controlled cooling of the melt in a two-stage flow channel. A temperature gradient along the flow channel quenches the melt prior to an area reduction in which the polymer undergoes solid-state orientation. Analysis of high-density polyethylene tapes extruded by this process shows that they have properties similar to samples hydrostatically extruded at 120°C. Infrared analysis was used to determine both the degree of crystallinity and degree of orientation in these tapes as well as previously prepared hydrostatically extruded samples.
