    Notes: Abstract Aspects of the intermittent fine structure of an odor plume were mimicked and experimentally modified in the wind tunnel using an air-pulsing device. Filaments of a behaviorally active blend of six sex-pheromone components created by the device in a temporally regular fashion elicited sustained upwind flight and source location in male Heliothis virescens.Males did not exhibit sustained upwind flight in significant numbers until a frequency of 4 filaments/s was delivered, at a loading of 1 μg of the major component, Z11-16: Ald, with the other components loaded at their appropriate ratios. A loading of 10 μg Z11-16: Ald was found to be optimal at this filament delivery rate. Electroantennogram recordings to different filament delivery rates of the complete blend indicated that a stationary male antenna can respond to the pulse rates used in this study. Importantly, when a main component necessary for upwind flight, Z9-14:Ald, was isolated into its own filaments and pulsed alternately against filaments of the five other components (including the other component essential for upwind flight, Z11-16: Ald), upwind flight to the source was significantly reduced (9%) compared to upwind flight and source location in response to filaments composed of the entire blend (30%), indicating that the complete pheromone blend must arrive on the antenna simultaneously for optimal evocation of sustained upwind progress. Neurophysiological evidence from other studies suggests that higher-order interneurons whose phasic response is enhanced when the entire blend is presented simultaneously may be of importance in explaining this behavioral difference stemming from synchronous vsasynchronous arrival of the components.

Helicoverpa zea males flew upwind and successfully contacted the source when presented with 2-component blends consisting of their principal conspecific sex pheromone component, (Z)-11-hexadecenal, plus small amounts of (Z)-9-tetradecenal, a key secondary component in the Heliothis virescens blend which has heretofore been considered antagonistic to H. zea pheromone-mediated behavior. Neurophysiological studies of H. zea antennal receptor neurons and central interneurons had suggested that this unexpected antagonistic effect on behavior might occur. 2. When the amount of (Z)-9-tetradecenal in the blend reached 15% relative to the principal component its effect did become antagonistic with significantly more H. zea males remaining quiescent. Five-to-fifteen percent (Z)-9-tetradecenal is emitted by H. virescens in its pheromone blend, levels that evoked optimal upwind flight and source contact in H. virescens males. 3. As suggested by studies of H. virescens antennal receptor neurons, H. virescens males were unresponsive to the reciprocal inter-specific blend, comprised of (Z)-11-hexadecenal plus various percentages of (Z)-9-hexadecenal. 4. Receptors that allow such mutual replacement of compounds might permit significant shifts in pheromone systems; a single mutation that drastically alters the female sex pheromone blend could still be carried in a population due to the successful attraction of normal males by mutant females.

    Notes: Abstract Sex pheromone gland volatiles from individualHeliothis virescens (F.) females were collected and analyzed on an SP-2330 capillary gas-liquid chromatography column for identification and quantification of the compounds emitted. Only four of the seven compounds previously reported as pheromone components appeared consistently in the volatile collections: 14:Ald, Z9-14:Ald, 16:Ald, and Z11-16:Ald. The female glands did not emit the same amounts of these compounds throughout a 24-hr period; they emitted maximum quantities between 6 and 11 hr after the onset of scotophase with the remainder of the photoperiod having minimal emission rates. Although the absolute quantities fluctuated, the percent compositions of the compounds remained about the same throughout the 24-hr period.

    Notes: Abstract Each of the seven compounds that have been identified from femaleHeliothis virescens sex pheromone glands was examined for its ability to elicit sexual responses from male moths in a flight tunnel. The two compounds initially described as pheromone components, (itZ)-11-hexadecenal and (itZ)-9-tetradecenal, were necessary for behavioral activity to occur. Of the remaining five compounds, hexadecanal was most consistent in elevating behavioral activity of males when it was added to treatments. Live, calling females elicited greater sexual activity from males than did the 7-compound mixture on rubber septa.

    Notes: Studies on the metabolism of the tricyclic antidepressant trimipramine, 5-(3-dimethylamino-2-methylpropyl)-10, 11-dihydro-5H-dibenz[b, f] azepine, in the rat are described. Many metabolites of trimipramine (TMP) were isolated from rat urine after enzymatic hydrolysis and their structures were identified by a gas chromatographic/mass spectrometric method, before and after appropriate chemical derivatization. Besides unchanged TMP, 20 metabolites were characterized (underivatized, and after acetylation), of which 12 had undergone alicyclic (C10 or C11) oxidation. This is a hitherto unreported metabolic pathway for TMP in the rat.

    Notes: An assay for the determination of the neuromuscular blocking agents pancuronium bromide and vecuronium bromide in plasma or urine has been developed. This method is based on syringe application of sample extracts to the moving belt surface and single metastable transition monitoring of the elimination of acetic acid from the ion of m/z 543 during chemical ionization. The analysis shows good linearity over three orders of magnitude and is capable of analyzing for the compounds below the 5 ng ml-1 level. The assay has been used in preliminary pharmacokinetic studies of the biological fluids of surgical patients. The utility of the method for simultaneous determination of the deacetylated metabolites of these two drugs has also been investigated.

    Notes: The thermospray mass spectra of arteether and 16 of its potential metabolites all showed strong [M + NH4]+ ions and with only a few exceptions these compounds also showed spectral peaks corresponding to [M + NH4 - HOR]+ and [M + H - HOR]+, where OR represents the alkoxy or hydroxy group at the 12-position. A method for quantifying the metabolites was developed in which the plasma was spiked with an internal standard (the propyl ether analog of arteether), extracted using a C-18 solid-phase cartridge, then subjected to thermospray high-performance liquid chromatographic/mass spectrometric analysis using selected ion monitoring and a C-18 reversed-phase analytical column. Following the intravenous administration of arteether (11.6 mg kg-1), the plasma was found to contain 12 metabolites of arteether in the 10-1000 ng ml-1 range 15 min post-injection, and within 60 min two of these metabolites attained higher concentrations than that of the parent compound, while several other of the metabolites attained concentrations similar to the parent compound. The pseudo-first-order half-life of arteether was found to be 10.0 ± 0.6 min, while the apparent half-lives of most of the metabolites were in the 15-30 min range. Nine of these metabolites were identified by comparison to authentic reference standards and the structures of three remaining metabolites were tentatively assigned from their spectral and chromatographic properties. The metabolic pathways leading to these 12 metabolites was a rather complex, multiple-step process, but most of the metabolites arose from an enzymatic oxidation at one of three sites; 3α, 9α, or the CH2 of the side-chain. Conversion of the endoperoxide group to an cyclic ether was not a major pathway. The in vitro antimalarial activity of reference standards of several of the metabolites was determined and all of those tested were found to be active in the low nanogram per milliliter range.

    Notes: The sequences of synthetically prepared methylphosphonate oligodeoxyribonucleotides have been verified using ionspray tandem mass spectrometry with sample introduction via flow injection. The technique involves the use of product-ion scans from multiply protonated (4+ and 5+) precursors. Among the ions detected are several series of fragments of different charge states that indicate the base sequence of the intact molecule. Oligomers as large as 18 bases have been successfully characterized.

    Notes: Matrix-assisted laser desorption/ionization (MALDI) mass spectrometry has been used to measure accurately the molecular masses of synthetic methylphosphonate oligodeoxyribonucleotides, up to 18 nucleotides in length. A simple method has been developed for the complete sequence verification of these compounds, which are intractable by classical means. Sequencing from the 5′ end of the molecule is possible because of inefficiencies in the synthetic procedure. Complementary information from the 3′ end can be obtained by partial hydrolysis of the methylphosphonate backbone.
