    Notes: The type III secretion system (TTSS) is a modular apparatus assembled by many pathogenic Gram-negative bacteria and is designed to translocate proteins through the bacterial cell wall into the eukaryotic host cell. The conserved components of the TTSS comprise stacks of rings spanning the inner and outer bacterial membrane and a narrow, needle-like structure projecting outwards. The TTSS of enteropathogenic E. coli is unique in that one of the translocator proteins, EspA, polymerizes to form an extension to the needle complex which interacts with the host cell. In this study we present the 3D structure of EspA filaments to c. 26 Å resolution determined from electron micrographs of negatively stained preparations by image processing. The structure comprises a helical tube with a diameter of 120 Å enclosing a central channel of 25 Å diameter through which effector proteins may be transported. The subunit arrangement corresponds to a one-start helix with 28 subunits present in five turns of the helix and an axial rise of 4.6 Å per subunit. This is the first report of a 3D structure of a filamentous extension to the TTSS.

    Notes: Map is an enteropathogenic Escherichia coli (EPEC) protein that is translocated into eukaryotic cells by a type III secretion system. Although not required for the induction of attaching and effacing (A/E) lesion formation characteristic of EPEC infection, translocated Map is suggested to disrupt mitochondrial membrane potential, which may impact upon subsequent functions of the organelle such as control of cell death. Before secretion, many effector proteins are maintained in the bacterial cytosol by association with a specific chaperone. In EPEC, chaperones have been identified for the effector proteins translocated intimin receptor (Tir) and EspF, and for the translocator proteins EspB and EspD. In this study, we present evidence that the Tir-specific chaperone, CesT, also performs a chaperone function for Map. Using a combination of biochemical approaches, we demonstrate specific interaction between CesT and Map. Similar to other chaperone–effector pairings, binding is apparent at the amino-terminus of Map and is indicated to proceed by a similar mechanism to CesT:Tir interaction. Map secretion from a cesT mutant strain (SE884) is shown to be reduced and, importantly, its translocation from this strain after infection of HEp-2 cells is almost totally abrogated. Although other chaperones are reported to have a bivalent binding specificity, CesT is the first member of its family that chaperones more than one protein for translocation.

In this investigation, we compare the multiplication rates and morphogenetic responses of the two most studied Tetrahymena species, T. pyriformis and T. thermophila, at supraoptimal temperatures. Although the upper temperature limits differ greatly in the two species, the pattern of growth responses to high temperature is for the most part similar, with some differences in detail. The transient recovery of cell division at the highest temperature that allows cell division, characteristic of T. pyriformis, is observed in a less distinct form in T. thermophila. Moreover, there is a remarkable difference in developmental response, with drastic abnormalities in patterning of oral structures during the transient recovery of cell division in T. pyriformis, and far more limited abnormalities under similar conditions in T. thermophila. The abnormalities result from spatial disorder in the alignment and orientation of basal body pairs within the early oral primordium, followed by failures in the realignment that normally occurs as oral structures (membranelles and undulating membrane) mature. Both the initial spatial disorder and the failures in realignment are far more severe in T. pyriformis than in T. thermophila.

This study asks two questions: 1) whether Hsp90 is involved in the regulation of cortical patterning in Tetrahymena, and 2) if it is, whether specific defects in this regulation can be attributed to functional insufficiency of the Hsp90 molecule. To address question I, we compared the effects of a specific inhibitor of Hsp90, geldanamycin, on population growth and on development of the oral apparatus in two Tetrahymena species, T. pyriformis and T. thermophila. We observed that geldanamycin inhibits population growth in both species at very low concentrations, and that it has far more severe effects on oral patterning in T. pyriformis than in T. thermophila. These effects are parallel to those of high temperature in the same two species, and provide a tentative affirmative answer to the first question. To address question 2, we ascertained the base sequence of the genes that encode the Hsp90 molecules which are induced at high temperatures in both Tetrahymena species, as well as corresponding sequences in Paramecium tetraurelia. Extensive comparative analyses of the deduced amino acid sequences of the Hsp90 molecules of the two Tetrahymena species indicate that on the basis of what we currently know about Hsp90 both proteins are equally likely to be functional. Phylogenetic analyses of Hsp90 amino acid sequences indicate that the two Tetrahymena Hsp90 molecules have undergone a similar number of amino acid substitutions from their most recent common ancestor, with none of these corresponding to any known functionally critical region of the molecule. Thus there is no evidence that the Hsp90 molecule of T. pyriformis is functionally impaired; the flaw in the control of cortical patterning is more likely to be caused by defects in mechanism(s) that mediate the response to Hsp90, as would be expected from the “Hsp90 capacitor” model of Rutherford and Lindquist.

    Notes: SYNOPSIS. The protargol technic was used in a study of the development of oral, cirral, and dorsal primordia of Urostyla weissei fixed during division, reorganization, and regeneration following transection at different levels. While the course of development is similar in all situations, differences were observed in the way in which some primordia are initiaily formed. The primordium of the new AZM always appears posterior to the old AZM. It develops into an entire new membranellar band in dividing cells and in opimers (posterior fragments from equatorial transections), while it eventually joins with a portion of the old AZM in reorganizers, promers (anterior fragments from equatorial transections) and “large opimers” (cells whose anterior tip has been cut off). The UM-primordium of proters is derived from disaggregation of the kinetosomes of the 2 old UM's, that of opisthes and opimers is formed “de novo” to the right of the AZM-primordium, while the UM-primordium of reorganizers, promers, and “large opimers” is of composite origin, partly “de novo” and partly from the old UM's. The UM primordium differentiates into the new UM's and the 1st frontal cirrus. The primordia of the remaining frontal, ventral, transversal (F-V-T) and marginal cirri originate as “streaks” of cilia, most of which are derived from re-alignment of the constituent cilia of certain pre-existing cirri. New cirri differendiate from the streaks, and replace the remaining old cirri. The streaks are formed similarly in all developmental situations, except for the 1st 3 F-V-T streaks. In proters, reorganizers, and promers, these originate from the posterior 3 frontal cirri, while in opisthes and opimers they are formed “de novo” to the right of the UM-primordium. In the “large opimers” these streaks are formed “de novo” behind the 1st 3 frontal cirri, in spite of the continued presence of these cirri at the anterior tip of the fragments. The site of formation of these streaks thus appears to be determined by an anteriorposterior gradient, rather than by any preformed cortical structure.The new dorsal bristle rows I to III develop from the proliferation of portions of the old rows, while rows IV and V originate from short kineties formed “de novo” on the right margin. New caudal cirri differentiate at the posterior ends of the new rows I to III.The numbers of ventral cirral rows and transversal cirri are variable; these variations are correlated, and related to variations in numbers of developing streaks.A survey of hypotrich developmental patterns revealed extensive parallels, especially in the sites of appearance of primordia. The primordium site appears to be a more constant feature of cortical development than is the “source” of ciliary units. It is concluded that sites of primordia are determined by cellular gradients, with competent preformed structures being utilized if they are appropriately positioned within these gradients.

    Notes: . Homopolar doublets with twofold rotational symmetry were generated in Paramecium tetraurelia and in P. undecaurelia by electrofusion or by arrested conjugation. These doublets underwent a complex cortical reorganization over time, which led to their reversion to singlets. This reorganization involved a reduction in number of ciliary rows, a progressive inactivation and loss of one oral meridian, and a reduction and eventual disappearance of one cortical surface (semicell) situated between the two oral meridians. The intermediate steps of this reorganization included some processes that resemble those previously described in regulating doublets of other ciliates, and others that are peculiar to members of the “P. aurelia” species-group and some of its close relatives. The former included a disappearance of one cortical landmark (a contractile vacuole meridian) and transient appearance of another (a third cytoproct) within the narrower semicell. The latter included a reorganization of the paratene zone and the associated invariant (non-duplicating) region to occupy the entire narrower semicell and a redistribution of zones of most active basal-body proliferation within the opposite, wider semicell. The final steps of reorganization involved anterior displacement, invagination, and resorption of one of the two oral apparatuses and eventual disappearance of the associated oral meridian. An oral meridian deprived of its oral apparatus, either by spontaneous resorption or microsurgical removal, could persist for some time in “incomplete doublets” before regulating to the singlet condition. The phylogenetically widespread events encountered in the regulation of doublets to singlets suggest that Paramecium shares some of the global regulatory properties that are likely to be ancestral in ciliates. The more specific events are probably associated with the complex cytoskeletal architecture of this organism and with the frequent occurrence of autogamy that was described in the preceding study (Prajer et al. 1999).

    Notes: SYNOPSIS. Cells of T. pyriformis GL-C, transferred from a complete axenic medium to a medium lacking amino acids, cease dividing after several hours, and instead begin to undergo oral replacement. This process can be synchronized by a single long 33.8 C treatment. Oral replacement was also observed during stationary phase of normal culture growth in cells of strain GL-C and WH-6 (syngen 1).In strain GL-C the oral replacement primordium is initiated by the appearance of a small number of kinetosomes adjacent to the anterior end of kinety 1, just posterior to the undulating membrane (UM). The UM then loses its cilia and becomes disorganized, and is thus converted into a field of kinetosomes which is broadest near its posterior end. This UM-derived field then becomes joined with the much smaller field which had appeared earlier near the anterior end of kinety 1. As a consequence, the right margin of the UM-derived field becomes continuous with the anterior end of kinety 1, and thus comes to appear as an anterior extension of this kinety. The membranelles and UM of the new oral area differentiate within this composite field. While this is going on, the membranelles of the old oral area are progressively resorbed; these old membranelles always remain spatially separate from the oral replacement primordium.In strain WH-6, the stomatogenic field initially formed adjacent to kinety 1 is substantial, and the role of the UM kinetosomes in stomatogenesis is less obvious than in strain GL-C. The posterior portion of the UM probably contributes to the oral replacement primordium, while the anterior portion is resorbed. In this strain small supernumerary primordia are occasionally seen adjacent to the portion of kinety 2 which is nearest to the posterior region of the UM.It is suggested that the junction between the posterior portion of the UM and the neighboring cortex can serve as an inductive zone for initiation of stomatogenesis, the UM itself having a varying capacity for direct provision of kinetosomes for the stomatogenic field. The flexibility of the stomatogenic site in T. pyriformis is discussed in relation to the apparent restriction of potentialities in peniculine ciliates such as Paramecium.

    Description / Table of Contents: Zusammenfassung Zur Bestimmung der Oberflächen-OH-Gruppen im Knochenmineral, welches als unreiner Hydroxyl-apatit beschrieben werden kann, wurde Lithium-Aluminiumhydrid in Diglymelösung verwendet. Die Menge der OH-Gruppen wurde aus der entwickelten Wasserstoffmenge berechnet. Ein Vergleich der auf diesem Wege ermittelten Menge an OH-Gruppen mit der aus der spezifischen Oberfläche (BET N2-Adsorption) und der Kristallstruktur errechneten zeigt ein Defizit von mehr als 50%. Zur Deutung werden mehrere Möglichkeiten diskutiert. Die Annahme, daß echte Fehlstellen für die Oberflächen-OH-Gruppen vorliegen, wird bevorzugt.
    Notes: Summary Lithium aluminium hydride reagent has been employed in the determination of surface OH groups in bone mineral which may be described as an impure hydroxyapatite. The dehydrated sample is dropped into a diglyme solution of LiAlH4 and hydrogen gas evolution is measured. One mole of hydrogen is evolved by each mole of OH groups. The total surface is previously measured by the B.E.T. nitrogen adsorption method. Thus it is possible to compare the number of surface OH groups found per unit area with that estimated on the basis of the known crystal lattice. Less than fifty percent of the estimated number is found. Several possible explanations for the apparent discrepancy are offered. Perhaps the most compelling one is the suggestion of an actual deficiency of surface hydroxyl groups.

    Notes: Abstract We have analyzed Ca2+ currents in two neuroblastoma-motor neuron hybrid cell lines that expressed normal or glutamine-expanded human androgen receptors (polyGln-expanded AR) either transiently or stably. The cell lines express a unique, low-threshold, transient type of Ca2+ current that is not affected by L-type Ca2+ channel blocker (PN 200-110), N-type Ca2+ channel blocker (ω-conotoxin GVIA) or P-type Ca2+ channel blocker (Agatoxin IVA) but is blocked by either Cd2+ or Ni2+. This pharmacological profile most closely resembles that of T-type Ca2+ channels [1-3]. Exposure to androgen had no effect on control cell lines or cells transfected with normal AR but significantly changed the steady-state activation in cells transfected with expanded AR. The observed negative shift in steady-state activation results in a large increase in the T-type Ca2+ channel window current. We suggest that Ca2+ overload due to abnormal voltage-dependence of transient Ca2+ channel activation may contribute to motor neuron toxicity in spinobulbar muscular atrophy (SBMA). This hypothesis is supported by the additional finding that, at concentrations that selectively block T-type Ca2+ channel currents, Ni2+ significantly reduced cell death in cell lines transfected with polyGln-expanded AR.

    Notes: Abstract A study of callase activity in relation to cytogenetical expressions in cytoplasmic male sterile and male fertile Petunia indicated differential timing of the localized enzyme activity. Enzyme activity was determined by a new test. The possible relations between the mode of action of the extrachromosomal gene, timing of the enzyme activity, and male sterility are discussed.
