Notizen: Abstract: We develop a characterization method of electroconvection structures in a planar nematic liquid crystal layer by a study of the electric current transport. Because the applied potential difference has a sinusoidal time dependence, we define two electric Nusselt numbers corresponding to the in-phase and out-of-phase components of the current. These Nusselt numbers are predicted theoretically using a weakly nonlinear analysis of the standard model. Our measurements of the electric current confirm that both numbers vary linearly with the distance from onset until the occurence of secondary transitions. A systematic comparison between our theoretical and experimental results, using no adjusted parameters, demonstrates moderate agreement, but discrepancies remain. Electric transport measurements during electroconvection represent a quantitative test of the standard model completely independent from optical probes. Thus, the technique described here can be a useful complement to traditional structural measurements.

Notizen: Abstract The objective of this study was to develop a method to indelibly mark mammary cells to follow cell lineage. This was done by infecting mouse mammary glands in whole organ culture (WOC) with a retroviral vector containing the β-galactosidase (BAG) gene. Abdominal mammary glands from BALB/c mice primed with estrogen and progesterone were removed and injected with 30 µl of concentrated vector prepared from CRE BAG 2 (ATCC 1858 — CRL) cells or left uninjected and co-cultured with CRE BAG 2 feeder layers for 48 or 96 hours. Glands were floated on siliconized lens paper and incubated in Waymouth's 752/1 medium supplemented with insulin, aldosterone, hydrocortisone, prolactin, and epidermal growth factor for 5 days. Integration of BAG was determined with a colorimetric assay using X-gal, a β-galactoside analog, in whole mounts and collagenase digested glands. Blue cells in collagenase digests and blue areas in whole mounts indicated that BAG had integrated into mammary cells during whole organ culture.

Notizen: Abstract We studied the influences of food type, food quantity, water currents, starvation and light on growth and reproduction of the sea hare Aplysia oculifera (Adams and Reeve, 1850) under laboratory conditions. Out of five species of algae served as food, Enteromorpha intestinalis promoted the fastest growth of A. oculifera, Ulva spp. slower growth, Cladophora sp. allowed maintenance of steady body mass, and the brown algae Colpomenia sp. and Padina pavonia were rejected by the sea hares. When sea hares were exposed to four levels of water currents, growth rates decreased as water currents increased. Sea hares fed on 50% ration grew slower than those fed on 100% ration (ad libitum). During 10 days of starvation sea hares lost weight, but when subsequently fed 100% ration they recovered and grew at a rate similar to those fed continuously with 100% ration. Under shade and under natural sunlight sea hares grew at the same rates. Whenever growth rates decreased, sea hares began to spawn at a smaller body size. A. oculifera demonstrated physiological plasticity that adapted them to varied and unpredictable environmental conditions. At different conditions of food availability they applied different tactics of resource allocation between growth and reproduction.
