    Notes: Abstract Acetyl coenzyme A (CoA) biosynthesis in spinach chloroplasts has been investigated by following the incorporation of bicarbonate and acetate into fatty acids under a variety of conditions. Both substrates were readily incorporated into fatty acids in a light-dependent manner by intact photosynthesising chloroplasts, but when the concentrations of these substrates were adjusted to those found in vivo, i.e. 200 μM acetate, 10 μM bicarbonate, then acetate was found to supply carbon atoms for fatty acids biosynthesis via acetyl CoA at forty times the rate of bicarbonate. It is proposed that extra-chloroplastic free acetate is the pricipal substrate for chloroplasts acetyl CoA biosynthesis in spinach.

    Notes: Abstract Of the total adenylate-kinase activity in 10-d-old barley and wheat leaves, 40–50% is localised in the chloroplasts, while in mature spinach leaves 50–70% of the enzyme is chloroplastic. The extra-chloroplastic adenylate-kinase activity is associated with the mitochondria, very little, if any, is freely soluble in the cytoplasm. The adenylate pool of the cytoplasm could have access to adenylate-kinase activity in the intermitochondrial space because of the free permeation of adenylates across the outer mitochondrial membrane. Thus the adenylate pool of the cytoplasm could be subject to adenylate-kinase equilibrium. The mitochondrial adenylate kinase appeared to the localised exclusively in the intermembrane space.

    Notes: Abstract (Ammonia plus 2-oxoglutarate)-dependent O2 evolution by intact chloroplasts was enhanced three- to five fold by 2 mM L- and D-malate, attaining rates of 9–15 μmol mg-1 Chl h-1. Succinate and fumarate also promoted activity but D-aspartate and, in the presence of aminooxyacetate, L-aspartate inhibited the malate-promoted rate. A reconstituted chloroplast system supported (ammonia plus 2-oxoglutarate)-dependent O2 evolution at rates of 6-11 μmol mg-1 Chl h-1 in the presence of MgCl2, NADP(H), ADP plus Pi (or ATP), ferredoxin and L-glutamate. The concentrations of L-glutamate and ATP required to support 0.5 V max were 5 mM and 0.25 mM, respectively. When the reaction was initiated with NH4Cl, O2 evolution was preceded by a lag phase before attaining a constant rate. The lag phase was shortened by addition of low concentrations of L-glutamine or by preincubating in the dark in the presence of glutamate, ATP and NH4Cl. Oxygen evolution was inhibited by 2 mM azaserine and, provided it was added initially, 2 mM methionine sulphoximine. The (ammonia plus 2-oxoglutarate)-dependent O2 evolution was attributed to the synthesis of glutamine from NH4Cl and glutamate which reacted with 2-oxoglutarate in a reaction catalysed by ferredoxin-specific glutamate synthase using H2O as the ultimate electron donor. The lag phase was attributed to the establishment of a steady-state pool of glutamine. L-Malate did not affect the activity of the reconstituted system.

    Notes: Abstract A reconstituted spinach chloroplast system containing thylakoids, stroma and 0.1 mM NADPH supported O2 evolution in the presence of oxidised glutathione (GSSG). The properties of the reaction were consistent with light-coupled GSSG-reductase activity involving H2O as eventual electron donor. The reconstituted system also supported dehydroascorbate-dependent O2 evolution in the presence of 0.6 mM reduced glutathione (GSH) and 0.1 mM NADPH with the concomitant production of ascorbate. The GSSG could replace GSH in which case the production of GSH preceded the accumulation of ascorbate. The data are consistent with the light-dependent reduction of dehydroascorbate using H2O as eventual electron donor via the sequence H2O→NADP→GSSG→dehydroascorbate. Approximately 30% of the GSH-dehydrogenase activity of spinach leaf protoplasts is localised in chloroplasts: this could not be attributed to contamination of chloroplasts by activity from the extrachloroplast compartment. Washed intact chloroplasts supported the uptake of ascorbate but the uptake mechanism had a very low affinity for ascorbate (Km approximately 20 mM). The rate of uptake of ascorbate was less than the rate of light-dependent reduction of dehydroascorbate and too slow to account for the rate of H2O2 reduction by washed intact chloroplasts.

    Notes: Abstract Transketolase (E.C. 2.2.1.1.) has been partially purified from wheat (Triticum aestivum, cv. Sappo) and spinach (Spinacia oleracea) leaves. The fully-active enzyme is a tetramer of relative molecular mass (Mr) of 150 kMr requiring thiamin pyrophosphate for maximal activity, and dissociating into a 74 kMr dimer in its absence or in dilute solution. The chloroplastic transketolase (over 75% of the cellular total) is magnesium-stimulated but the cytosolic form is magnesium-insensitive. Both chloroplastic and cytosolic transketolase showed similar broad specificities towards several ketose phosphate substrates including fructose 6-phosphate and sedoheptulose 7-phosphate. Wheat and spinach leaf transketolases are not light-activated and closely resemble the yeast enzyme in many of their properties.

    Description: Measurements of the acoustic impedance of a two-dimensional Helmholtz resonator, i.e., a cavity coupled to the environment through a high-aspect ratio slot are presented as a function of the velocity of a steady flow above the orifice of the resonator. The structure of the unsteady flow above the orifice is outlined from hot-wire measurements. A simplified model of it is proposed and analyzed, leading to a successful correlation of both the resistance and the reactance of the resonator.

    Description: Annoyance expressed in a railway noise survey is compared with that from two road traffic and three aircraft surveys in order to determine whether responses to various types of environmental noise are source-specific. Railway noise is found to be less annoying than other noises at any given high noise level. Railway noise annoyance rises less rapidly with increasing noise level. At high levels, this gap in reactions averages about 10 dB; it ranges from 4 dB to more than 20 dB. The methods used for comparing the surveys are examined. It is found that methodological uncertainties lead to imprecise comparisons and that different annoyance scales yield different estimates of intersurvey differences.

    Description: A model is developed for the effects of uniform and boundary-layer mean flow on the attenuation and propagation of harmonically excited sound waves in an extended reaction lined cylindrical duct. A duct geometry consisting of an annular outer region of bulk material surrounding an inner cylinder of air is utilized. A numerical solution is obtained for the coupled wave equations governing the motion of the sound in both the inner and annular regions. It is found that the numerically predicted attenuation and propagations constants are in excellent agreement with measured values using Kevlar as the liner material for plane-wave mode (O,O) excitation over a wide range of mean flows and sound frequency. The boundary-layer effects are determined to be unimportant, at least for plane-wave sound. In addition, numerical studies indicate small differences between the use of either the radial velocity or the radial displacement boundary conditions.
