    Notes: Abstract The paper deals with the numerical solution techniques for the traction boundary integral equation (BIE), which describes the opening (and sliding) displacements of the surface of the traction loaded crack or arbitrary planform embedded in an elastic infinite body (buried crack problem). The traction BIE is a singular integral equation of the first kind for the displacement gradients. Its solution poses a number of numerical problems, such as the presence of derivatives of the unknown function in the integral equation, the modeling of the crack front displacement gradient singularity, and the regularization of the equation's singular kernels. All of the above problems have been addressed and solved. Details of the algorithm are provided. Numerical results of a number of crack configurations are presented, demonstrating high accuracy of the method.

    Notes: Summary Glycophorins alpha and delta are the carriers of the antigens of the MNSs blood system; this report documents the presence of three glycophorin mutations in two individuals of a 16 member family. Erythrocytes were examined by serology, sodium dodecyl sulfate electrophoresis, and immunoblotting. The inheritance pattern and immunoblot profile revealed: (1) A variant Dantu glycophorin showed properties consistent with a delta-alpha glycophorin hybrid structure, previously noted in other individuals. The gene responsible for the Dantu glycophorin in this family is linked to a gene coding for an M-specific alpha glycophorin. (2) Another variant glycophorin, Mi-III glycophorin, was first revealed by immunoblotting and subsequently confirmed by erythrocyte antigen typing. This autosomal dominant trait is associated with N blood group activity and the inheritance pattern indicates that it could be a variant of delta glycophorin. (3) In the individuals with both Dantu and Mi-III glycophorins a delta glycophorin deficiency was observed suggesting that a deletion or alteration of delta gene may exist cis to the Dantu gene. Our findings that document clustering of multiple mutations in MNSs gene loci in the propositus family are very unusual as such variants are relatively rare.

    Notes: Abstract  Lycopersicon hirsutum G1.1560 is a wild accession of tomato that shows resistance to Oidium lycopersicum, a frequently occurring tomato powdery mildew. This resistance is largely controlled by an incompletely dominant gene Ol-1 near the Aps-1 locus in the vicinity of the resistance genes Mi and Cf-2/Cf-5. Using a new F2 population (n=150) segregating for resistance, we mapped the Ol-1 gene more accurately to a location between the RFLP markers TG153 and TG164. Furthermore, in saturating the Ol-1 region with more molecular markers using bulked segregant analysis, we were able to identify five RAPDs associated with the resistance. These RAPDs were then sequenced and converted into SCAR markers: SCAB01 and SCAF10 were L. hirsutum-specific; SCAE16, SCAG11 and SCAK16 were L. esculentum-specific. By linkage analysis a dense integrated map comprising RFLP and SCAR markers near Ol-1 was obtained. This will facilitate a map-based cloning approach for Ol-1 and marker-assisted selection for powdery mildew resistance in tomato breeding.

    Notes: Abstract Based upon the strength criterion proposed by Gol'denblat and Kopnov, the second-, fourth-, and sixth-rank strength tensors in the three-dimensional case have been determined for each of the crystal classes from consideration of invariant transformations of the strength function. A cubic polynomial form has been proposed as an improved strength criterion for anisotropic brittle materials by using the constitutive laws in continuum mechanics. The methods of determination of the strength coefficients and the strength envelope in the biaxial stress state for graphite are presented in detail by relating the cubic polynomial strength criterion to the experimental test data. Numerical examples are presented to show that the theoretical and experimental results are in good agreement. A comparison with the results previously published is presented.

    Notes: Abstract Two phospholipases A2 (PLA2) fromNaja naja atra andNaja nigricollis snake venoms were subjected to tyrosine modification withp-nitrobenzenesulfonyl fluoride (NBSF) atpH 8.0. Three major NBS derivatives from each PLA2 were separated by high-performance liquid chromatography. The results of amino acid analysis showed that only two Tyr residues out of nine were modified, and the modified residues were identified to be Tyr-3 and Tyr-63 (or Tyr-62) in the sequence. Spectrophotometric titration indicated that the phenolic group of Tyr-3 and Tyr-63 (or Tyr-62) had apK of 10.1 and 11.0, respectively. The reactivity of Tyr-3 toward NBSF was not affected in the presence or absence of Ca 2+; however, the reactivity of Tyr-63 (or Tyr-62) toward NBSF was greatly enhanced by Ca2+. Modification of Tyr-63 (or Tyr-62) resulted in a marked decrease in both lethality and enzymatic activity. Conversely, modification of Tyr-3 inN. naja atra PLA2 could cause more than a sixfold increase in lethal potency, in sharp contrast to the loss of enzymatic activity. Tyrosine-63-modifiedN. naja atra PLA2 exhibited the same Ca2+-induced difference spectra as that of native PLA2, indicating that the Ca2+-binding ability of Tyr-63-modifiedN. naja atra PLA2 was not impaired. However, Tyr-3-modified PLA2 and all Tyr-modifiedN. nigricollis CMS-9 were not perturbed by Ca2+, revealing that the Ca2+-binding ability have been lost after tyrosine modification. These results suggest that Tyr-62 inN. nigricollis CMS-9 and Tyr-3 in both enzymes are involved in Ca2+ binding. AtpH 8.0, both native PLA2 enzymes enhance the emission intensity of 8-anilinonaphthalene sulfonate (ANS) dramatically, while all of the Tyr-modified derivatives did not enhance the emission intensity at all either in the presence or absence of Ca2+, suggesting that the hydrophobic pocket that interacts with ANS might be the substrate binding site, in which Tyr-3 and Tyr-63 (or Tyr-62) are involved.
