    Notes: Cellulase production by strains of Myrothecium verrucaria, Stachybotrys atra and Trichoderma viride was examined. Myrothecium verrucaria was found to give the greatest yields. A variety of media were examined as potential substrates for the industrial production of cellulase. The salts content of the medium was varied and was found to affect cellulase production. Glucose, carboxymethylcellulose (CMC), filter paper and three industrial wastes were examined as possible cellulase inducers. Filter paper was found to be the most effective, followed by sugar cane bagasse and CMC.

    Notes: To study the effects of the growth rate of the hybridoma cell Mn12 on productivity, cell cycle, cell size, and shear sensitivity, six continuous cultures were run at dilution rate of 0.011, 0.021, 0.023, 0.030, 0.042, and 0.058 h-1. This particular hybridoma cell appeared to be unstable in continuous culture with respect to specific productivity, as a sudden drop occurred after about 30 generations in continuous culture, accompanied by the appearance of two populations with respect to the cytoplasmic lgG content. The specific productivity increased with increasing growth rate. The shear sensitivity of the cell, as measured in a small air-lift loop reactor, increased with increasing growth rate. The mean relative cell size, as determined with a flow cytometer, increased with increasing growth rates. Furthermore, the fraction of cells in the S phase increased, and the fraction of cells in the G1/G0 phase decreased with increasing growth rates. © 1993 John Wiley & Sons, Inc.

    Notes: The role of computers in the monitoring and control of fermentation processes has increased steadfastly. The ultimate utility of the machines will not depend on the availability of online sensors but also on the availability of techniques that combine direct measurements, leading towards estimates of variable closely related to the microbial process or its control. In this article, a methodology for on-line and noninterfering evaluation of the volumetric mass-transfer coefficient Kla is developed. A detailed presentation of the procedure, called “the static method,” is given. Its feasibility is proved through implementation of the method on an antibiotic fermentation process. These experiments indicate that operator actions meant to modify the oxygen-transfer conditions can be checked on-line. The quantitative value of the static method is ascertained by comparing the experimental results with Kla estimates obtained with the “gassing-out” method. A sensitivity analysis was carried out, revealing the need for temperature and pressure corrections and showing that the precision of the oxygen analyzer determines the precision of the static method.

    Notes: We studied the effect of organic solvents on the kinetics of porcine pancreatic lipase (pp) for the resolution of racemic glycidol through esterification with butyric acid. We quantified ppl hydration by measuring water sorption isotherms for the enzyme in the solvents/mixtures tested. The determination of initial rates as a function of enzyme hydration revealed that the enzyme exhibits maximum apparent activity in the solvents/mixtures at the same water content (9% to 11% w/w) within the associated experimental error. The maximum initial rates are different in all the media and correlate well with the logarithm of the molar solubility of water in the media, higher initial rates being observed in the solvents/mixtures with lower water solubilities. The data for the mixtures indicate that ppl apparent activity responds to bulk property of the solvent. Measurements of enzyme particle sizes in five of the solvents, as function of enzyme hydration, revealed that mean particle sizes increased with enzyme hydration in all the solvents, differences between solvents being more pronounced at enzyme hydration levels close to 10%. At this hydration level, solvents having a higher water content lead to lower reaction rates; these are the solvents where the mean enzyme particle sizes are greater. Calculation of the observable modulus indicates there are no internal diffusion limitations. The observed correlation between changes in initial rates and changes in external surface area of the enzyme particles suggests that interfacial activation of ppl is only effective at the external surface of the particles. Data obtained for the mixtures indicate that ppl enantioselectivity depends on specific solvent-enzyme interactions. We make reference to ppl hydration and activity in supercritical carbon dioxide. © 1994 John Wiley & Sons, Inc.

    Notes: ATP production, measured by the luciferin-luciferase assay, is an indicator of bacterial metabolic activity. This enzymatic assay yields rapid results (〈 5 minutes), permitting multiple measurements and establishment of ATP growth curves in order to study the kinetics of antibiotics in bacterial populations. The measurement of free or extracellular ATP, total ATP (extra and intracellular) and the ratio of free to total ATP are additional means of studying the bacteriostatic or bactericidal activity of antibiotics. An increase in free ATP is an indicator of extracellular movement due to alteration of the cell wall. The ratio free ATP/total ATP × 100 ≥ 50%, indicates bacteriallysis. These assays were used to study the effects of 14 antibiotics on two reference strains of Escherichia coli ATCC 25922 and Staphylococcus aureus 25923.

    Notes: The chemiluminescence of peripheral blood monocytes and alveolar macrophages was determined in the presence of luminol and lucigenin, either before or after the cell adherence to the luminometer curvettes. In the case of monocytes, cell adherence induces an increase of luminol-dependent chemiluminescence and has almost no effect on the lucigenin-dependent chemiluminescence. However, it shows a strong inhibition of the lucigenin-dependent chemiluminescence and almost no effect on luminol-dependent chemiluminescence, in the case of alveolar macrophages. These results show that adhesion to plastic alters the metabolic burst of both monocytes and alveolar macrophages. Although the mechanisms are poorly understood, they seem to be related to the modifications that take place during the differentiation of peripheral monocytes to alveolar macrophages.

    Notes: A procedure for the preparation of about 50 g batches of tRNA from 25 kg E. coli W is described. The method involves phenolic extraction of the cells, batch absorption of the tRNA on DEAE-cellulose, washing the DEAE-cellulose and packing it into a column, elution of the tRNA from the column and precipitation of the tRNA with ethanol. The method is less time and labor consuming than the methods described in the literature and can be carried out with relatively simple equipment.

    Notes: The relation between the intrinsic rate of the glucose-fructose isomerization catalyzed by whole-cell immobilized glucose isomerase and the substrate concentration can be described with the kinetic model The numerical values of k1, k-1, k2, and k-2 have been determined from low-conversion experiments starting from pure glucose or fructose solutions, and are presented as a function of pH and temperature. The difference between the overall chemical reaction rate determined in high-conversion experiments and that calculated from the individual k1, k-1, k2, and k-2 values is less than 10%.

    Notes: In recent years considerable effort has been made in the Netherlands toward the development of a more sophisticated anaerobic treatment process, suitable for treating low a strength wastes and for applications at liquid detention times of 3-4 hr. The efforts have resulted in new type of upflow anaerobic sludge blanket (UASB) process, which in recent 6 m3 pilot-plant experiments has shown to be capable of handling organic space loads of 15-40 kg chemical oxygen demand (COD)·m-3/day at 3-8 hr liquid detention times. In the first 200 m3 full-scale plant of the UASB concept, organic space loadings of up to 16 kg COD·m-3/day could be treated satisfactorily at a detention times of 4 hr, using sugar beet waste as feed. The main results obtained with the process in the laboratory as well as in 6 m3 pilot plant and 200 m3 full-scale experiments are presented and evaluated in this paper. Special attention is given to the main operating characteristics of the UASB reactor concept. Moreover, some preliminary results are presented of laboratory experiments concerning the use of the USB reactor concept for denitrification as well as for the acid formation step in anaerobic treatment. For both purposes the process looks feasible because very satisfactory results with respect to denitrification and acid formation can be achieved at very high hydraulic loads (12 day-1) and high organic loading rates, i.e., 20 kg COD·m-3/day in the denitrification and 60-80 kg COD·m-3/day in the acid formation experiments.
