    Notes: Hydrogen-bonded complexes of the photoacid 1-naphthol with NH3 and ND3 were investigated by resonant two-photon ionization, spectral hole burning, and fluorescence spectroscopies. Although the intermolecular vibrations are weak in both absorption and emission, with typical Franck–Condon factors 〈2% relative to the electronic origin, all six intermolecular modes were identified, namely the hydrogen bond stretch σ, the ammonia torsion τ, two in-plane wags β1 and β2, and two out-of-plane rocking motions ρ1 and ρ2. Several ammonia torsional excitations were observed, with spacings in good agreement with the S0- and S1 state effective torsional barriers derived by Humphrey and Pratt [J. Chem. Phys. 104, 8332 (1996)]. The β1, β2, and ρ2 vibrational excitations exhibit large (2–8 cm−1) torsional splittings, which indicate strong anharmonic coupling with the ammonia internal rotation. The observed Franck–Condon factors of the intermolecular stretching vibration imply a contraction of the O–H⋅⋅⋅N hydrogen bond by (approximate)0.01 Å upon S1←S0 excitation. © 2001 American Institute of Physics.

    Notes: In Bradyrhizobium japonicum, the nitrogen-fixing symbiont of soybeans, we have identified a haem uptake system, Hmu, that comprises a cluster of nine open reading frames. Predicted products of these genes include: HmuR, a TonB-dependent haem receptor in the outer membrane; HmuT, a periplasmic haem-binding protein; and HmuUV, an ABC transporter in the inner membrane. Furthermore, we identified homologues of ExbBD and TonB, that are required for energy transduction from the inner to the outer membrane. Mutant analysis and complementation tests indicated that HmuR and the ExbBD–TonB system, but not the HmuTUV transporter, are essential for haem uptake or haem acquisition from haemoglobin and leghaemoglobin. The TonB system seems to be specific for haem uptake as it is dispensable for siderophore uptake. Therefore, we propose the existence of a second TonB homologue functioning in the uptake of Fe-chelates. When tested on soybean host plants, hmuT-hmuR and exbD-tonB mutants exhibited wild-type symbiotic properties. Thus, haem uptake is not essential for symbiotic nitrogen fixation but it may enable B. japonicum to have access to alternative iron sources in its non-symbiotic state. Transcript analysis and expression studies with lacZ fusions showed that expression of hmuT and hmuR is induced under low iron supply. The same was observed in fur and irr mutant backgrounds although maximal induction levels were decreased. We conclude either that both regulators, Fur and Irr, independently mediate transcriptional control by iron or that a yet unknown iron regulatory system activates gene expression under iron deprivation. An A/T-rich cis-acting element, located in the promoter region of the divergently transcribed hmuTUV and hmuR genes, is possibly required for this type of iron control.

    Notes: Phosphatidycholine (PC), the major membrane phospholipid in eukaryotes, is found in only some bacteria including members of the family Rhizobiaceae. For this reason, it has long been speculated that rhizobial PC might be required for a successful interaction of rhizobia with their legume host plants in order to allow the formation of nitrogen-fixing root nodules. A major pathway for PC formation in prokaryotes involves a threefold methylation of the precursor phosphatidylethanolamine (PE). Here, we report on the isolation of a Bradyrhizobium japonicum gene (pmtA) encoding the phospholipid N-methyltransferase PmtA. Upon expression of the bradyrhizobial pmtA gene in Escherichia coli, predominantly monomethylphosphatidylethanolamine was formed from PE. PmtA-deficient B. japonicum mutants still produced low levels of PC by a second methylation pathway. The amount of PC formed in such mutants (6% of total phospholipids) was greatly decreased compared with the wild type (52% of total phospholipids). Root nodules of soybean plants infected with B. japonicum pmtA mutants showed a nitrogen fixation activity of only 18% of the wild-type level. The interior colour of the nodules was beige instead of red, suggesting decreased amounts of leghaemoglobin. Moreover, ultrastructure analysis of these nodules demonstrated a greatly reduced number of bacteroids within infected plant cells. These data suggest that the biosynthesis of wild-type amounts of PC are required to allow for an efficient symbiotic interaction of B. japonicum with its soybean host plant.

    Notes: Abstract. The STAT transcription factors form a family of signal transducers and activators of transcription. We sequenced the bovine STAT5B cDNA and both STAT5-encoding genes, STAT5A and STAT5B, representing the first complete description of any STAT5-encoding gene. DNA fiber FISH hybridization revealed that the genes reside only 40 kbp apart on BTA19. Both genes are segmented into 19 exons and all but two of the homologous exons are of equal size. The genes harbor a central block of nearly identical DNA sequence (97.5% sequence identity over 3373 bp), spanning from intron 5 to intron 9. Isolation and sequencing of the homologous segments from mouse revealed the same unusually high degree of intronic sequence conservation in these segments of the murine STAT5-encoding genes. However, the respective sequences are completely divergent between the two species. A comparison of the inter- and intragenic cDNA sequence preservation at nonsynonymous sites reveals that the DNA-binding domain is under the strongest selection pressure for both intergenic and factor-specific intragenic sequence preservation. The so-called ``SH3'' segment of the linker domain, in contrast, shows species-specific sequence identity in all but one amino acid residues in both factors, in cattle, human, and mouse. This indicates that the same species-specific selection pressure occurs on the linker domain from both factors, STAT5A and STAT5B. Thus, the comparison of evolutionary selection pressures resting on various domains suggests that the DNA-binding domain might contribute to differential DNA binding of STAT5A and STAT5B factors, while both might interact equally well with other cellular factors through a segment of the linker domain.

    Notes: Abstract This paper discusses why estimates of the benefits of reduced air pollution differ in accordance with the approach used. Estimates based on bottom-up studies of the damage costs related to air pollution usually turn out much lower than estimates based on assessments of the utility of reduced air pollution, obtained for instance by willingness to pay assessments. This is usually explained by the fact that the willingness to pay approach includes the utility aspect of non-market values, and for this reason, it is often preferred to the damage cost approach. This is, however, not the whole story. The paper shows why alternative approaches should not be considered as being in conflict, but rather as means to get supplementary information necessary to put a value on environmental quality. Information from bottom-up assessments of damage costs and from studies of the willingness to pay is used in a macroeconomic model to carry out an evaluation of the social costs of energy saving measures in Hungary.

    Notes: Abstract We apply a novel laser diagnostic technique — Resonant Holographic Interferometry (RHI) to measure the concentration of hydroxyl radical (∼2000 ppm) in a co-flow diffusion flame of diluted hydrogen and air stabilized on a Wolfhard-Parkerburner. This methodology is based upon the dispersion of light of frequency close to an electronic transition of a target molecule. The two-color setup utilized in RHI provides a two-dimensional distribution of the target species concentration and quantitative information can be obtained from the interferogram without requiring any calibration. To provide independent flame data for comparison, a two-dimensional numerical simulation was performed taking into account the effects of detailed chemical kinetics and transport phenomena. In spite of a number of simplifying assumptions made in the simulation, computational and experimental results are in good agreement with respect to the magnitude and width of the region where OH is found. We do observe a difference of approximately 1 mm in the flame position due to the simplifying assumptions made in the simulation. The comparison between the experimental and numerical results clearly demonstrated the potential of RHI in flame diagnostics.
