    Notizen: Summary Anthers of male fertile, cytoplasmic male sterile (cms), and restored male fertile Petunia hybrida, are analyzed for cytochrome c oxidase (cox) activity in subsequent stages of microsporogenesis, and compared with anthers of male fertile, cms-S and cms-C Zea mays. The cox activity is determined in anther extracts and cytochemically. In petunia anthers, the first differences in cox activity occur from meiosis onward. However, at these stages, the initial symptoms of degeneration are already apparent. It is suggested that the decline in enzyme activity of the cms petunia anthers is the result rather than the cause of the non-formation of functional pollen. In maize anthers, the cox activity of sterile-type anthers is reduced in comparison with fertile-type anthers from premeiosis onward. There are also consistent cytochemical differences in the mitochondrial organization of cox activity between pollen of cms-S and male ferile maize anthers. In fertile-type mitochondria, the DAB reaction product indicating cox activity is localized in the cristae and within the space between the outer and inner limiting membranes of the organelles. In mitochondria of pollen of cms-S maize, cox activity is only observed between the outer and inner membranes of the mitochondria. The biochemical and cytochemical differences are observed at stages of development at which no structural signs of degeneration are apparent. The results suggest that cms in maize correlates with deviations in cytochrome c oxidase activity.

    Notizen: Summary Phenotypic revertants from several kinds of mutations, including deletions, have been detected by pollen analysis at the wx and Adh loci in maize. Mutations in these genes give phenotypic revertants with median frequencies of 0.7 and 0.5×10−5, respectively. However, the nature of such revertants can only be analyzed following their recovery from conventional matings. In the current study large seed populations derived from crosses involving several bz (bronze) locus mutations in maize were examined for reversion to a Bz (purple) expression. Deletion, insertion and point mutations were included in the study. Principally, over 2 million gametes of the bz-R mutation, which is shown here to be associated with a 340 base pair deletion within the transcribed region of the gene, have been screened for reversion. No revertants from it or any of the other bz mutations have been recovered, even though a total of almost 5 million gametes from homoallelic crosses have been examined to date. Results from seed analysis are discussed in reference to those from pollen analysis in maize.

    Notizen: Summary Inoculated and non-inoculated seedlings of Zea mays were grown in agricultural soils under aseptic and non-sterile conditions. Acetylene reduction activity and microbial counts were determined after 7 and 30 days of growth. Irrespective of the soil type Azotobacter spp. were commonly isolated under maize cultivation. Inoculation of agricultural soils with a suspension of A. chroococcum led to an increase in Azotobacter numbers, although this effect diminished with time. Nitrogenase activity was detected on maize roots and increased in response to the inoculation with A. chroococcum, showing that this associative growth could be of primary importance for the plant. The results of assays for acetylene reduction activity indicated that the nitrogenase activity was associated only with the root systems.

Application of 40 kg N ha−1 plus A. chroococcum caused a significant increase in maize yield. Azotobacter inoculation was more efficient at lower doses (40 kg N ha−1) than at high doses (80 kg N ha−1) of urea.

    Notizen: Abstract  We hypothesized that the integration of trees and shrubs in agricultural landscapes can reduce NO3 – leaching and increase utilization of subsoil N. A field survey was conducted on 14 farms on acid soils in the subhumid highlands of Kenya, where there is little use of fertilizers, to determine the effect of vegetation types (VT) on soil NH4 + and NO3 – to 4 m depth. The VT included maize (Zea mays) with poor growth and good growth, Markhamia lutea trees scattered in maize, natural weed fallow, banana (Musa spp.), hedgerow, and eucalyptus woodlot. The effect of VT on NH4 + was small (〈1 mg N kg–1). NO3 – within a VT was about constant with depth below 0.25 m, but subsoil NO3 – varied greatly among VT. Mean NO3 –-N concentrations at 0.5–4 m depth were low beneath hedgerow and woodlot (〈0.2 mg kg–1), intermediate beneath weed fallow (0.2–0.7 mg kg–1), banana (0.5–1.0 mg kg–1) and markhamia (0.5–1.6 mg kg–1), and high beneath both poor (1.0–2.1 mg kg–1) and good (1.9–3.1 mg kg–1) maize. Subsoil NO3 – (0.5–4 m) was agronomically significant after maize harvest with 37 kg N ha–1 m–1 depth of subsoil beneath good maize and 27 kg N ha–1 m–1 depth beneath poor maize. In contrast, subsoil NO3 – was only 2 kg N ha–1 m–1 depth beneath woodlot and hedgerow. These results demonstrate that the integration of perennial vegetation and the rotation of annual and perennial crops can tighten N cycling in agricultural landscapes.

    Notizen: Summary Foreign pollen tubes in the stigma of Zea mays can be prevented from reaching the ovary cavity by the unusual length of the pollen tube pathway. A simple and rapid procedure is described for overcoming this difficulty by pollinating the basal parts of the stigmas without removing the ensheathing bracts (“husks”). The method maintains high humidity in the vicinity of the ovaries, and by conserving photosynthetic tissues probably also ensures a more normal O2 /CO2 balance in the neighbourhood of the stigmas than do bagging procedures. It is shown that Sorghum pollen tubes readily reach the ovary after pollination by the method. Their presence induces some of the characteristic post-pollination effects caused by Zea pollen tubes, but they frequently also stimulate premature enlargement of the nucellus and lysis of nucellar cells. Although Sorghum tubes have been traced across the inner ovary wall, they have not been seen to enter the micropyle, and hybrid embryos have not yet been obtained.

    Notizen: Summary A chromosome doubling technique, involving colchicine treatment of an embryogenic, haploid callus line of maize (Zea mays L., derived through anther culture), was evaluated. Two colchicine levels (0.025% and 0.05%) and three treatment durations (24, 48, and 72 h) were used and compared to untreated controls. Chromosome counts and seed recovery from regenerated plants were determined. No doubled haploid plants were regenerated from calli without colchicine treatment. After treatment with colchicine for 24 h, the callus tissue regenerated about 50% doubled haploid plants. All of the plants regenerated from the calli treated with colchicine for 72 h were doubled haploids, except for a few tetraploid plants. No significant difference in chromosome doubling was observed between the two colchicine levels. Most of the doubled haploid plants produced viable pollen and a total of 107 of 136 doubled haploid plants produced from 1 to 256 seeds. Less extensive studies with two other genotypes gave similar results. These results demonstrate that colchicine treatment of haploid callus tissue can be a very effective and relatively easy method of obtaining a high frequency of doubled haploid plants through anther culture.

    Notizen: Summary Two free-space marker procedures (Prussian blue and lanthanum nitrate) were employed to determine the pathway(s) followed by water and solutes in the transpiration stream after their introduction into the xylem of small and intermediate bundles, and the effectiveness of the suberin lamellae of the bundle-sheath cells as a barrier to the movement of tracer ions (Fe3+ and La3+). Judged from the distribution of Prussian-blue crystals (insoluble, crystalline deposits resulting from the precipitation of ferric ions by ferrocyanide anions) and lanthanum deposits, water and the tracer ions moved readily from the lumina of the vessels into the apoplast (cell wall continuum) of the phloem and bundle-sheath cells via portions of vessel primary walls not bearing lignified secondary wall thickenings. Prussian blue and lanthanum deposits were abundant on the bundlesheath cell side of the bundle sheath/mesophyll interface but few occurred on that of the mesophyll, indicating that the suberin lamella is an effective barrier to apoplastic movement of both ferric and lanthanum ions. The presence of Prussian-blue crystals and lanthanum deposits in the compound middle lamella of the radial wall of the bundle-sheath cells indicates that the compound middle lamella provides an apoplastic pathway for transpirational water from the xylem to the evaporating surfaces of the mesophyll and epidermal cells.

    Notizen: Summary Arabinogalactan proteins (AGPs) are proteoglycans detected in high amounts at plant cell surfaces; however, details of their subcellular localization are largely unknown. Immunolocalization studies with the anti-AGP monoclonal antibody LM2 have indicated that this AGP epitope is associated with secretory compartments such as endoplasmic reticulum and Golgi apparatus within plant cells actively producing and secreting AGPs. The LM2 epitope contains a β-linked glucuronic acid residue and occurs in the polysaccharide moiety of AGPs. We have localized this AGP epitope also to the tonoplast and to cytoplasmic strands. Endomembrane association of AGPs was confirmed with two other monoclonal antibodies, JIM13 and MAC207, both reacting with carbohydrate AGP epitopes containing GlcpA-β(1→3)-D-GalpA-α(1→2)-L-Rha residues. Immunocytochemistry is supported by biochemical analysis which shows that LM2 reacts with the microsomal fraction and also with low-molecular-weight material of the detergent phase after Triton X-114 phase separation prepared from maize roots. Our results indicate that some AGP epitopes are closely associated with endomembranes.
