    Notes: Summary Field studies ofPolyrhachis moesta revealed that colonies were facultatively founded by more than one queen, even though each established colony was inhabited by only one queen with her workers. In pleometrotic colonies, it was found that cofoundresses frequently exhibited cooperative behaviors, including trophallaxis. The results of DNA fingerprinting showed that multiple foundresses were not genetically to one another. It seems that they aggregate randomly and independently of their relatedness. A discussion of the reasons why foundresses ofP. moesta do not aggregate preferentially with genetically related females.

To evaluate further the signal transduction mechanisms involved in the short-term modulation of Na-K-ATPase activity in the mammalian kidney, we examined the role of phospholipase C-protein kinase C (PLC-PKC) pathway and of various eicosanoids in this process, using microdissected rat proximal convoluted tubules. Dopamine (DA) and parathyroid hormone (either synthetic PTH1-34 or PTH3-34) inhibited Na-K-ATPase activity in dose-dependent manner; this effect was reproduced by PKC530-558 fragment and blocked by the specific PKC inhibitor calphostin C, as well as by the PLC inhibitors neomycin and U-73122. Pump inhibition by DA, PTH, or arachidonic acid, and by PKC activators phorbol dibutyrate (PDBu) or dioctanoyl glycerol (DiC8) was abolished by ethoxyresorufin, an inhibitor of the cytochrome P450-dependent monooxygenase pathway, but was unaffected by indomethacin or nordihydroguaiaretic acid, inhibitors of the cyclooxygenase and lipoxygenase pathways of the arachidonic acid cascade, respectively. Furthermore, each of the three monooxygenase products tested (20-HETE, 12(R)-HETE, or 11,12-DHT) caused a dose-dependent inhibition of the pump. The effect of DA, PTH, PDBu or DiC8, as well as that of 20-HETE was not altered when sodium entry was blocked with the amiloride analog ethylisopropyl amiloride or increased with nystatin. We conclude that short-term regulation of proximal tubule Na-K-ATPase activity by dopamine and parathyroid hormone occurs via the PLC-PKC signal transduction pathway and is mediated by cytochrome P450-dependent monooxygenase products of arachidonic acid metabolism, which may interact with the pump rather than alter sodium access to it.

    Notes: Abstract Overcoming apoptosis resistance to chemotherapy and radiation may lead to a reduction in gastric cancer death. We hypothesize that the apoptotic machinery in gastric cancer cells is dependent upon specific cellular conditions. In the course of our study of the expression of apoptosis-related genes in human gastric cancer cell lines, we have identified a cDNA clone which predicts an alternative form of caspase-9. The caspase-9 variant, which we designated as caspase-9 beta, retained a truncated structure of native caspase-9 without its catalytic domain and was expressed in seven cell lines from human gastric cancer. Among the cell lines examined, MKN-28 cells, which exhibited the most resistance against apoptotic stimuli, expressed the highest level of caspase-9 beta. The induction of apoptosis by staurosporine or actinomycin D was markedly suppressed in caspase-9 beta-transfected HeLa cells. These results are consistent with our hypothesis that the caspase-9 beta may be an endogenous dominant-negative molecule which attenuates apoptotic activity in human gastric cancer cells.

    Notes: Abstract The supersaturated superfluid 3 He-4 He mixture in the presence of quantized vortices is considered. A specific type of the lability against the vortex core expansion arises from concentrating 3 He atoms onto the vortex core. This affects significantly the region of the 3 He concentrations at which the rate of nucleating stable 3 He-concentrated phase can be noticeable. The most clear case of the vortex structure is a straight-line vortex with the uniformly distributed 3He atoms along the core. Such lability leads to some features in the thermal activation and quantum tunneling nucleation, resulting, in particular, in a relatively small supersaturation of about 1% which can be achieved. The critical value of the supersaturations corresponding to the vortex lability is dispersed because of the finite size of the 3 He-concentrated phase along the vortex core. We analyze this dispersion and its effect on the quantum separation in supersaturated 3 He-4 He mixtures. The value of the supersaturation conserves its order of the magnitude, changing only by a numerical factor. We also emphasize an important difference of the coefficient in the term responsible for the lability from the superfluid density ρs. The supersaturation observed in experiments lies within the range of the vortex lability and, probably, is connected with this lability.
