    Notes: The vibrational relaxation of the ν4 mode of CH3NO2 liquid and its molecular dynamics are investigated. Temperature and solvent effects on vibrational dephasing and correlation times are discussed in terms of the adiabatic impact theory to infer that, in the molecular collision, the attractive part of intermolecular potential is explored. The statistical uncorrelation of rotational and vibrational relaxations is experimentally demonstrated.

    Notes: The pattern of cytospindle assembly and the modifications of the microtubular cytoplasmic network during division of Paramecium are studied by means of indirect immunofluorescence. The assembly of cytospindle starts at two independent areas placed respectively around the proter's and opisthe's buccal overture. The moment of the microtubule bundles’appearance depends on their distance from the buccal opening, with those closest appearing 1st. The existence of microtubule organizing centers that act transiently during division of Paramecium is discussed.

    Notes: The mechanism of porous silicon (PS) film growth remains a debate among researchers. This study investigates the effect of current density (J) and anodization time (t) on the film thickness, optical properties, and microstructure of films grown at low current densities. The samples reported herein were generated from polished and unpolished 〈100〉 single crystal wafers, with resistivities of 0.1 and 0.6 Ω cm, respectively. Current densities of 1–10 mA/cm2 and anodization times ranging from 3 to 900 s were used. This work shows that film growth is at first linear but beyond a given anodization time (dependent on J), the film produced is not continuous, but is best described as a finite number of stacked sublayers with each sublayer having a unique microstructure, porosity, and optical response. This article also shows that as the PS film grows, second and third order peaks appear in the reflectance spectra. This data, together with modulations in the index of refraction spectra and visual evidence from cross-sectional scanning electron microscopy (SEM) images demonstrate that submicron PS thin films grown at low current densities have a multilayer structure. Above a certain thickness, the modulation in the properties due to the sublayers phases out and the films appear as if they were single layered films. SEM is used to show the progression, from cross-sectional imaging, from a layered or stacked PS film structure to an apparently homogeneous film with depth. When using low current densities, single layer films are only obtained at very short times. © 2000 American Institute of Physics.

    Notes: A basic program is described that runs on any PC-IBM. This program allows data acquisition either from most interfaciable devices (by means of IEEE 488 or RS 232), or suitable cards such as the 8255. During acquisition, recorded data can be processed and displayed, and several commands can be executed so as to modify the acquisition itself in real time. The set of commands forms a sort of programming language and an ASCII file of such commands can be executed. Some applications, already in use in our laboratory, are also described.

    Notes: The first Auger electron–ion coincidence experiment on the nitrogen molecule excited by electron impact is described. The kinetic energy releases of the N++ and the N+ fragments in the 43–72 eV binding energy range have been measured.     Notes: New multireference, configuration-interaction potential energy surfaces are reported for the interaction of Ar with the CH radical in its ground (X 2Π) and second excited (B 2Σ−) electronic states. These potential energy surfaces are then used in an adiabatic analysis of the rovibronic levels of the ArCH(X) and ArCH(B) van der Waals complexes. A qualitative discussion of the expected features in the B˜←X˜ electronic spectrum of ArCH is presented, and these are compared with the experimental spectrum reported earlier by Lemire et al. [J. Chem. Phys. 99, 91 (1993)].

    Notes: High-resolution SI nuclease mapping of mRNA synthesised in vivo, in vitro run-off transcription with RNA polymerase from Streptomyces lividans and gene fusions were used to analyse the transcriptional organization of the Sall restriction-modification system of Streptomyces albus G. The sallR and sallM genes that encode the restriction endonuclease and its cognate methyltransferase constitute an operon which is mainly transcribed from sal-pR1, a promoter located immediately upstream of saIIR, with two possible minor promoters further upstream. Another promoter, sal-pM, is within the 3′ end of the saIIR coding region, and allows expression of the modification gene in the absence of sal-pR1. The sal-pM promoter might be involved in the establishment of modification prior to restriction endonuclease activity. Sequences upstream of the apparent transcriptional start sites for sat-pR1 and sal-pM show similarity with the −10 region of typical vegetatively expressed eubacterial promoters, but appropriately centered −35 regions are absent.

    Notes: The gene responsible for the optochin-sensitive (OptS) phenotype of Streptococcus pneumoniae has been characterized. Sequence comparisons indicated that the genes involved encoded the subunits of the F0 complex of an H+-ATPase. Sequence analysis and transformation experiments showed that the atpC gene is responsible for the optochin-sensitive resistant (OptS/OptR) phenotype. Our results also show that natural as well as laboratory OptR isolates have arisen by point mutations that produce different amino acid changes at positions 48, 49 or 50 of the ATPase c subunit. The nucleotide sequence of the F F0 complex of the Streptococcus oralis ATPase has also been determined. In addition, comparison of the sequence of the atpCAB genes of S. pneumoniae R6 (OptS) and M222 (an OptR strain produced by inter-species recombination between pneumococcus and S. oralis), and S. oralis revealed that, in M222, an interchange of atpC and atpA had occurred. We also demonstrate that optochin specifically inhibited the membrane-bound ATPase activity of the S. pneumoniae wild-type (OptS) strains, and found a 100-fold difference between OptS and OptR strains, both in growth inhibition and in membrane ATPase resistance.

    Notes: Changes in expression of ribosomal protein genes during growth and stationary phase of Streptomyces coelicolor A3(2) in liquid medium were studied. Proteins being synthesized were pulse-labelled with [35 S]-methionine, separated by two-dimensional poly-acrylamide gel electrophoresis, and quantified using the Bioimage computer software. Most of the ribosomal proteins were synthesized throughout the life cycle. Exceptions were two proteins whose synthesis drastically decreased at the approach of stationary phase. These two proteins were identified in purified ribosomes as homologues of Escherichia coli ribosomal proteins L10 and L7/L12, using antibodies raised against fusion proteins between these ribosomal proteins and Escherichia coliβ-galactosldase. The genes (rplJ and rplL) encoding the L10 and L7/L12 proteins were contained in a 1.2 kb BamHl fragment that was cloned and sequenced. The linkage and order of the genes coincide with other L10-L7/L12 operons. However, L11 and L1 genes were not present immediately upstream of the L10 gene, as is the case for E. coli and other bacteria. Instead, two open reading frames of unknown function were found immediately upstream of the L10 gene, in an adjacent 1.9 kb BamHl fragment.

    Notes: Resistance to third-generation cephalosporins in a clinical isolate of Streptococcus pneumoniae was shown to be due to the production of altered forms of penicillin-binding proteins (PBPs) 2X and 1A. The cloned PBP2X gene from the resistant strain was able to transform a susceptible strain to an intermediate level of resistance. The resulting transformant could be transformed to the full level of resistance of the clinical isolate using the cloned PBP1A gene from the latter strain. Chromosomal DNA from the resistant strain (and from other resistant strains) could readily transform a susceptible strain to the full level of resistance to third-generation cephalosporins (〉250-fold for cefotaxime; 〉100-fold for ceftriaxone) in a single step (transformation frequency of about 10-5). The resistant transformants obtained with chromosomal DNA were shown by gene fingerprinting to have gained both the PBP1A and PBP2X genes from the DNA donor.
