    Notizen: Summary We have investigated the intragenomic DNA sequence homologies of twelve species of birds representing five orders, and emphasizing Galliformes. This study differs in two important ways from the classical approaches taken in constructing and evaluating phylogenies based on DNA sequence similarities. Comparisons are made on the basis of sequence homologieswithin genomes of related birds, rather than between genomes. DNA is reassociated at 50°C in 0.5M phosphate buffer; these conditions allow formation and detection of duplexes containing more mismatch than would normally be permitted using more stringent conditions, affording an opportunity to observe more ancient sequence homologies. Thermal stability profiles of DNA duplexes formed under these conditions are the basis of comparison; three general patterns were observed. This approach emphasizes differences in sequence composition between genomes while the more traditional method of intergenomic tracer DNA hybridization at higher stringency emphasizes sequence similarities. No correlation was found between taxonomic position and intragenomic sequence composition, either within or between lineages. The thermal stability profiles of DNA duplexes formed within avian genomes did not reflect the biological similarities inferred from morphology, karyotype, and studies of interspecific hybridization. While all of the differences observed could have occurred over geological time, it was surprising that the genomes of the domestic chicken and the Red Jungle Fowl (Gallus gallus) differ in their sequence compositions. It appears that amplification/reduction events and/or positional changes occur rather often during evolution of a lineage.

These traits were (i) paired vs. single female spikelets; (ii) two-ranked vs. many-ranked ears; (iii) non-indurated vs. indurated glumes; (iv) inclination of the kernels toward the rachis, and (v) distichous vs. polystichous central staminate spike. All traits showed a simple mode of inheritance except for paired female spikes, which appeared to be controlled by two genes. The loci controlling these major changes were mapped with RFLP markers to four chromosomal regions. These results support the suggestion that maize became differentiated from teosinte with as few as five major gene changes.

    Notizen: Abstract We investigated the reproductive biology of Etheostoma chienense in the Bayou du Chien drainage of western Kentucky. Etheostoma chienense is similar in its ecology and reproductive biology to other members of the E. squamiceps complex. However, E. chienense is opportunistic in its choice of egg deposition substrates, as nests were found on a variety of natural and anthropogenic items. Due to an apparent lack of suitable spawning substrates, we added half-cylindrical ceramic tiles to several stretches of stream to increase potential nest productivity. Egg-clutches attached to artificial substrates were twice the size of egg clutches attached to naturally occurring materials. Laboratory experiments were conducted to determine nest rock size and mate choice preferences. Both the size of the male and of the nest rock appeared to be important parameters to spawning females. The life history information gathered here has significant management implications. Artificial spawning substrates should be placed in appropriate microhabitats (i.e., shallow, low-flow reaches in headwaters). Nest cavity vertical height should be about 3.0 cm and tiles should be spaced at least 0.5 m apart. We feel the use of surrogates (e.g., E. oophlyax) to investigate other types of spawning cover, and restoration of riparian buffer zones among other actions, would particularly benefit recovery efforts for this endangered species.

    Notizen: Abstract Muscular fatigue in the training athlete or military recruit has been hypothesized to cause increased bone strain that may contribute to the development of a stress fracture. Under normal circumstances, muscles exert a protective effect by contracting to reduce bending strains on cortical bone surfaces. In vivo strain studies in dogs show that muscle fatigue following strenuous exercise elevates bone strain and changes strain distribution. However, a similar experiment has yet to be performed in humans. The purpose of this work was to test the hypothesis in humans that strenuous fatiguing exercise causes an elevation in bone strain. It was also hypothesized that this elevation is greater in younger people than in older people due to the decline in muscle strength and endurance that normally occurs with age. To test these hypotheses, strain in the tibiae of seven human volunteers was measured during walking before and after a period of fatiguing exercise. Neither hypothesis was sustained. Post-hoc analysis of the strain data suggests that strain rate increases after fatigue with a greater increase in younger as opposed to older persons. Although not conclusive, this suggests that it is strain rate, rather than strain magnitude, that may be causal for stress fracture. © 1998 Biomedical Engineering Society.
