    Notes: Bovine β-1,4-galactosyltransierase is an efficient catalyst for the regioselective transfer of galactose from UPD-galactose, generated in situ with the UDP-glucose/UDP-glucose-4-epimerase system, to the kaurane glycosides stevioside (1) and Steviolbioside (2), affording the corresponding galactosyl derivatives 3 and 4 in high yields. By a combination of 2D NMR techniques (COSY, TOCSY, ROESY, HMQC, and HMBC), the structure of the products is established as 13-[(β -D-galactopyranosyl-(1 → 4)-β-D-glucopyranosyl-(1 → 2)- β -D-glucopyranosyl)oxy]kaur-16-en-19-oic acid β -D-glucopyranosyl ester (3) and 13-[(β-D-galactopyranosyl-(1 → 4)- β-D-glu-copyranosyl-(1 → 2)- β-D-glucopyranosyl)oxy]kaur-16-en-19-oic acid (4).

    Notes: The conversion of the enzymatic hydrolysate of shellfish chitin waste to single-cell protein was investigated as part of a comprehensive waste treatment program. Forty-two yeasts were screened for ability to assimilate the monomer of chitin, N-acetylglucosamine, which has been shown to be the sole product of enzymatic hydrolysis of chitin. The Yeast Pichia Kudriavzevii was selected for study, based on ability to grow at high temperature (37°C and above), low pH (4.0 ± 0.5), and in a nutritionally simple medium. Growth rates of P. kudriavzevii were similar on N-acetylglucosamine and on the chitin hydrolysate. Dependencies of specific growth rate on temperature, pH, medium composition, and oxygen tension were studied. The variations of yield, protein content, and total nucleic acid content with the specific growth rate were evaluated. The amino acid distribution of the protein of P. kudriavzevii was obtained.

    Notes: A 10.5-m3 concentric tube jet loop reactor was used to study the influence of the working liquid volume, mean superficial air velocity, operating pressure, downcomer aeration, liquid jet velocity, and two ratios of draft tube/reactor diameter (Dt/D) on liquid circulation time (Tc). The experiments were carried out in a water-air system with the use of the acid pulse method. Results showed that circulation time was independent of the working liquid volume over a certain minimum liquid level, whereas downcomer aeration and Dt/D ratio appeared as amenable parameters to achieve a high degree of control over liquid circulation and mixing efficiency, and to optimize the overall reactor performance. Increasing the operating pressure caused a reduction of the liquid circulation rate. However, ionger residence times of the air bubbles and the higher mass transfer driving force that result at higher pressures improve oxygen utilization. The relationship between Tc and air load was independent of the operating pressure, provided the correlation is given as a function of the mean superficial air velocity. Neither liquid circulation nor gas holdup were significantly affected by liquid jet velocity. © 1995 John Wiley & Sons, Inc.

    Notes: Biotechnological techniques, including biofilters and biotrickling filters are increasingly used to treat air polluted with VOCs (Volatile Organic Compounds). In this work, the start-up, the effect of the gaseous ammonia addition on the toluene removal rate, and the problems of the heat accumulation on the performance of a laboratory scale biofilter were studied. The packing material was sterilized peat enriched with a mineral medium and inoculated with an adapted consortium (two yeast and five bacteria). Start-up showed a short adaptation period and an increased toluene elimination capacity (EC) up to a maximum of 190 g/m3/h. This was related to increased CO2 outlet concentration and temperature gradients between the packed bed and the inlet (Tm-Tin). These events were associated with the growth of the microbial population. The biofilter EC decreased thereafter, to attain a steady state of 8 g/m3/h. At this point, gaseous ammonia was added. EC increased up to 80 g/m3/h, with simultaneous increases on the CO2 concentration and (Tm-Tin). Two weeks after the ammonia addition, the new steady state was 30 g/m3/h. In a second ammonia addition, the maximum EC attained was 40 g/m3/h, and the biofilter was in steady state at 25 g/m3/h. Carbon, heat, and water balances were made through 88 d of biofilter operation. Emitted CO2 was about 44.5% of the theoretical value relative to the total toluene oxidation, but accumulated carbon was found as biomass, easily biodegradable material, and carbonates. Heat and water balances showed strong variations depending on EC. For 88 d the total metabolic heat was -181.2 × 103 Kcal/m3, and water evaporation was found to be 56.5 kg/m3. Evidence of nitrogen limitation, drying, and heterogeneities were found in this study. © 1998 John Wiley & Sons, Inc. Biotechnol Bioeng 60: 483-491, 1998.

    Notes: Lipase from Aspergillus niger was used for the selective hydrolysis of the 3-O-acetate of cephalosporin C to give an intermediate useful for further chemical elaborations. This lipase was purified to homogeneity and its properties compared with previously published data that present some discrepancies. The lipase proved to be very effective in catalyzing 3-O-acetate hydrolysis and versatile toward substitution on the β-lactamic ring. In fact, as an example, two other cephalosporinic derivatives, cephalotin and cefotaxime, were efficiently deacetylated. The lipase was immobilized on Eupergit C and employed continuously in either a column or a batch reactor for 2 months without appreciable loss of activity. © 1996 John Wiley & Sons, Inc.

    Notes: The transesterification activity, autolysis, thermal stability and conformation of subtilisin Carlsberg, made soluble in dioxane by covalent linking to methoxypoly(ethylene glycol) (PEG), were investigated as a function of the concentration of water in the medium. Electrospray mass spectrometry showed that the modified enzyme preparation was a mixture of proteins containing from 2 to 5 covalently linked PEG chains per subtilisin molecule. PEG-subtilisin catalyzed transesterification between vinyl butyrate and 1-hexanol was optimum at 0.55 MH2O, while hydrolysis prevailed above 2 MH2O. There was a decrease in the overall enzyme activity with increasing water concentration because of autolysis and denaturation of the enzyme. Subtilisin powder and celite-immobilized subtilisin were more stable and less susceptible to autolysis than the PEG-modified enzyme. Circular dichroism and intrinsic protein-fluorescence studies showed that the conformation of PEG-subtilisin did not change as a function of water concentrations between 0 and 9 M. The Km,app value of PEG-subtilisin for 1-hexanol was highly influenced by water, which behaved as a competitive inhibitor in the transesterification reaction with an affinity for the enzyme similar to that of the alcohol. The Km,app for the acylating agent was not significantly modified by water. Lyoprotectants such as sorbitol and free PEG did not influence the activity of PEG-subtilisin but notably increased the activity of subtilisin powder and celite-immobilized subtilisin. The addition of 1.7-5.5 M water, however, rendered enzyme preparations containing no additives as active as those containing the lyoprotectants. © 1997 John Wiley & Sons, Inc.     Notes: Proton transfer processes in both the ground and excited states in anils of aromatic α-hydroxyaldehydes (salicylaldehyde, 2-hydroxynaphthalene-1-carbaldehyde and the novel 10-hydroxyphenanthrene-9-carbaldehyde) have been studied by a combination of spectroscopic techniques. Solution 1H and 13C NMR is used to establish the position of the tautomeric equilibria. UV-visible absorption and fluorescence spectral data help to characterize the existence, in all cases, of excited-state intramolecular proton transfer (ESIPT) pheonmena. Semi-empirical calculations involving full geometry optimization and calculation of heats of formation for the ground state (AM1) and vertical excitation energies and oscillator strengths (INDO/S) are in agreement with the experimental observations.

    Notes: A reproducible, rapid procedure for the determination of glutathione in human blood by micellar electrokinetic chromatography has been developed. Whole blood samples were deproteinized with 5% w/v sulfosalicylic acid (final concentration). After centrifugation, the supernatant was directly injected for analysis, without further derivatization. Separations were performed by using an uncoated capillary of 30 cm effective length and 50 μm internal diameter (ID), 50 mM Tris-HCl, 30 mM sodium dodecyl sulfate (SDS), pH 7.00, as running buffer, and 10-20 kV. On-line detection was carried out at 214 nm and a detection limit in the range of femtomoles was achieved. Under the same experimental conditions, we resolved a mixed standard solution containing glutathione in its oxidized and reduced forms, lipoamide and α-lipoic acid. The corresponding migration times were reproducible. The present method allows rapid determination of these compounds, which play a critical role in oxidative stress, in cellular defense against injurious agents and whose levels are related to the toxicology and metabolism of several toxins and drugs, such as antineoplastic agents.
