    Notes: Abstract  Dehydrins are water-soluble lipid-associating proteins that accumulate during low-temperature or water-deficit conditions, and are thought to play a role in freezing- and drought-tolerance in plants. Dhn genes exist as multi-gene families in plants. Previously, we screened lambda genomic libraries of two barley cultivars in an effort to isolate all of the barley Dhn genes. We identified 11 unique Dhn genes and estimated a total of 13 Dhn genes in the barley genome. To extend the collection, we used an alternative source of clones, a 1.5×Morex barley BAC library. In this library, we found nine Dhn genes that we described previously and one new Dhn gene, Dhn12. The Dhn12 gene encodes an acidic YSK2 dehydrin. The Dhn12 gene is located on chromosome 6H, and shows a different expression pattern from all other Dhn genes identified previously. RT-PCR results show that Dhn12 expression is embryo-specific. Dhn12 is not expressed in seedling shoots under any of the conditions tested, including non-stressed as well as dehydrated, or cold-, ABA- or NaCl-treated seedlings.

    Notes: Abstract  We previously identified 11 unique barley Dhn genes and found, using wheat-barley addition lines, that these genes are dispersed on four chromosomes 3H, 4H, 5H, 6H. In the present work, more precise positions of barley Dhn genes were determined using gene-specific PCR and 100 doubled haploid lines developed from a cross of Dicktoo and Morex barley. Dhn10 is located on 3H between saflp106 and ABG4. Dhn6 is at the previously determined position on 4H between SOLPRO and BCD265a. Dhn1 and Dhn2 are at the previously determined position on 5H between mR and saflp172. The Dhn locus previously called Dhn4a on barley 5H or Dhn2.2 on T. monococcum 5A is in fact Dhn9 and maps to a revised position between BCD265b and saflp218. Dhn3, Dhn4, Dhn7 and Dhn5 each map to the same position on chromosome 6H, suggesting that the previously reported separation of Dhn3, Dhn4 and Dhn5 may reflect limitations in the accuracy of Southern blot data. In addition to clarifying the map positions of these important stress-related genes, these results illustrate the advantage of gene-specific probes for the mapping of individual genes in a multi-gene family.

    Notes: Summary The extractability of125I-labelled plasma albumin from bone pieces and from powdered bone has been compared after both in vivo and in vitro incorporation. The results show that albumin is more readily extracted from bone pieces than from bone powder which implies that tissue disruption exposes additional protein adsorption sites. It is suggested that incorporation of plasma albumin into calcified matrix during bone formation occurs mainly as a result of its strong interaction with bone mineral.

    Notes: Summary Plasmaα 2HS-glycoprotein is specifically accumulated in calcified tissues. In the present studies this glycoprotein was isolated from plasma and after iodination with iodine-125 was injected intravenously into young rabbits. The tissue distribution and plasma disappearance rate of this radioactively labeled material were determined. Of the various tissues studied, bone showed the greatest retention of labeled glycoprotein expressed as percentage of the injected dose per gram tissue relative to the plasma content. The rate of loss of iodinatedα 2HS-glycoprotein from plasma was similar to that ofα 2HS-glycoprotein labeled endogenously by using14C-glucosamine or3H-glucosamine. The uptake of exogenously labeled3I-α 2HS-glycoprotein into bone tissue expressed as a percentage of the injected dose was similar to that of endogenously labeled14C-α 2HS-glycoprotein. These results suggest that the125I-labeled material can be used to study further the metabolism ofα 2HS-glycoprotein by bone tissue.

    Notes: Abstract The spectrum of plasma proteins present in human cortical bone and permanent dentine has been determined. One plasma glycoprotein, theαHS-glycoprotein, was found to be present at a high concentration in both bone and dentine and was shown to be concentrated in the mineralized tissues with respect to the other plasma proteins by factors of between 30 and 100. In this respect theαHS-glycoprotein is analogous to the G2B-glycoprotein and α-glycoprotein of bovine and rabbit b one, respectively. The binding ofαHS-glycoprotein and albumin to calcium phosphates generated within serum samples has been studied at different serum: precipitate ratios. In each case all theαHS-glycoprotein was removed from the samples and theαHS-glycoprotein was concentrated with respect to albumin by factors ranging from 370 at the highest serum: precipitate ratio to 25 at the lowest ratio. The plasmaαHS-glycoprotein concentrations of patients with Paget's disease of bone were shown to be substantially lower than the normal range, with significant negative correlation between theαHS-glycoprotein concentration and the plasma alkaline phosphatase activity.

    Notes: Summary The passage of tissue fluid through cortical bone has been investigated using radioactively labelled macromolecules as markers. The results suggest that in the cortex of young rabbit femur the movement of tissue fluid is in the same net direction as blood, mainly from the endosteal to the periosteal surface. Some albumin is incorporated from extravascular tissue fluid into calcified matrix at sites of bone formation. Polyvinylpyrrolidone, average molecular weight 35,000 is able to pass through extravascular tissue fluid in bone but isnot incorporated into calcified matrix. In rabbits made vitamin D deficient, much less albumin is retained in regions of bone formation than is the case with controls. Albumin adsorbs to the surface of calcium phosphate precipitates and it is suggested that this mechanism may be mainly responsible for its incorporation into bone.

    Notes: Abstract Blends obtained from Biopol D600G and polyamide 6 reveal in DSC investigations multiphase structure with a distinct crystalline polyamide 6 phase. Due to rapid crystallization of the polyamide 6 the crystallization of the Biopol D600G is retarded. The grade of crystallization of Biopol D600G is lower in the blends than in the pure state, as calculated from the melting enthalpies. Crystallization of polyamide 6 in the blends is faster and results in increasing of the grade of crystallization of polyamide 6 phase comparing to the unblended component.

    Notes: Abstract New polyurethanes with mesogenic units in the main chain due to the use of a liquid crystalline chain extender were synthesized from 4,4'-methylenebis(cyclohexyl isocyanate) (HMDI)using diisocyanates of different trans, trans isomer content, a low molecular diol4,4'-bis(6-hydroxyhexoxy)biphenyl (BHHBP) and a high molecular poly(hexyleneadipate)diol (PHA). The growth of trans, trans isomer content in HMDI used to syntheses of PU induces monotonic growth of melting point, rectilinear growth of crystallization temperatures and the growth of crystallization enthalpy, both for hard segment polyurethanes and block polyurethanes. The increase of trans, trans isomer content in HMDI increases also glass transition temperatures and dynamic storage modulus of the polyurethanes.

    Notes: Abstract Blends obtained from polyamide 6 and a thermoplastic polyurethane compatibilized by diblock copolyurethaneamides were investigated by means of DMTA and DSC. The blends were prepared by compounding in a Brabender mixer. The compatibilizer affected the glass transition temperature of the amorphous phase of the blends. The non-isothermal crystallization temperature of the polyamide phase was lowered in the presence of the polyurethane and the copolyurethaneamide.
