    Notes: The ciliated protozoan Paramecium contains hemoglobin in heterogeneous monomeric forms. In particular, Paramecium caudatum is characterized by the presence of a major component called Hb10 and a basic component named bHb. We found that in P. caudatum both of these hemoglobin components show some variation according to stock. The types and distributions of these hemoglobin components were examined on 16 stocks in five different syngens and one stock in an unidentified syngen using high performance liquid chromatography. The results indicate that in a variety of stocks the major component, Hb10, was divided into three types, A, B or A + B, and that the basic hemoglobin component was composed of a combination of two or three variants out of four possible, i.e. bHb 1, bHb 2, bHb 3 and bHb 4. Neither the Hb10 types nor the bHb variants, however, could be used to distinguish syngen in P. caudatum, since all of the Hb10 types and bHb variants were widely distributed over syngens and identical profiles appeared to some stocks in different syngens.

    Notes: Starch gel electrophoresis and isoelectric focusing were performed on sarcoplasmic proteins from three taxa of cobitid fish in Japan, Cobitis biwae, C. taenia striata and C. l. taenia. These taxa are hardly distinguishable from each other by external appearance or morphological characters. Electrophoretic patterns of sarcoplasmic proteins from white muscle suggested fixed allelic differences between C. biwae and C. t. striata at two protein-coding loci (parvalbumin and creatine kinase). The third taxon, C. t. taenia, showed composite banding patterns with respect to the above two proteins, suggesting C. t, taenia originated from hybridization between C. biwae and C, t. striata. Previously, C. t, taenia and C. t. striata were believed to have shared a common ancestor subsequent to their divergence from C. biwae.

    Notes: Abstract Dryobalanops aromatica is an emergent canopy tree species with hermaphroditic bee-pollinated flowers. This species is distributed in the lowland dipterocarp forests of Malaya, Sumatra, Rhio Archipelago and Borneo. The flowering of many different dipterocarp species was observed in some parts of Brunei in 1991, and all over Brunei in 1992. The outcrossing rates of D, aromatica were estimated in 1991 and 1992 in the secondary forest, and in 1992 in the primary forest. Selective harvesting of D. aromatica has occurred in the secondary forest about 20 years ago. Four enzyme systems with a total of six isozyme loci were used to estimate the single locus and the multilocus outcrossing rates. The multilocus outcrossing estimates ranged from 0.794 to 0.856, with a mean of 0.816. These estimates are similar to those for other tropical tree species. Although the flowering density of D. aromatica in the secondary forest was lower than in the primary forest, the multilocus outcrossing estimates were not significantly different. The variation between years in the secondary forest was also not significant. These findings may be largely the result of the high flowering tree density of this species, even in the secondary forest. In this study, the outcrossing rates of D. aromatica were maintained by high flowering density regardless of the size structure or the topography of the habitat.

    Notes: Abstract Mouse monoclonal antibody (mAb) Pgf-II specific for a 72-kDa major cell-surface protein (72K-CSP) derived from Porphyromonas gingivalis OMZ 409 was prepared. Immunoblotting analysis revealed that mAb Pgf-II reacted with 72K-CSP but not with 41-kDa fimbrial subunit protein (41K-fimbrilin) derived from P. gingivalis 381. Electron microscopic observation revealed that P. gingivalis OMZ 409 possessed peritrichous, thin fimbriae on their surface. Immunogold electron microscopy also demonstrated that mAb Pgf-II bound to the 72K-CSP examined with the gold particles arranged along the fibril array originating from the cell surface of the bacteria. These findings suggested that P. gingivalis 72K-CSP was identifiable as another fimbriae (termed Pg-II fimbriae) different from the fimbriae (termed Pg-I fimbriae) composed of a 41K-fimbrilin. Using multipin peptide synthesis technology, 102 sequential overlapping peptides covering the entire 514 amino-acid stretch of Pg-II fimbriae were synthesised. Seven immunodominant regions within Pg-II fimbrial protein molecule, which definitely reacted with the serum of patients with periodontal diseases, were detected.

    Notes: Abstract We previously established murine hybridomas producing a monoclonal antibody monospecific against three glucosyl-transferases (I, SI and S) of Streptococcus mutans which contribute to dental caries formation. Here, we developed a new immunochemical technique (cross-dot system) with which individual levels of glucosyltransferases expressed by S. mutans can be evaluated. We also examined glucosyltransferase production and in vitro artificial plaque formation by a reference strain and several clinical isolates of S. mutans. The findings indicate that the levels of glucosyltransferases produced greatly vary with the cells and the culture medium, and that the cells producing high levels of both glucosyltransferase-SI and glucosyltransferase-I enzymes may possess high in vitro artificial plaque forming ability. We suggest that the cross-dot system will be useful for estimating the cariogenic potential of S. mutans isolates.

    Notes: The effects of stereoregularity on the low-temperature relaxation processes were studied by dynamic mechanical measurements on isotactic and syndiotactic polyisobutyl methacrylates (iso-PiBMA and syn-PiBMA). The α, β, and γ relaxation processes were observed in both stereoregular forms. Both the α, and β loss peaks were at lower temperatures for iso-PiBMA than for syn-PiBMA. The γ loss peak was observed at about -155°C at 30 Hz for both forms, and the apparent activation energy of this process was same for both samples within experimental error (6.7 ± 0.5 kcal/mole). It was reduced from these results that the α and β processes are both considerably influenced by the isotactic configuration but the γ process is not.

    Notes: The sorption properties of atactic polypropylene (APP) and isotactic polypropylene (IPP) were studied by equilibrium sorption of various organic solvents. The variation of the Flory-Huggins interaction parameter χ for the APP-CCl4 system at 25°C was expressed as a function of the volume fraction v2 of polymer by the relation: x = 0.113 exp {1.879 v2}. The average molecular weight Mc of the polymer chains between successive crystallites for IPP subjected to different thermal treatments was calculated to be 250 to 350 by the equation of Flory and Rehner. From the variation of Mc with solvent concentration, we estimated the number fraction of polymer chains actually contributing to elastic deformation. The clustering function for solvent in the polymer calculated by the method of Zimm and Lundberg decreased linearly from a positive value to -1 with increasing solvent concentration. Clustering of solvent molecules was found to occur more easily in APP than in IPP.

    Notes: Potentiometric titration in various 1-1 salt solutions was carried out to study the dissociation behavior of the poly(allylammonium) cation (PAAH+). The titration curves show that proton release from cationic PAAH+ is markedly suppressed by addition of simple salt and depends on the counterion species. From an analysis of the titration curves it is found that the electrostatic free energy change ΔGel upon the proton release is negative in all systems examined, and its absolute value decreases with increasing ionic strength of the medium. Also, at a constant added salt concentration (0.1 mol L-1) ΔGel depends significantly on the counterion species added and increases in the order $\[ {\rm ClO}_{\rm 4}^ - 〉 {\rm NO}_3^ - 〉 {\rm Cl}^ - 〉 {\rm Br}^ - 〉 {\rm SCN}^ - 〉 {\rm I}^ - . \]$ The value of ΔGel correlates well with the electron donor constant En of the counterions. The dissociation behavior of PAAH+ is discussed in terms of electrostatic interaction among the ionized groups on the chain, the charge shielding effect, and the ion-solvent interaction of the added counterions.

    Notes: The hydrophobic association behavior of p-propylbenzenesulfonate ions (n-PBS- and iso-PBS-) around poly(allylammonium) cation (PAAH+ was studied by absorption and 1H nuclear magnetic resonance (NMR) spectroscopy. In the presence of PAAH+Cl-, the broadening (hypochromism) of absorption band of PBS- were observed. In addition, all 1H NMR signals of PBS- exhibited up-field shift which resulted from the intermolecular ring current shift. These results indicate the hydrophobic association of negative PBS- around PAAH+. The hydrophobic association arises from the accumulation of counterions around the polyion, and is stabilized by the hydrophobic interaction between propyl groups and the stacking interaction between benzene rings. The association of iso-PBS- ions is rather weaker than that of n-PBS-, suggesting that the steric hindrance of isopropyl groups prevents an effective association of hydrophobic counterions. Furthermore, through viscosity and Cl- activity measurements, it was found that the binding of associated PBS-,s to PAAH+ causes the change of its surroundings to hydrophobic character and its conformation. © 1994 John Wiley & Sons, Inc.
