    Notes: Several R2 somaclonal families were derived from plants regenerated from a salt-resistant callus of the salt-sensitive rice cultivar ‘I Kong Pao’ (IKP). The family R2-1-23, in the presence of NaCl exhibited higher yield performances than the initial cultivar. This improvement in salinity resistance, however, was not transmitted to following generations; despite a higher number of spikelets per plant, family R3-1-23 did not perform better than the initial cultivar because of a very low seed set. This somaclonal family, its initial being the cultivar IKP, the breeding line IR31785 (extremely salt-sensitive) and the cultivar ‘Aiwu’ (moderately salt-resistant), were used as parents for production ofhybrids. Four crosses, IKP×R3-1-23, IR31785 ×R3-1-23, IR31785× IKP and IKP בAiwu’, were performed. Most of the F1 hybrids cultivated in the absence of salt exhibited increased performances compared with the mid-parent, suggesting an heterosis effect for yield-related parameters. F2 populations were screened for salinity resistance and a clear improvement for yield in stress conditions was recorded for populations derived from IK×R3-1-23, IR31785×R3-1-23 and IR31785×IKP, although the mean level of increase over the mid-parent (RIMP) varied depending on the population, the presence or absence of stress, and the quantified parameters. The results are discussed in relation to the usefulness of in vitro selection for obtaining interesting somaclonal variants useful to be integrated in classical breeding programmes for salinity resistance in rice.

    Notes: We describe a rapid method for determining nucleotide sequences directly from total genomic DNA. This technique was used to determine genomic DNA sequences in various prokaryotic and eukaryotic microorganisms with a G+C content between 40 and 50%, e.g. Escherichia coli, Vibrio cholerae, Bacillus subtilis and Saccharomyces cerevisiae. Furthermore, the method was applied to accurately sequence up to 300 DNA base pairs in Photorhabdus luminescens, whose genome sequencing is currently under way. Taken together, these results provide evidence that our technique can be widely used to easily and efficiently determine genomic DNA sequences.

    Notes: Flagellar motility helps bacteria to reach the most favourable environments and to successfully compete with other micro-organisms. These complex organelles also play an important role in adhesion to substrates, biofilm formation and virulence process. In addition, because their synthesis and functioning are very expensive for the cell (about 2% of biosynthetic energy expenditure in Escherichia coli) and may induce a strong immune response in the host organism, the expression of flagellar genes is highly regulated by environmental conditions. In the past few years, many data have been published about the regulation of motility in polarly and laterally flagellated bacteria. However, the mechanism of motility control by environmental factors and by some regulatory proteins remains largely unknown. In this respect, recent experimental data suggest that the master regulatory protein-encoding genes at the first level of the cascade are the main target for many environmental factors. This mechanism might require DNA topology alterations of their regulatory regions. Finally, despite some differences the polar and lateral flagellar cascades share many functional similarities, including a similar hierarchical organisation of flagellar systems. The remarkable parallelism in the functional organisation of flagellar systems suggests an evolutionary conservation of regulatory mechanisms in Gram-negative bacteria.

    Notes: Ralstonia metallidurans, formerly known as Alcaligenes eutrophus and thereafter as Ralstonia eutropha, is a β-Proteobacterium colonizing industrial sediments, soils or wastes with a high content of heavy metals. The type strain CH34 carries two large plasmids (pMOL28 and pMOL30) bearing a variety of genes for metal resistance. A chronological overview describes the progress made in the knowledge of the plasmid-borne metal resistance mechanisms, the genetics of R. metallidurans CH34 and its taxonomy, and the applications of this strain in the fields of environmental remediation and microbial ecology. Recently, the sequence draft of the genome of R. metallidurans has become available. This allowed a comparison of these preliminary data with the published genome data of the plant pathogen Ralstonia solanacearum, which harbors a megaplasmid (of 2.1 Mb) carrying some metal resistance genes that are similar to those found in R. metallidurans CH34. In addition, a first inventory of metal resistance genes and operons across these two organisms could be made. This inventory, which partly relied on the use of proteomic approaches, revealed the presence of numerous loci not only on the large plasmids pMOL28 and pMOL30 but also on the chromosome. It suggests that metal-resistant Ralstonia, through evolution, are particularly well adapted to the harsh environments typically created by extreme anthropogenic situations or biotopes.

    Notes: Anaerobic digestion of olive mill wastewaters is generally performed in anaerobic contact bioreactors where the removal of toxic phenols is often unsatisfactory. In the present work we show that a granular activated carbon packed-bed biofilm reactor can be successfully used to achieve effective and reproducible wastewater decontamination even at high organic loads. A comparison of 16S rRNA gene sequences of the inoculum and of biomass samples from different districts of the reactor revealed enrichment of specific microbial populations, probably minor members of the inoculum and/or of the olive mill wastewaters. They mainly consisted of the members of Proteobacteria, Flexibacter-Cytophaga-Bacteroides, and sulphate-reducing bacteria. The dominant sequence among Archaea (70% of clones) was closely related to Methanobacterium formicicum.

    Notes: Abstract. We study potential signals of neutralino dark matter indirect detection by neutrino telescopes in a wide range of CMSSM parameters. We also compare with direct detection potential signals taking into account in both cases present and future experiment sensitivities. Only models with neutralino annihilation into gauge bosons can satisfy cosmological constraints and current neutrino indirect detection sensitivities. For both direct and indirect detection, only next generation experiments will be able to really test this kind of models.

    Notes: Abstract. We experimentally study the relaxation towards mechanical equilibrium following the surface avalanche of a granular pile and discuss it in the general context of glassy materials and jamming transition. Two coexisting dynamics are observed in the surface layer: a short time exponential decay related to rapid isolated grains displacements, and intermittent reactivation bursts consisting in spatially correlated moves. The competition of both dynamics results in long-lived intermittent transients, the total duration of which can last more than a thousand of seconds. The typical time-scales, and the two-time relaxation function of these transients are measured. Analyzing the response of the surface layer to localized perturbations provides new evidence for spatial correlations. We further analyse the relaxation function in a probabilistic framework, which allows to discuss different possible scalings according to the choice of specific hypothesis. Following some considerations on the observed time-scales, we finally discuss the similarities with aging systems which are suggested by our description of the observed dynamics.

    Notes: Abstract: Using 155Gd Mössbauer spectroscopy down to 27 mK, we show that, in the geometrically frustrated pyrochlore Gd2Sn2O7, the Gd3+ hyperfine levels are populated out of equilibrium. From this, we deduce that the hyperfine field, and the correlated Gd3+ moments which produce this field, continue to fluctuate as T ↦ 0. With a model of a spin 1/2 system experiencing a magnetic field which reverses randomly in time, we obtain an analytical expression for the steady state probability distribution of the level populations. This distribution is a simple function of the ratio of the nuclear spin relaxation time to the average electronic spin-flip time. In Gd2Sn2O7, we find the two time scales are of the same order of magnitude. We discuss the mechanism giving rise to the nuclear spin relaxation and the influence of the electronic spin fluctuations on the hyperfine specific heat. The corresponding low temperature measurements in Gd2Ti2O7 are presented and discussed.

    Notes: Abstract. Type 1 and P-pili are prototype members of Class I fimbriae produced by Gram-negative bacteria. Despite common structural characteristics, the low level of amino acid sequence conservation among the Class I major fimbrial subunits (pilins) indicates considerable evolutionary distance between members of this superfamily. We highlight here structural relatedness between Class I pilins from their two-dimensional sequence analysis using hydrophobic cluster analysis (HCA) and secondary structure predictions (PHD program). We present evidence that all members of the Class I pilin family have clear structural relatedness and suggest that classification based on phylogenetic analysis of Class I pilins into seven subfamilies correlates with differences in structural properties of the amino acid sequences. Using a sensitive alignment process (HCA), we identified 29 residues in topohydrophobic positions which probably play a prominent role in folding. The most striking aspects that distinguish the different pilin subfamilies are (i) large variation in the length of the loops connecting the structurally conserved regions and (ii) intrasubfamily sequence signature motifs located on regions predicted to be in the β-conformation. We suggest that these ``intrasubfamily sequence signature motifs'' are part of interactive surfaces which participate in subunit–subunit interactions. These motifs prove highly useful in characterizing and classifying new Class I fimbriae that have not yet been described and whose sequence diverges appreciably from those of characterized groups. (After the submission of our manuscript, the experimental structure of Class I pilus subunits was published. In light of these actual pilin structures, a comparison has been made between the predicted results and the crystal structure in the Note Added in Proof.)

    Notes: Abstract. In this paper we study formal moduli for wildly ramified Galois covering. We prove a local-global principle. We then focus on the infinitesimal deformations of the Z/p Z-covers. We explicitly compute a deformation of an automorphism of order p which implies a universal obstruction for p〉2. By deforming Artin-Schreier equations we obtain a lower bound on the dimension of the local versal deformation ring. At last, by comparing the global versal deformation ring to the complete local ring in a point of a moduli space, we determine the dimensions of the global and local versal deformation rings.
