    Notes: Consumption of ground beef has been implicated in the majority of outbreaks of disease from Escherichia coli O157:H7. There is a great need for rapid, sensitive, and specific microbial detection and isolation methods for the detection and recovery of pathogens such as E. coli O157:H7. Evanescent wave, fiber optic biosensors are an innovative, cutting-edge technology with the potential to meet such a need. Food pathogens such as E. coli O157:H7 can be detected in minutes using the biosensor assay rather than days using conventional methods. The biosensor assay is sensitive and presently has the ability to directly detect 102 CFU of E. coli O157:H7 in E. coli O157:H7-seeded PBS or ground beef. In this study, it was determined that once the pathogen had been positively identified, it could then be recovered by culturing the cells captured on the waveguide used in the biosensor assay. E. coli O157:H7-seeded PBS or ground beef samples were assayed using an evanescent wave, fiber optic biosensor. After a positive identification on the biosensor, the waveguide was added to modified LB medium supplemented with 10 μg/mL acriflavin (mLB) and incubated at 42C for a minimum of 4 h followed by selection on Rainbow O157 agar incubated 16 to 18 h at 37C. E. coli O157:H7 was confirmed using agglutination with E. coli O157:H7 antiserum. It was possible to recover E. coli O157:H7 colonies in E. coli O157:H7-seeded PBS or ground beef when 1 mL samples containing at least 2.0 × 103 CFU of E. coli O157:H7 had been injected into the biosensor. It took only 24 h to detect the target pathogen, recover the pathogen from the sample, and grow isolated colonies. This total time included sample preparation, detection, enrichment culture, and growth of cells to produce isolated colonies on a selective and differential medium. The identification of isolated colonies was then quickly confirmed using slide agglutination with the appropriate antiserum. This method significantly improved the speed of the confirmation of the presence of the pathogen and of the isolation of the pathogen, from 4 days using conventional methods to one day using the biosensor assay.

    Notes: While many aspects of the collaborative process have been discussed in the management literature, the connection between collaboration and the dynamics of institutional fields has remained largely unconsidered. Yet, collaboration is an important arena for inter–organizational interaction and, therefore, a potentially important context for the process of structuration upon which institutional fields depend. In this paper, we argue that institutionalization and collaboration are interdependent; institutional fields provide the rules and resources upon which collaboration is constructed, while collaboration provides a context for the ongoing processes of structuration that sustain the institutional fields of the participants.

    Notes: Inter-organizational collaboration has been linked to a range of important outcomes for collaborating organizations. The strategy literature emphasizes the way in which collaboration between organizations results in the sharing of critical resources and facilitates knowledge transfer. The learning literature argues that collaboration not only transfers existing knowledge among organizations, but also facilitates the creation of new knowledge and produce synergistic solutions. Finally, research on networks and interorganizational politics suggests that collaboration can help organizations achieve a more central and influential position in relation to other organizations. While these effects have been identified and discussed at some length, little attention has been paid to the relationship between them and the nature of the collaborations that produce them. In this paper, we present the results of a qualitative study that examines the relationship between the effects of interorganizational collaboration and the nature of the collaborations that produce them. Based on our study of the collaborative activities of a small, nongovernmental organization (NGO) in Palestine over a four-year period, we argue that two dimensions of collaboration – embeddedness and involvement – determine the potential of a collaboration to produce one or more of these effects.

There are hundreds of monogenic disorders for which hearing loss is one manifestation of a syndrome or the only disorder and therefore is nonsyndromic. Herein we review the supporting evidence for identifying over 30 genes for dominantly and recessively inherited, nonsyndromic, sensorineural deafness. The state of knowledge concerning their biological roles is discussed in the context of the controversies within an evolving understanding of the intricate molecular machinery of the inner ear.

    Notes: It is widely believed that the initial degradation of proteins contained in grazed forage is mediated by rumen micro-organisms, but the authors’ recent work suggests that the plant cells themselves contribute to their own demise. In the present study the responses of Lolium perenne leaves to the rumen environment were investigated by using an in vitro system which simulates the main stresses of the rumen but from which rumen micro-organisms were excluded. Degradation of leaf protein and the accumulation of amino acids in tissue and bathing medium occurred over a time-scale that is relevant to rumen function, and in a near 1 : 1 ratio. Significant loss of nuclear material was observed after 6 h incubation and chloroplasts became morphologically more spherical as the incubation progressed. In situ localization suggested that ribulose 1,5 bisphosphate carboxylase/oxygenase was broken down within chloroplasts which from cytology were judged to be intact. We conclude from these data that plant metabolism may play a significant role in breaking down plant proteins within relatively intact organelles in the rumen. The determinations of chlorophyll content and cell viability revealed that the plant processes occurring in the simulated rumen were similar but not identical to those of natural senescence.

    Notes: Root elongation in wheat seedlings (Triticum aestivum L. cv. Atlas 66) was inhibited by micromolar activities of SeO42–. SeO42– inhibition was enhanced by supplementation of the rooting medium with CaCl2, MgCl2, SrCl2, or the reduction of pH. These solute treatments, as well as the addition of tris (ethylenediamine)cobalt3+, enhanced the uptake of Se by the roots. The results are interpreted to reflect an elevated PM-surface activity of SeO42– caused by solute-induced reductions of plasma membrane (PM) surface negativity. (PM-surface electrical potential is sometimes measured electrophoretically as the zeta potential.) This study complements an extensive literature documenting the suitability of an electrostatic model (Gouy-Chapman-Stern), based almost entirely upon experiments with cations rather than anions. The close correspondence among uptake, intoxication, and model-computed SeO42– activity at the PM surface adds credibility to the model and its evaluated parameters. The model may be useful for the interpretation of other plant-anion interactions, and phosphate and sulphate nutrition in acidic soils are considered as examples.
