    Notes: A high-speed data-collection system for large-unit-cell crystals is presented, using the Fuji Imaging Plate as an X-ray detector and a rotating-anode generator as the X-ray source. It is an automatic data-acquisition system that requires almost no manual intervention. The quality of data collected on the system is discussed. Merging R values ranged from 0.04 to 0.05. Compared with a four-circle diffractometer, data reproducibility was better, isomorphous/anomalous Patterson maps were almost identical in quality and data from a small-molecule crystal, cytidine, were of almost the same quality. Protein structures were refinable using the data measured on the system, the final crystallographic R value of the 2.2 Å 3-isopropylmalatedehydrogenase structure being 0.185 and that of the 1.88 Å Flammulina veltipes agglutinin structure being 0.199.

    Notes: The properties of a new blue-colored Imaging Plate (IP), Fuji type UR-III, were examined with respect to spatial resolution and sensitivity to Cu Kα radiation. The spatial resolution of the new IP, estimated from the full width at half-maximum of the diffraction profile of a glutathione synthetase crystal, was 270 μm, smaller than that in a conventional IP, Fuji type BA (330 μm) and larger than that in an X-ray film, Kodak DEF-5 (210 μm). An estimation of the peak-to-background ratios of diffraction spots from a hen egg white lysozyme crystal shows that the new IP is at least 25 times more sensitive to Cu Kα radiation than the X-ray film. This new IP is a useful area detector to collect diffraction data from large-unit-cell crystals.

    Notes: The design and performance of a new water-cooled and doubly bent crystal monochromator for Compton scattering experiments are described. Its mechanical system is similar to that of a usual bent cylindrical X-ray mirror. A monolithic Si (111) crystal is mounted on a water-cooled and cylindrically polished copper crystal holder with liquid Ga–In alloy between them in order to obtain good thermal and mechanical contact. The sagittal bending radius of the holder, Rs, is 508 mm. The holder is mounted on a mechanical bender making a meridian radius, Rm, of 596 m. The performance of the monochromator, which has been installed at the Photon Factory Accumulator Ring beamline NE1, is as follows. The focused beam size, flux and energy resolution for 60 keV X-rays are 0.5 mm in height and 2.0 mm in width, 5 × 1012 photons s−1 and about 60 eV, respectively. The new monochromator gives one order higher brightness at the sample position and also better energy resolution than the previous monochromator. The overall momentum resolution for high-resolution Compton scattering experiments becomes 0.08 atomic units, and the background can be reduced by a third, because it is possible to insert a fine slit after the analyser crystal without losing any Compton scattering signal.

    Notes: The electron-density map of an Si crystal is drawn by the maximum-entropy method (MEM) with 30 structure factors which were determined very accurately on an absolute scale by the Pendellösung method [Saka & Kato (1986). Acta Cryst. A42, 469-478]. It is shown that the following three beneficial points arise from the use of the MEM for accurate structure analysis. Firstly, a precise electron-density map can be obtained; the existence of bonding electrons is clearly visible in the maximum-entropy map, even though no forbidden reflections are included in the analysis. The structure factors calculated from the maximum-entropy map for the 222 and the 442 forbidden reflections show good agreement with those measured by different experiments. The final R factor was 0.05% as a consequence of the high accuracy of the measured structure factors. Secondly, there is no need to seek a better structural model which could be a complicated and time-consuming process in accurate structure analysis. Thirdly, the resolution of the maximum-entropy map is much higher than that of the map drawn by conventional Fourier transformation. It is also found in the case of silicon that phase information is not absolutely necessary because exactly the same density-distribution map can be obtained without any phase information as the map drawn with all the phase information.
