    Notes: Abstract This article addresses the methodological problem of the non-linear representation of philosophical systems in a computerized knowledge base. It is a problem of knowledge representation as defined in the field of artificial intelligence. Instead of a purely theoretical discussion of the issue, we present selected results of a practical experiment which has in itself some theoretical significance. We show how one can represent different philosophies using CODE, a knowledge engineering system developed by artificial intelligence researchers. The hypothesis is that such a computer based representation of philosophical systems can give insight into their conceptual structure. We argue that computer aided text analysis can apply knowledge representation tools and techniques developed in artificial intelligence and we estimate how philosophers as well as knowledge engineers could gain from this cross-fertilization. This paper should be considered as an experiment report on the use of knowledge representation techniques in computer aided text analysis. It is part of a much broader project on the representation of conceptual structures in an expert system. However, we intentionally avoided technical issues related to either Computer Science or History of Philosophy to focus on the benefit to enhance traditional humanistic studies with tools and methods developed in AI on the one hand and the need to develop more appropriate tools on the other.

    Notes: Abstract The 25 April 1992 Cape Mendocino earthquake generated a tsunami characterized by both coastal trapped edge wave and non-trapped tsunami modes that propagated north and south along the U.S. West Coast. Both observed and synthetic time series at Crescent City and North Spit are consistent with the zero-order edge wave mode solution for a semi-infinite sloping beach depth profile. Wave amplitudes at Crescent City were about twice that observed at North Spit, in spite of the fact that the source region was three times farther from Crescent City than North Spit. The largest observed amplitude was due to an edge wave which arrived almost three hours after the initial onset of the tsunami; since such waves are highly localized nearshore, this suggests that the enhanced responsiveness at Crescent City is at least partly due to local dynamic processes. Furthermore, the substantially delayed arrival of this wave, which was generated at the southern end of the Cascadia Subduction Zone, has significant implications for hazard mitigation efforts along the entire U.S. West Coast. Specifically, this study demonstrates that slow-moving but very energetic edge wave modes could be generated by future large tsunamigenic earthquakes in the CSZ, and that these might arrive unexpectedly at coastal communities several hours after the initial tsunami waves have subsided.

    Notes: Abstract We previously described the isolation of a variant subline of HL-60 cells that does not differentiate in response to nitric oxide (NO)-generating agents or to cGMP analogs [7]. The variant cells have normal guanylate cyclase activity and normal NO-induced increases in the intracellular cGMP concentration. We now show that the variant cells have normal cGMP-dependent protein kinase (G-kinase) activity, both by an in vitro and in vivo assay, and using two-dimensional gel electrophoresis we have identified six G-kinase substrates in the parental cells. Of these six proteins, we found considerably less phosphorylation of one of the proteins in the variant cells than in parental cells, both in vitro and in intact cells, and by 35S-methionine/35S-cysteine incorporation we found much less of this protein in the variant cells than in parental cells. The protein is a shared substrate of cAMP-dependent protein kinase (A-kinase); since cAMP analogs still induce differentiation of the variant cells, it appears that the NO/cGMP/G-kinase and cAMP/A-kinase signal transduction pathways share some but not all of the same target proteins in inducing differentiation of HL-60 cells.

    Notes: Abstract We have purified from human placenta a low molecular mass substance that inhibits cAMP-dependent protein kinase and activates protein kinase C. This protein kinase regulator was purified in three steps: (1) homogenizing placentas in chloroform/methanol and extracting the regulator into water; (2) eluting a strong anion exchange high performance liquid chromatography (HPLC) column with a quaternary gradient; and (3) eluting a reversed-phase HPLC column with a binary gradient. The regulator was found to be highly purified by HPLC, thin-layer chromatography (TLC) and laser desorption ionization mass spectrometry with a molecular mass of 703 Daltons by the latter procedure. The physical and biochemical properties of this protein kinase regulator suggest that it is a phospholipid but it did not co-elute by HPLC or by TLC with any of the known phospholipid activators of protein kinase C.

    Notes: Abstract A full-length cDNA clone encoding apple (Malus domesticus) polyphenol oxidase (PPO) was isolated from a fruit peel cDNA library. Southern analysis indicated that apple PPO is encoded by a divergent multigene family. By northern analysis, PPO mRNA was only detected in a fruit sample taken one week after full bloom. PPO mRNA accumulated in wounded tissues, and also in peel tissue showing the symptoms of superficial scald, a post-harvest disorder. The induction of PPO mRNA provides the first evidence for transcriptional control of PPO expression after wounding or the manifestation of a physiological disorder.

    Notes: Abstract The expression of seven genes from the anthocyanin biosynthesis pathway was determined in different tissues of Shiraz grapevines. All of the tissues contained proanthocyanidins, but only the berry skin accumulated anthocyanins. In most tissues, all of the flavonoid genes except UDP glucose-flavonoid 3-o-glucosyl transferase (UFGT) were expressed, but UFGT expression was only detected in berry skin. Similar patterns of expression were observed in the skin of other red grapes. In white grapes, UFGT expression was not detected. White grape cultivars appear to lack anthocyanins because they lack UFGT, although they also had decreased expression of other flavonoid pathway genes.
