    Notizen: Summary An investigation of the larval dragonfly fauna associated with the plant, Sagittaria platyphylla, was conducted in a small pond. Despite the presence of several larval anisopteran species in the pond, only Pachydiplax longipennis larvae were found on Sagittaria plants. A study of the microspatial distribution of P. longipennis larvae on S. platyphylla indicated that larvae use the various regions of a plant in a highly non-random fashion. Larvae show a strong preference for the leaf axil area. A generalized predator, the bluegill sunfish (Lepomis macrochirus), was allowed to selectively eat either of two larvae placed in various plant regions. This experiment indicated that larvae in a leaf axil area were significantly less susceptible to bluegill predation than larvae positioned in other plant regions. The microspatial distribution of starved larvae revealed that larvae with high hunger levels occupied the leaf axil area significantly less than well fed larvae, suggesting 1) larvae do not use these regions as feeding sites, and 2) high hunger levels may induce a behavioral shift in habitat use, with starved larvae forced into areas of high predation risk by the need to fulfill nutritional requirements.

    Notizen: Abstract A detailed methodology is described for fluorescence-activated cell sorting (FACS) of porcine muscle cells that have been transfected to express green fluorescent protein (GFP). Cells are liberated from porcine skeletal muscle and primary cultures are transfected with DNA encoding GFP. Primary cultures are subjected to immunocytochemistry using a primary muscle-specific monoclonal antibody followed by incubation with a phycoerythrin-conjugated second antibody. Transfected myoblasts aresorted from fibroblasts using forward angle light scatter and ninety degree light scatter, phycoerythrin fluorescence, and GFP fluorescence. These procedures allow for isolation of genetically-engineered porcine muscle cells more rapidly than traditional clonal selection procedures. Consequently, FACS provides porcine myoblast populations that retain the majority of their replicative capacity and are not contaminated with non-myogenic cells.

    Beschreibung: Following the breakup of the Space Shuttle Columbia during reentry a NASA/Contractor investigation team was formed to examine the probable damage inflicted on Orbiter Thermal Protection System elements by impact of External Tank insulating foam projectiles. The authors formed a working subgroup within the larger team to apply the Smooth Particle Hydrodynamics code SPHC to the damage estimation problem. Numerical models of the Orbiter's tiles and of the Tank's foam were constructed and used as inputs into the code. Material properties needed to properly model the tiles and foam were obtained from other working subgroups who performed tests on these items for this purpose. Two- and three-dimensional models of the tiles were constructed, including the glass outer layer, the main body of LI-900 insulation, the densified lower layer of LI-900, the Nomex felt mounting layer, and the Aluminum 2024 vehicle skin. A model for the BX-250 foam including porous compression, elastic rebound, and surface erosion was developed. Code results for the tile damage and foam behavior were extensively validated through comparison with Southwest Research Institute foam-on-tile impact experiments carried out in 1999. These tests involved small projectiles striking individual tiles and small tile arrays. Following code and model validation we simulated impacts of larger foam projectiles on the examples of tile systems used on the Orbiter. Results for impacts on the main landing gear door are presented in this paper, including effects of impacts at several angles, and of rapidly rotating projectiles. General results suggest that foam impacts on tiles at about 500 mph could cause appreciable damage if the impact angle is greater than about 20 degrees. Some variations of the foam properties, such as increased brittleness or increased density could increase damage in some cases. Rotation up to 17 rps failed to increase the damage for the two cases considered. This does not rule out other cases in which the rotational energy might lead to an increase in tile damage, but suggests that in most cases rotation will not be an important factor.

    Beschreibung: This report compares seven double-plate penetration predictor equations for accuracy and effectiveness of a shield design. Three of the seven are the Johnson Space Center original, modified, and new Cour-Palais equations. The other four are the Nysmith, Lundeberg-Stern-Bristow, Burch, and Wilkinson equations. These equations, except the Wilkinson equation, were derived from test results, with the velocities ranging up to 8 km/sec. Spreadsheet software calculated the projectile diameters for various velocities for the different equations. The results were plotted on projectile diameter versus velocity graphs for the expected orbital debris impact velocities ranging from 2 to 15 km/sec. The new Cour-Palais double-plate penetration equation was compared to the modified Cour-Palais single-plate penetration equation. Then the predictions from each of the seven double-plate penetration equations were compared to each other for a chosen shield design. Finally, these results from the equations were compared with test results performed at the NASA Marshall Space Flight Center. Because the different equations predict a wide range of projectile diameters at any given velocity, it is very difficult to choose the "right" prediction equation for shield configurations other than those exactly used in the equations' development. Although developed for various materials, the penetration equations alone cannot be relied upon to accurately predict the effectiveness of a shield without using hypervelocity impact tests to verify the design.
