Notes: Pk-REC is a protein which binds to DNA and catalyzes the central step of recombination and repair. The protein was crystallized using the hanging-drop vapour-diffusion method with PEG as a precipitant. Two orthorhombic crystal forms I and II with the same space group P212121 were obtained at pH 8.0 using PEG 3000 and PEG 550 monomethylether, respectively. The unit-cell parameters were a = 151, b = 174, c = 241 Å for form I and a = 151, b = 176, c = 300 Å for form II, indicating that the asymmetric unit contains more than 20 molecules.

Notes: Crystals of the complex between granulocyte colony-stimulating factor and its soluble receptor were obtained by a vapour-diffusion method using ammonium sulfate as a precipitant. Addition of 1,4-dioxane was critical in order to grow the crystals to sufficient sizes. Cryoprotection was essential in order to collect diffraction data at atomic resolution. Two kinds of crystal forms were obtained depending on the cryoprotectants. In a cryosolvent with the same salt concentration as in the crystallization conditions, the crystal belonged to the space group I4122. At higher salt concentrations, the crystal was converted to a different space group P41212 (P43212) with the same unit-cell parameters.

Notes: Abstract: We studied the changes in the electronic structure of SrTi1-xRuxO3 across the metal-insulator transition. The parent compound, SrTiO3, is a well known diamagnetic insulator; whereas the doped compound, SrTi1-xRuxO3, becomes a ferromagnetic metal above x C = 0.35. The techniques used in the study were photoemission (PES) and O 1 s X-ray absorption (XAS) spectroscopy. The experimental spectra were analyzed in terms of band structure and Hubbard model calculations. The PES and XAS spectra of SrTi1-xRuxO3 show the Ru 4 d bands growing in the band gap of SrTiO3 . The analysis in terms of the Hubbard model indicates that the Ti 3 d and Ru 4 d bands are mostly decoupled. This suggests that the metal-insulator transition is a percolation transition like that of metals embedded in a rare gas matrix. Electron correlation effects are present in this system, but they do not seem to play a major role in the transition.

Notes: Abstract. A Coulomb excitation experiment was carried out with a 66Zn beam bombarding a ${}^{\rm nat}$ Pb target. Four E2 matrix elements and the quadrupole moment of the 21 + state were derived with the least-squares search code GOSIA. According to the B(E2) values, the ground band can be interpreted as a quasirotational band. It was found that the 21 + level has a positive quadrupole moment, which may be interpreted as a soft triaxial deformation.

Notes: Abstract The characteristics of regulation of the gene encoding the third xylanase (Xyn III) of a filamentous fungus, Trichoderma reesei PC-3–7, were studied by Northern blot analysis. A partial DNA sequence (185 bp) of the xyn3 gene was obtained by PCR amplification of genomic DNA of T. reesei PC-3–7 and sequenced. This xyn3 gene fragment was used as a probe for Southern and Northern blot analysis. The expression of the xyn3 gene was regulated at the transcriptional level. The xyn3 mRNA was expressed in mycelia of T. reesei PC-3–7 induced by Avicel, l-sorbose and sophorose, but not by xylose, xylooligosaccharides and birchwood xylan. Furthermore, it was observed that xyn3 was synchronously expressed with egl1 but not with xyn1 and xyn2 by l-sorbose, indicating that the xyn3 gene may be coordinately expressed with cellulase genes. By Southern blot analysis, the xyn3 gene was confirmed to exist as a single copy in both strains of T. reesei PC-3–7 and QM9414. However, no xyn3 mRNA appeared in the mycelia induced by any kind of inducers in T. reesei QM9414 even when total RNA was used in large excess, suggesting that the xyn3 gene in T. reesei QM9414 is in the dormant state and cannot be expressed. Therefore, T. reesei PC-3–7 may be a very useful strain for elucidating the induction mechanism of xylanase biosynthesis by cellulosic and xylanosic substrates, and also the regulatory correlation between cellulase and xylanase induction.

Notes: We observed the x-ray-excited optical luminescence (XEOL) of erbium-doped silicon (Si:Er) thin films to make a site-selective x-ray absorption fine structure (XAFS) measurement of an optically active Er atom. The undulator beam was used for the increment of the electron population in the excited state, and following XEOL at an infrared wavelength of 1.54 µm with minimum absorption loss in the host Si was detected. The edge-jump and XAFS oscillation were successfully obtained at the Er LIII-edge. This spectrum originated from inner-shell excitation and relaxation of only the optically active Er atom, indicating that site-selectivity at an atomic level was achieved.

Notes: The praseodymium-exchanged mordenite degassed at 873 K acted as a highly active heterogeneous photocatalyst for the stoichiometric decomposition of N2O. Analysis of Pr L3-edge EXAFS revealed that the Pr species were isolated each other in the large channel of zeolite, and that the active Pr species were coordinated by ca. four oxygen atoms and one Al atom. On the other hands, the inactive Pr species are surrounded by ca. 10 oxygens of adsorbed waters. The former low-coordinated Pr species allowed a strong adsorption of N2O, which could be the first step of the decomposition reaction.

Notes: Abstract The nematode-trapping fungusArthrobotrys ellipsospora developed an adhesive knob and trapped nematodes when cultured on a low-nutrient medium. It also trapped polystyrene beads in the same way. The adhesive knob produced mucus that was stained with alcian blue, while mycelium of the fungus was stained with periodic acid/Schiff (PAS). The amount of mucus increased with in days after culturing in the low-nutrient media. The fungus completely lost its ability to trap nematodes when treated with EDTA and EGTA, but it recovered the ability after incubation in the presence of a low concentration of Ca (10−6–10−7 M) for 1 h. Calmodulin inhibitor W-7 also inhibited the trapping ability of the fungus, and there was a significant (p〈0.05) difference between the effects of W-7 and W-5. Ca-binding protein was also detected in the fungus.

Notes: Abstract The Kii Bifurcation Current is often found along the southwest coast of the Kii Peninsula, and its frequency of occurrence reaches about 70% in the period from 1988 to 1996 (Takeuchi et al., 1998a). In order to clarify the structure and short-period variability of the Kii Bifurcation Current, detailed observations were made four times on board the R/V Seisui-maru of Mie University on October 29–31, 1996, on June 24–26, 1997, October 14–16, 1997, and December 3–4, 1997. The measured horizontal structure of the Kii Bifurcation Current indicates that the eastern portion of the Current (eastward flow near Cape Shionomisaki) consists of a part of the current zone of the Kuroshio. It is shown that the current structure, including the Kii Bifurcation Current in the vicinity of Cape Shionomisaki, is stable when the Kuroshio is flowing in a stationary straight path, but that the current structure is considerably changed when small-scale eddies pass by the cape. Such short-period variation can be monitored by using the daily variation of the sea level difference between Kushimoto and Uragami. In particular, in the case of October 29–31, 1996, when an eminent small-scale eddy passed by Cape Shionomisaki, and when the Kuroshio axis tentatively moved southwards about 50 km apart from the coast, the Kii Bifurcation Current seems to have disappeared.

Notes: Abstract: High-spin states of 155Gd were populated by using the 154Sm(α,3nγ)155Gd reaction at E α= 33 MeV. γ-γ coincidence, E γ singles, excitation function, and the DCO ratios were measured. we have identified three intermediate bands with ΔI= 2 feeding the positive yrast band. The bands are interpreted as such candidate bands that are mixed with the negative-parity ground state band. This observation can provide a plausible explanation for unusually large population of the positive-parity yrast band observed in a recent Coulomb excitation.
