    Description: The gene for von Recklinghausen neurofibromatosis (NF1), one of the most common autosomal-dominant disorders of humans, was recently mapped to chromosome 17 by linkage analysis. The identification of two NF1 patients with balanced translocations that involved chromosome 17q11.2 suggests that the disease can arise by gross rearrangement of the NF1 locus, and that the NF1 gene might be identified by cloning the region around these translocation breakpoints. To further define the region of these translocations, a series of chromosome 17 Not I-linking clones has been mapped to proximal 17q and studied by pulsed-field gel electrophoresis. One clone, 17L1 (D17S133), clearly identifies the breakpoint in an NF1 patient with a t(1;17) translocation. A 2.3-megabase pulsed-field map of this region was constructed and indicates that the NF1 breakpoint is only 10 to 240 kilobases away from 17L1.     Notes: Based on the substrate specificity for 5-lipoxygenase and the known stereochemical course of the reaction, a hypothetical model of the enzyme active site was developed and used to design 2 types of selective inhibitors of 5-lipoxygenase. Both inhibitor types used aromatic rings in place of (Z)-olefins of the substrate and were designed to mimic the nonpolar end of arachidonic acid. One inhibitor type used a carboxylic-acid interaction with the O-binding centre of the enzyme in analogy with known cyclooxygenase inhibitors, whereas a second type employed a hydroxylamine function to interact with a presumed tyrosine or cysteinyl radical predicted to be in the enzyme active site. Selective 5-lipoxygenase inhibitors were 7-(hexyloxy) naphthalene-2-acetic acid (1) and N-methyl;-N(7-propoxynaphthalene-2-ethyl)hydroxylamine (2). Structure-activity relationships for both types of inhibitors are discussed.

    Notes: The diffusion coefficient values of GaIII in potassium nitrate and potassium chloride supporting electrolytes have been obtained polarographically. The half-wave potential, E1/2, and E3/4-E1/4 values have also been determined. The E3/4 - E1/4 values confirm the irreversibility of GaIII reduction in KNO3 and KCl electrolytes.

    Notes: This study provides electrochemical and LCEC determination for three of the most abundant and most important isoflavones, Biochanin A, formononetin, and genistein. Cyclic voltammetry and pH-potential diagrams show that the isoflavones undergo pH-dependent irreversible oxidations. The LCEC determination of the isoflavones has been optimized for mobile phase composition and detection potential. Sub-nanogram detection limits have been achieved.

    Notes: Many techniques are available for the determination of diffusion coefficients of ions in aqueous solutions. A review of classical and modern techniques is presented. Non-electrochemical methods are discussed briefly. Electrochemical methods reviewed include chronoamperometry, chronopotentiometry, polarography, and hydrodnamics. Selected values of the diffusion coefficients determined by these electrochemical methods of the metallic ions AgI, BiIII, CdII, CuII InIII, PbII, TlI, and ZnII are presented.

    Notes: Cyclic voltammetry was used to characterize the oxidation-reduction behavior of a series of novel hexacoordinate silicon(IV) porphyrin complexes. Each (Por)SiX2 [Por = the dianions of the porphyrins: tetrakis(p-tolyl)porphyrin (TTP), tetraphenylporphyrin (TPP), tetrakis(p-trifluoromethylphenyl)porphyrin (TTFP), octaethylporphyrin (OEP), × = F, Cl, O3SCF3] complex undergoes one to two oxidations and one to two reductions, all occurring at the porphyrin ring. No silicon(IV) reduction is observed for any of the porphyrin complexes. The effects of solvent, supporting electrolyte, substituents on the porphyrin ring, axial ligands, certral metal, and ruffling of the porphyrin ring are described.

    Notes: Previous work has established the presence of an unbalanced chromosome abnormality [+der(1),t(1;7)(p11;p11)] in some therapy-associated myelodysplastic disorders. Recently the EGF receptor has been found to reside at 7p11. Using a probe specific for erb B oncogene, which encodes a truncated form of the EGF receptor, we examined RNA and DNA derived from bone marrow and peripheral blood mononuclear cells from three patients with myelodysplastic syndromes (MDS) and one with acute lymphocytic leukemia (ALL), all bearing an abnormal clone in their bone marrow with a similar unbalanced 1;7 translocation. DNA-excess slot blot hybridization to 5′-32p-labeled cellular RNA revealed from ten- to thirtyfold enhancement in accumulation of mRNA specific for erb B in both peripheral blood and bone marrow cells of the three MDS patients when compared to normal controls. In addition, enhancement of H-ras mRNA accumulation was detected in some, though expression of other genes such as actin, N-ras, myc, src, B-lym, and 20 other genes was not found to be enhanced. Increased erb B expression was not apparent in mononuclear cells from patients with other hematologic disorders such as chronic lymphocytic leukemia, Hodgkin's disease, or lymphoma. Southern blot analysis of restriction-enzyme-cleaved DNA from three MDS patients with an unbalanced 1;7 translocation revealed that erb B gene was amplified at least twentyfold in peripheral blood white blood cells, while levels of actin hybridization were comparable to those of the controls. No such amplification was evident in the ALL patient. Our data suggest that +der(1),t(1;7)(p11;p11) chromosomal anomalies can be specifically associated with amplification of erb B DNA and RNA sequences.

Neu synthetisiert werden 2-Methyl-bicyclo[3.2.1]octen-(2), 2-Methyl-bicyclo[2.2.2]octen-(2) und 3-Methyl-bicyclo[3.2.1]octen-(2) durch Dehydratisierung der entsprechenden tertiären Alkohole, 2-Methylen-bicyclo[3.2.1]octan, 2-Methylen-bicyclo[2.2.2]octan und 3-Methylen-bicyclo[3.2.1]octan durch Wittig-Reaktion aus den zugrundeliegenden Ketonen sowie 1-Methyl-bicyclo[2.2.2]octen-(2). Der Verlauf der Wasserabspaltungen wird diskutiert.
