    Notes: Ab initio MO calculations, using both minimal (STO-3G) and extended (Roos-Siegbahn) basis sets are reported for the systems methanethiol-imidazole, methanethiol-imidazole-formaldehyde, and methanethiol-imidazole-formamide, which, together with a point-change representation of a long α-helix, form models for the active site of papain. It is shown that the large electric field exerted by the helix in the active-site region is responsible for the presence of the essential residues Cys 25 and His 159 in the form of an ion pair RS- ··· ImH+, which is crucial for a recently proposed mechanism for the catalytic action of the enzyme. Also, an explanation is given for the anomalies in measured pK values for these residues. Detailed studies on the (sub)systems show that minimal basis sets lack the flexibility necessary for describing the type of proton transfer involved. We conclude that α-helices are essential parts of enzymes and that they play a significant role in the catalytic process.

    Description: The rearing of the botfly Cephenemyia auribarbis (Diptera: Oestridae) After several failures we finally succeeded in rearing botflies Cephenemyia from third-instar larvae. From a red deer shot at 13 March 1998 about one hundred larvae of Cephenemyia auribarbis (Meigen, 1824) were collected. Ten individuals reached the pupal stage. After 22 days, one fullgrown male died in the pupa, and two males emerged and lived for 17 and 18 days respectively. This is one of the few recorded succesful attempts to rear botflies.

    Notes: Abstract   Rhizomania is a serious disease of sugar beet, caused by beet necrotic yellow vein virus (BNYVV). The disease can only be controlled by the use of resistant cultivars. The accession Holly contains a single dominant gene for resistance, called Rz. The identification of a locus for resistance that differs from Rz would provide possibilities to produce cultivars with multiple resistance to BNYVV. Inheritance of resistance to BNYVV was studied by screening progenies of crosses between resistant plants of the accessions Beta vulgaris subsp. maritima WB42 and B. vulgaris subsp. vulgaris Holly-1–4 or R104. Observed and expected segregation ratios were compared to elucidate whether the resistance genes in the three accessions are alleles or situated on different loci. STS markers, linked to the genes for resistance, were used to study the segregation in more detail. The results demonstrated that the genes for resistance to BNYVV inHolly-1-4 and WB42 are closely linked. The gene for resistance in R104 is at the same locus as in Holly-1-4, and also closely linked to the gene in WB42. As the Holly resistance gene has been named Rz, the name Rz2 is proposed to refer to the resistance gene in WB42. Consequently, the gene Rz should be referred to as Rz1.

    Notes: Summary Experiments were carried out for adding the chromosome carrying resistance to beet root nematode (Heterodera schachtii) from the wild Beta species of the section Patellares (B. procumbens, B. webbiana and B. patellaris) to the genome of B. vulgaris. Preliminary experiments indicated that crosses between the wild species and B. vulgaris cultivars of the mangold type yielded on average more viable F1 hybrids than crosses with sugar and fodderbeet. However, crossability varied strongly between individual parental combinations. It was concluded that most types of B. vulgaris can be hybridized with the wild species of the section Patellares if a sufficient number of pair-crosses is made. Crosses between diploid cultivars or species of the section Vulgares and diploid wild species of the section Patellares yielded many hybrids which, however, were highly sterile. From crosses between tetraploid B. vulgaris and the wild species a great number of viable allotriploid and allotetraploid hybrids was obtained. In the backcross progenies of allotriploid hybrids 26% alien monosomic additions occurred, of which 4.1% carried the resistance bearing chromosome of B. procumbens or B. patellaris. The programme will be continued by sereening progenies of the resistant monosomic addition plants for the occurrence of resistant disomic introgression products.

    Notes: Abstract Erythromycin (ERT) has been shown to reduce the 5-aminolevulinic acid (ALA) synthesizing capacity of a normal (N) chlorophyllous sugarbeet callus, grown under light, in contrast to a habituated achlorophyllous non-organogenic (HNO) callus of the same species. Similar effects were obtained on total hemes and on catalase which is a hemoprotein used as marker. The effect of ERT, which is an inhibitor of plastid differentiation and of chlorophyll synthesis, was reversed in the N callus by a supply of glycine and succinate. The compounds are the precursors of ALA synthesized through 5-aminolevulinic acid synthase (ALAS) which is implied in the Shemin pathway. The involvement of ALAS appeared to be favoured when plastids were undifferentiated (HNO callus) or when plastids were inefficient (N callus under darkness or under light after ERT treatment).

    Notes: Abstract In vitro vegetative multiplication of sugarbeet was obtained by culturing of inflorescence explants. Subapical segments or 5-mm-long tips from nine varieties developed axillary shoots (up to 50 per tip) on a medium containing indolebutyric acid (IBA) and benzylaminopurine (BAP). Zeatin was ineffective as cytokinin. Gibberellic acid (GA3) enhanced the process. Such vegetative shoots were subsequently isolated and were each allowed to develop up to 20 supplementary axillary shoots on a multiplication medium containing IBA, BAP, and naphthaleneacetic acid (NAA). Rooting of shoots was obtained in the absence of growth regulators and plants were established.

    Notes: Die Darstellung der freien Farbsäuren des Betanins und Betanidins wird beschrieben, ihre Bruttoformeln werden erörtert. Beide Verbindungen sind optisch aktiv. Sie spalten mit starker Natronlauge in der Hitze etwa 1/3 ihres Stickstoffes als Ammoniak ab. Quantitative Decarboxylierungsversuche und potentiometrische Titration weisen für das Glucosid wie für das Aglykon auf drei Carboxylgruppen, von denen eine durch eine basische Gruppe neutralisiert zu sein scheint, und eine schwach saure (phenolische) Gruppe hin. Beim oxydativen Abbau des Betanidins wurde L-Asparaginsäure erhalten.

Die Spaltung ist eine innere Disproportionierung; als Mechanismus wird eine β-(oder vinyloge β-)Eliminierung diskutiert. - Sieben einseitig hydroxyl-substituierte Diphenyläther wurden neu dargestellt.
