    Notes: Abstract. Pex is a newly discovered gene (also called Phex) whose mutation is the cause of X-linked hypophosphatemia. Other members of this gene family encode endopeptidases that activate or inactivate endocrine and paracrine factors. Though embryonic bone expresses mRNA for the Pex gene at relatively high levels, we have found Pex expression to be widespread in adult organs and to be poorly expressed in adult bone. This led to the hypothesis that Pex mRNA expression changes with age. To test this, genetically normal mice of the B6C3H hybrid strain were studied at 0 (newborn), 2, 3, 10, and 72 weeks of age. Organs known to express Pex were collected, and RNA was extracted from them. Following reverse transcription, cDNA was amplified by the polymerase chain reaction with primers for Pex and G3PDH, a housekeeping gene. The amplimers were separated by electrophoresis, blotted onto nylon membranes, and hybridized with radioactively labeled internal oligonucleotide probes. The radioactivity was quantified, and the data were analyzed as the Pex/G3PDH ratio. The brain samples had high levels of Pex mRNA expression that rose slightly with age. Calvaria, kidney, and lung samples had the highest Pex mRNA expression at birth. In these organs Pex mRNA expression fell with age to undetectable or barely detectable levels. Thymus, heart, and skeletal muscle samples had low Pex mRNA expression at birth that did not change with age. Some organs showed a decline in G3PDH levels with age, but Pex expression decreased more, leading to a reduced Pex/G3PDH ratio. The widespread expression of mRNA for Pex suggests a role beyond that of phosphate homeostasis. The high level of expression in newborn animals suggests a role in growth and development. This seems to occur in addition to its role for the endocrine regulation of phosphate homeostasis by as yet unknown humoral agents that must occur throughout life. In summary, Pex mRNA expression is high in brain and bone at birth. Expression remains high in brain with age but falls with age in bone, kidney, and lung.

    Notes: Abstract We demonstrate ways to incorporate nondeterminism in a system designed to formalize the reasoning of agents concerning their abilities and the results of the actions that they may perform. We distinguish between two kinds of nondeterministic choice operators: one that expresses an internal choice, in which the agent decides what action to take, and one that expresses an external choice, which cannot be influenced by the agent. The presence of abilities in our system is the reason why the usual approaches towards nondeterminism cannot be used here. The semantics that we define for nondeterministic actions is based on the idea that composite actions are unravelled in the strings of atomic actions and tests that constitute them. The main notions used in defining this semantics are finite computation sequences and finite computation runs of actions. The results that we obtain meet our intuitions regarding events and abilities in the presence of nondeterminism.

    Notes: Abstract Micromachining technologies were established to fabricate microelectrode arrays and devices for interfacing parts of the central or peripheral nervous system in case of neuronal disorders. The devices were part of a neural prosthesis that allows simultaneous multichannel recording and multisite stimulation of neurons. Overcoming the brittle mechanics of silicon, we established a process technology to fabricate light-weighted and highly flexible polyimide based devices. Concerning the challenging housing demands close to the nerve to prevent mechanical induced nerve traumatization, we integrated interconnects to decouple the nerve interface from plugs and signal processing electronics. Hybrid integration with a new assembling technique—the MicroFlex interconnection (MFI)—has been applied for the connection of the flexible microsystems to silicon microelectronics. In this paper, we present different shapes and applications of the flexible electrodes: sieve electrodes for regeneration studies, cuff electrodes for interfacing peripheral nerves, and a retina implant for ganglion cell stimulation. The discussion is focused on electrode and material properties and the hybrid assembly of a fully implantable neural prosthesis.

    Notes: Abstract Objectives: Thirty years ago, a drug therapy consultation service centre was established simultaneously with the Institute of Clinical Pharmacology based at the university hospital in Magdeburg. The combination of therapeutic drug monitoring and a drug information service including individual patient-oriented drug dosage and bedside consultation permits an effective support of the treatment of drug related therapeutic problems. Currently, the service offers drug monitoring for 161 drugs and additional 46 metabolites. Methods and discussion: In a representative 2-year period (1997–1998), drug monitoring was performed for 6293 patients, and 569 drug-related inquiries were answered. The main categories of required drug information were: pharmacokinetics/metabolism/analytical problems (38%), adverse drug reactions/drug safety (24%), therapeutic drug use/drug indication (21%) and pharmaceuticals (9%). Further activities of the institute refer to teaching of undergraduates, continuing education of local physicians in recent aspects of drug therapy, providing therapeutic bulletins and conducting supporting clinical studies.

    Notes: Abstract Co-artemether is an oral tablet of artemether (20 mg) and lumefantrine (120 mg) for the treatment of falciparum malaria. Administration in the presence of mefloquine is likely, as co-artemether may be used following failure of antimalarial prophylaxis or treatment with mefloquine. Objective: The effects on the QTc interval were compared among treatment with three doses of mefloquine (500, 250, 250 mg over 12 h) followed by six doses of co-artemether (6 × 4 tablets over 60 h) and either treatment alone. The study was performed in a randomised, double-blind, parallel group design in 14 healthy male subjects per dose group. Methods: Electrocardiograms (ECGs) were recorded before dosing and repeatedly thereafter. The Bazett formula was used to calculate the QTc interval. The maximum and average QTc intervals for the first, third and sixth dosing intervals of co-artemether treatment were compared among treatments. Drug plasma concentrations were determined at identical times with the ECG recordings for exploratory pharmacokinetic/pharmacodynamic evaluation. Results: No clinically relevant differences in the QTc interval were observed after sequential administration of mefloquine and co-artemether relative to either treatment given alone, and there were no clinically relevant study drug-related effects on the QTc interval after either treatment. Plasma drug measurements revealed adequate systemic exposure to artemether, dihydroartemisinin, lumefantrine and mefloquine, well in line with the clinical setting. No correlation between the length of the QTc interval and plasma drug concentrations was found for any of the compounds. Conclusions: Untoward effects on the QTc interval are unlikely to occur when co-artemether is administered following prophylaxis or treatment with mefloquine.

    Notes: Abstract The study was designed to demonstrate - for the first time in humans - that oxidative stress in the heart indicated by lipid peroxidation is associated with time--dependent changes in the enzymatic antioxidative defense. For this purpose, we analyzed the oxygen radical metabolism in 69 myocardial biopsies (taken between the fifth day and 6 years after transplantation) of 31 heart transplant recipients who were suspected of suffering from increased formation of oxygen radicals in the allograft. The levels of lipid peroxides (LPO), glutathione peroxidase (GSH-Px), total-, copper/zinc- and manganese superoxide dismutase (t-SOD, CuZnSOD, MnSOD) were compared in 3 post-transplantation periods (5-90 d vs. 91-365 d vs. 1 y). Significantly increased LPO levels were found (0.27±0.04 vs. 0.13±0.02 vs. 0.27±0.04 nmol/mg protein) in the first and third period. Increased activities of GSH-Px (39.8±3.8 vs. 30.2±4.1 vs. 76.±6.5 mU/mg protein), t-SOD (1.57±0.10 vs. 1.30±0.14 vs. 2.44±0.23 U/mg protein) and CuZnSOD (1.09±0.08 vs. 0.93±0.13 vs. 2.05±0.21 U/mg protein) occurred only in the third period. For calculation of time courses more precisely, the single data with respect to time were analyzed with a curve fitting program. Except for the first period, the allograft LPO and GSH-Px levels rose for up to 6 years after transplantation. However, the t-SOD and CuZnSOD activities switched from increase to decrease in the third period. The study provided indication for: first, the potency of the human heart to time-limited increase of the enzymatic antioxidative defense, and secondly, the inability of human heart allografts - despite this adaptation - for complete prevention of myocardial oxidative stress.

    Notes: Abstract. The development of dentin and of enamel share a common starting locus: the dentinoenamel junction (DEJ). In this study the relationship between enamel and dentin crystals has been investigated in order to highlight the guiding or modulating role of the previously mineralized dentin layer during enamel formation. Observations were made with a high-resolution electron microscope and, after digitalization, image-analysis software was used to obtain digital diffractograms of individual crystals. In general no direct epitaxial growth of enamel crystals onto dentin crystals could be demonstrated. The absence of direct contact between the two kinds of crystals and the presence of amorphous areas within enamel particles at the junction with dentin crystals were always noted. Only in a few cases was the relationship between enamel and dentin crystals observed, which suggested a preorganization of the enamel matrix influenced by the dentin surface structure. This could be explained either by the existence of a proteinaceous continuum between enamel and dentin or by the orientation of enamel proteins by dentin crystals.

    Notes: Abstract. A sexually dimorphic distribution of galanin in the preoptic region of the molly and goldfish has previously been demonstrated. Females of these species lack galanin-immunoreactive perikarya in the preoptic nucleus. In contrast, we have found, in female red salmon, galanin-immunoreactive neurons in the magnocellular preoptic nucleus, located far lateral to the preoptic recess, whereas many immunoreactive fibers are present in the preoptic area in both genders. In addition, many immunoreactive neurons have been seen in the nucleus preopticus periventricularis and nucleus lateralis tuberis, also in both sexes. These findings support the notion that galanin may play a gender-specific role in red salmon.

    Notes: Abstract. Gastrin stimulates gastric acid secretion by acting on the cholecystokinin B/gastrin receptor (CCK-BR). The localization of this receptor at the cellular level showed conflicting results in animal studies and has not been described in man by immunohistochemistry. The aim of the present study is to characterize the precise cellular location of the CCK-BR in the human stomach. Polyclonal antisera were raised against different epitopes of the CCK-BR molecule and used for immunohistochemical investigations. CCK-BR mRNA was detected in paraffin tissue sections by the highly sensitive method of in situ reverse transcriptase-polymerase chain reaction (RT-PCR). Using immunohistochemistry, CCK-BR could successfully be localized in gastric parietal cells. In the majority of parietal cells, CCK-BR immunoreactivity was present at the basolateral cell membrane domain. In some parietal cells, a granular pattern of immunoreactivity was exclusively confined to the cytoplasm of the cells. CCK-BR mRNA was found in parietal cells and in enterochromaffin-like (ECL) cells by means of in situ RT-PCR. No expression of CCK-BR was found in the gastric antral mucosa. Our data support the concept that gastrin stimulates gastric acid secretion directly via CCK-B receptors on parietal cells and indirectly by inducing histamine release from histamine-containing ECL cells, which contributes to acid secretion by parietal cells.
