    Description: A worldwide initiative in structural genomics aims to capitalize on the recent successes of the genome projects. Substantial new investments in structural genomics in the past 2 years indicate the high level of support for these international efforts. Already, enormous progress has been made on high-throughput methodologies and technologies that will speed up macromolecular structure determinations.     Description: Endocytic proteins such as epsin, AP180, and Hip1R (Sla2p) share a conserved modular region termed the epsin NH2-terminal homology (ENTH) domain, which plays a crucial role in clathrin-mediated endocytosis through an unknown target. Here, we demonstrate a strong affinity of the ENTH domain for phosphatidylinositol-4,5-bisphosphate [PtdIns(4,5)P2]. With nuclear magnetic resonance analysis of the epsin ENTH domain, we determined that a cleft formed with positively charged residues contributed to phosphoinositide binding. Overexpression of a mutant, epsin Lys76 --〉 Ala76, with an ENTH domain defective in phosphoinositide binding, blocked epidermal growth factor internalization in COS-7 cells.     Description: Hairpin formation by single-stranded DNA molecules was exploited in a DNA-based computation in order to explore the feasibility of autonomous molecular computing. An instance of the satisfiability problem, a famous hard combinatorial problem, was solved by using molecular biology techniques. The satisfiability of a given Boolean formula was examined autonomously, on the basis of hairpin formation by the molecules that represent the formula.     Notes: The number density of dangling bonds (spin density) and the hydrogen content as well as the hydrogen-bonding configuration were independently controlled in the glow-discharge hydrogenated amorphous Si-Ge alloys using a cathode-heating technique. A tentative mechanism is presented in terms of the top surface process as well as the growth zone.

    Notes: Solid-phase crystallization processes of molecular-beam-deposited amorphous GaAs with various As/Ga composition ratios are investigated by reflection high-energy electron diffraction, Rutherford backscattering spectroscopies, and time-resolved reflectivity measurements at various annealing temperatures. In a low-temperature region ((approximately-less-than)300 °C), solid-phase epitaxial growth takes place, and the activation energy of the growth rate (1.6 eV) is shown to agree well with that of the ion-implanted amorphized GaAs. In a high-temperature region ((approximately-greater-than)400 °C), polycrystallization takes place, followed by a realignment of the polycrystal to a single crystal. The epitaxial growth rate is more strongly affected by the As/Ga composition ratio for the low-temperature crystallization than for the high-temperature crystallization. The interface roughnesses between crystallized and amorphous layers are evaluated to be 14–20 nm from the reflectivity measurements and the transmission electron microscopy. The atomic bonding state in the amorphous GaAs is examined by Auger electron spectroscopy, and no local precipitation of As atoms is detected in the nearly stoichiometric film.

    Notes: The localized density of states g(E) near midgap for undoped, B2H6-doped, and PH3-doped hydrogenated amorphous silicon is evaluated from the electrophotographic (xerographic) method. The density of hole traps decreases with increasing B2H6 concentration and that of electron traps decreases with increasing PH3. These features are qualitatively similar to those deduced from the deep-level transient spectroscopy, the photothermal deflection spectroscopy, the time-of-flight measurements, and the isothermal capacitance transient spectroscopy. The energy positions of neutral dangling bonds D0 and doubly occupied D− are estimated to be 0.85–0.88 eV from the valence band and 0.62–0.67 eV from the conduction band, respectively. Comparison of the energy peaks of electron and hole traps shows that the positive correlation energy of dangling bonds is about 0.3–0.4 eV.

    Notes: Summary A prenatal diagnosis of GM1-gangliosidosis was made in a pregnancy at risk, on the basis of a deficiency of β-galactosidase activity demonstrated in cultured aminiotic fluid cells. Biochemical analyses were performed in the aborted fetus. GM1-ganglioside β-galactosidase activity was reduced to 1% of the control value in both the brain and liver of the affected fetus. Lamellar bodies suggestive of membranous cytoplasmic bodies were found in cells of basal ganglions, while the accumulation of GM1-ganglioside in the brain was not remarkable.
