    Notes: Summary In the first inbred generation (I1) of cv. Black 4495 a dark green, slowly growing mutant (coded ds) was found, whereas the I1 of its self-compatible dihaploid, B16, comprised this ds mutant and in addition the mutants virescens (v) and yellow margin (ym). The occurrence of ds and ym might trace back to diploid S. phureja, one of the ancestors of Black 4495. No lethal mutants were observed in I1 of B16. Analysis of I1 of cv. Gineke revealed a simplex condition for virescence and either duplex or triplex heterozygosity for one lethal gene. On the other hand, the I1 of its dihaploid, G254, segregated for virescence and for three different lethal genes. It is shown that both in B16 and in G254 homozygosity of an S-bearing translocation causes early death of embryo and endosperm, thus preventing seed development. From this study it appeared that the three lethal genes from G254 affect germination rate of the seeds. The genotypes at 11 loci of B16 and G254 are presented.

    Notes: Summary Three dihaploids of Solanum tuberosum (two self-compatible, one self-incompatible) were found to be heterozygous for a monogenic recessive virescent mutant. Intercrossing resulted in the expected 3 : 1 ratio only in crosses involving one self-compatible and one self-incompatible parent. Self-compatible x self-compatible matings produced F1's in which 6:1 was found. The same ratio was observed in the self progeny of the two self-compatible dihaploids. This significant deviation could be explained by assuming linkage (25% crossing-over) between v and an S-bearing translocation. This translocation causes self-compatibility in the dihaploids used and early lethality when homozygous.

    Notes: Summary Investigations of the genetics of self-compatibility and self-incompatibility in dihaploids and diploid derivatives from cv. Gineke revealed the presence of S 1, S2 and S 3 at the S-locus of Gineke and in addition an S 1-allele on a translocation. By means of a complete tester set involving the S-alleles S 1, S2 and S 3 (all from Gineke) and S 4 (from Black 4495) it was demonstrated that some Gineke dihaploids were compatible with all six testers. This indicated a fourth S-allele in Gineke, which differs from those in the tester series and was therefore assigned S 5. Additional evidence was obtained from an analysis of F1's from crosses of two S 5-bearing dihaploids and one of the testers. So the S-genotype of cv. Gineke was identified as S 1S2S3S5/S1, the second S 1 being the S-allele on a translocated fragment.

    Notes: Complicated PCB mixtures can be separated in individual compounds using glass capillary gas chromatography, (GC)2. Depending on extraction and clean-up procedure it is also possible to separate and determine organochlorine pesticides at the same time.This (GC)2 technique can be used to determine the contents of individual chlorinated biphenyls in milk products and animal feedstuffs and in the analysis of complicated extracts of soil and vegetable material.Practical aspects concerning connection of the capillary, automatic splitless injection, repeatability of the retention time, quality of the column with respect to separation and adsorption and degradation of DDT are discussed.The detection of individual chlorinated biphenyls is possible at the ppb level in fats and vegetable materials, using an extraction and clean-up procedure, based on saponification of the sample.Preliminary results for milk, obtained from several areas, are shown.

    Notes: ---In comprehensive two-dimensional gas chromatography, two individual separations are coupled by means of a rotating thermal desorption modulator interface. The injection pulse introduced via the interface onto the second column should be as short as possible. Parameters affecting the modulator operation are studied. In the set-up used in this study, the temperature of the second column can be programmed independently from that of the first column. Optimization of the second-dimension separation to minimize peak broadening and maximize resolution is discussed and an elegant approach to determine second-dimension retention times using a non-constant modulation frequency is demonstrated. The high separation power of the comprehensive system is demonstrated by the analysis of technical and biota samples containing chlorinated biphenyls and toxaphene.

    Notes: A detection system is described which is based on fluorescent ion-pair formation between tertiary amine drugs and dimethoxyanthracene sulphonate. A dynamic micro-extraction principle is then used to isolate the ion-pairs from the excess reagent. The band broadening of this extraction detector was kept below 20% using standard auto-analyser equipment. With the enhanced selectivity and sensitivity of this approach it was possible to analyse chloropheniramine in urine at ppb concentrations using a pre-column clean-up trace enrichment step.

    Notes: The mass spectra and gas chromatographic properties of 17 synthetic fluoro-, chloro- and Bromomethoxybiphenyls and 12 dichlorodimethoxybiphenyls have been examined. From this representative series it appears that the position of the methoxy group (ortho, meta and para to the biphenyl bond) in all monomethoxy compounds examined, and the positions of the two methoxy groups in most of the dimethoxy compounds, can be assigned unambiguously by their difference in fragmentation pattern. The value of this method was shown by metabolism experiments in which 4,4′-difluoro- and 4,4′-dibromobiphenyl were fed to rats and 4,4′-dichlorobiphenyl was administered to plants. All hydroxylated metabolites found were identified by gas chromatography mass spectrometry. Relationships between structure and gas chromatographic retention time of these compounds are discussed.

    Notes: ---The potential of the on-line coupling of microcolumn liquid chromatography (μLC) using aqueous eluents with a flame ionization detector (FID) was evaluated. An eluent-jet interface was modified to allow the efficient introduction of the eluent into the FID. The potential of the method is demonstrated by the μLC-FID determination of lower alcohols and bis(2-hydroxyethylthio)alkanes on porous and non-porous stationary phases, respectively. Flow injection analysis (FIA)-FID experiments with highly polar, thermolabile, semi-volatile and non-volatile compounds like amino acids, organic acids, alkylphosphonic acids, and carbohydrates showed the developed configuration to be a promising approach for the detection of a wide range of analytes. Compared with a nebulization interface, the eluent-jet interface showed 4-10 times higher peaks for citric acid. Detection limits by FIA for all compounds were in the range of 0.2-5 ng injected. With ribose as test compound, plots of peak height vs. amount injected showed good linearity (r2 〉 0.999) in the range of 75-12,000 μg/mL. The repeatability showed relative standard deviations of less than 5%.
