    Notes: Abstract Primitive terrestrial life – defined as a chemical system able to transfer its molecular information via self-replication and to evolve – probably originated from the evolution of reduced organic molecules in liquid water. Several sources have been proposed for the prebiotic organic molecules: terrestrial primitive atmosphere (methane or carbon dioxide), deep-sea hydrothermal systems, and extraterrestrial meteoritic and cometary dust grains. The study of carbonaceous chondrites, which contain up to 5% by weight of organic matter, has allowed close examination of the delivery of extraterrestrial organic material. Eight proteinaceous amino acids have been identified in the Murchison meteorite among more than 70 amino acids. Engel reported that l-alanine was surprisingly more abundant than d-alanine in the Murchison meteorite. Cronin also found excesses of l-enantiomers for nonprotein amino acids. A large collection of micrometeorites has been recently extracted from Antarctic old blue ice. In the 50- to 100-μm size range, carbonaceous micrometeorites represent 80% of the samples and contain 2% of carbon, on average. They might have brought more carbon than that involved in the present surficial biomass. The early histories of Mars and Earth clearly show similarities. Liquid water was once stable on the surface of Mars, attesting the presence of an atmosphere capable of deccelerating C-rich micrometeorites. Therefore, primitive life may have developed on Mars as well and fossilized microorganisms may still be present in the near subsurface. The Viking missions to Mars in 1976 did not find evidence of either contemporary or past life, but the mass spectrometer on the lander aeroshell determined the atmospheric composition, which has allowed a family of meteorites to be identified as Martian. Although these samples are essentially volcanic in origin, it has been recognized that some of them contain carbonate inclusions and even veins that have a carbon isotopic composition indicative of an origin from Martian atmospheric carbon dioxide. The oxygen isotopic composition of these carbonate deposits allows calculation of the temperature regime existing during formation from a fluid that dissolved the carbon dioxide. As the composition of the fluid is unknown, only a temperature range can be estimated, but this falls between 0° and 90°C, which would seem entirely appropriate for life processes. It was such carbonate veins that were found to host putative microfossils. Irrespective of the existence of features that could be considered to be fossils, carbonate-rich portions of Martian meteorites tend to have material, at more than 1000 ppm, that combusts at a low temperature; i.e., it is an organic form of carbon. Unfortunately, this organic matter does not have a diagnostic isotopic signature so it cannot be unambiguously said to be indigenous to the samples. However, many circumstantial arguments can be made to the effect that it is cogenetic with the carbonate and hence Martian. If it could be proved that the organic matter was preterrestrial, then the isotopic fractionation between it and the carbon is in the right sense for a biological origin.

    Notes: Abstract. Ageing can be defined as the time-dependent decline of physiological functions of an organism. The molecular causes for the ageing process are multiple, involving both genetic and environmental factors. It has been proposed that antioxidants may positively influence the ageing process, protecting the organism against free radical-induced damage. Here we show that the antioxidant N-acetylcysteine (NAC) has a life-extending effect on Drosophila melanogaster. Dietary uptake of NAC results in a dose-dependent increase in median and maximum life span. Flies fed on 1 mg/ml NAC food live 16.6% longer; at 10 mg/ml, life span increases by 26.6%. We have examined the effect of NAC treatment on protein and RNA levels we observe an NAC-dependent increase in absolute amounts of total RNA and ribosomal RNA, but no differences in protein levels. The NAC effect on longevity may involve differential expression of specific mRNA genes, as suggested by RNA fingerprinting experiments.

    Notes: Abstract We amplified, via PCR, DNA segments from intron 1 of the tyrosine hydroxylase gene (TH01) and intron 40 of the von Willebrand factor gene (VWA) in ten nonhuman primate genera. In humans both introns contain polymorphic microsatellites with tetrameric repeats. Compared to the allelic ranges in human populations relatively short repeat arrays could be detected for the nonhuman primates typed, presumably reflecting an ancient precursor state at both microsatellite loci. Furthermore, our results provide evidence for an association of the average number of repeats present in different primate genera and their divergence time from man. DNA sequencing of VWA orthologues revealed a relatively high variability in the arrangement of repeats in the 5′-repeat arrays, the generation of which could probably be explained by polar mutational events.

    Notes: Abstract. Bioassay-directed identification of toxicants in an acetonic extract of a sediment of the riverine Spittelwasser in the industrial region of Bitterfeld (Germany) was conducted. For this purpose, a combination of chromatographical fractionation, chemical analysis, and a biotest battery including Vibrio fischeri (inhibition of bioluminescence), Daphnia magna (immobilization), and Scenedesmus vacuolatus (inhibition of cell multiplication) was applied. Major toxicants identified and confirmed were methyl parathion (D. magna), prometryn, N-phenyl-β-naphthalene amine, PAHs (S. vacuolatus), and tributyltin (all biotests). Toxicity to V. fischeri was dominated by elemental sulfur. Results indicate high toxicant loads in the sediment about 7 years after closedown of a majority of chemical production sites at Bitterfeld. Comparison of potential exposure and toxicity data indicate a severe hazard potential to aquatic organisms due to organic toxicants. The results illustrate the potency of a biotest battery for identification of toxicants in contaminated sediment within the frame of toxicity identification procedures.

    Notes: Abstract The distribution of the human endogenous retrovirus (HERV)-K genome was investigated by Southern-blot analyses using a HERV-K-env DNA probe. With the exception of one DNA-sample, obtained from a Chinese individual in whom an amplification of HERV-K was detected, Southern-blot analyses yielded identical hybridization patterns with DNA from peripheral blood lymphocytes of 37 normal healthy blood donors, with DNA from six tumor cell lines, or with 23 DNA samples prepared from various carcinoma tissues. To elucidate whether the integration of HERV-K genomes into the primate lineage occurred as a single event or as an integration with later expansion, we further examined the evolutionary history of HERV-K by Southern blot analyses with DNA samples from different primate species. We detected HERV-K genomes in Macaca mulatta and Macaca silenus, which represent Old World monkeys, but not in prosimians (Galago demidovii) and New World monkeys, represented by Saguinus fuscicollis, Saguinus oedipus, and Calliihrix iacchus. Thus, we assume that the infection of the primate lineage with HERV-K had occurred after the divergence of New World and Old World monkeys, but before the evolutionary expansion of large hominoids. In contrast to the apparent lack of HERV-Kenv sequences in DNA from tissue of the New World monkey Saguinus oedipus (cotton-top marmoset), we found HERV-K-DNA in the B95-8 cell-line, which is a Saguinus oedipus leukocyte cell-line, immortalized in vitro by Epstein-Barr virus (EBV) and cultivated in human cells. It may be speculated that HERV-K-DNA or HERV-K-particles were introduced into these cells during in vitro transformation with EBV.

    Notes: Abstract The behavioral and electroantennogram (EAG) responses of female Anopheles gambiae mosquitoes to pooled samples of freshly collected human sweat and human sweat incubated for 42–52 hr were tested. No behavioral or EAG response was obtained to pooled fresh sweat samples, whereas incubated pooled sweat samples produced a behavioral as well as an EAG response. GC-MS analysis of the headspace composition of the fresh sweat revealed ethanol (15.1% of the total amount of volatiles trapped), acetic acid (10.9%), and 3-hydroxy-2-butanone (9.5%) as the most abundant compounds; a wide range of ethyl esters was present as well. None of the ethyl esters was detected in the headspace collections from incubated sweat, while the relative amounts of ethanol, acetic acid, and 3-hydroxy-2-butanone were strongly reduced. In the latter collections, indole (27.9%), 1-dodecanol (22.4%), and 3-methyl-1-butanol (10%) were present in high amounts, while they were absent or present in only minor amounts in the headspace collections from fresh sweat. Geranyl acetone (6%) and 6-methyl-5-hepten-2-one (1.9%) were relatively abundant in both the fresh and incubated headspace samples. EAG responses were observed in response to indole, 6-methyl-5-hepten-2-one, and geranyl acetone.

    Notes: Abstract Canberra, the capital of Australia since 1911, has been developed into a modern city from its original site on a nearly treeless plain. Today the city has about 300,000 inhabitants and 500,000 trees. The authors were requested by the managers of the urban public tree resource to survey their asset and to develop a computer-based system that would aid them in anticipating future maintenance requirements and its costs. This paper reports on our response. We have surveyed 3,000 streets and parks in the city, noting the species, number, and condition of every public space tree. We have also obtained the dimensions of sample trees, noting their total height, maximum crown width, height of maximum crown width, diameter at maximum crown width, and height at crown break. A management system has been developed using Microsoft AccessTM. Using standard regression techniques available on the package JMPTM, we found that total tree height was related to age for all species and that all other parameters of interest were related to height or transformed values of height. We assumed a sigmoidal growth curve and calibrated 114 height/age curves to cover the 165,000 trees of the 340 species we have in our database. As well we used the data on tree condition to determine the rate at which populations change from healthy to stressed. By interviewing foremen and supervisors we were able to determine the maintenance treatments carried out in Canberra, the equipment used, and the number of trees that can be treated in a day, for each type of operation. The management system can be used to display the current inventory for each street or park, by suburb, in the database. It can also be used to model future increases in size or crown condition, to predict the operations that will be required as a consequence of tree growth or crown deterioration, and finally, by applying multipliers to equipment and personnel, to estimate the future costs of tree maintenance. Managers can use the system to anticipate problems such as uneven expenditure requirements in future years.

    Notes: Abstract Intravascular oxygenation and carbon dioxide removal remains a potentially attractive means for respiratory support in patients with acute or chronic respiratory failure. Our group has been developing an intravascular hollow fiber artificial lung that uses a pulsating balloon located within the fiber bundle to augment gas transfer. We previously reported on a simple compartmental model for simulating O2 exchange in pulsating intravascular artificial lungs. In this study we evaluate the O2 exchange model with gas exchange and PO2 measurements performed on an idealized intravascular artificial lung (IIVAL) tested in a water perfusion loop. The IIVAL has well-defined bundle geometry and can be operated in balloon pulsation mode, or a steady perfusion mode for determining the mass transfer correlation required by the model. The O2 exchange rates and compartmental O2 tensions measured with balloon pulsation in the IIVAL are within 10% of model predictions for flow and pulsation conditions relevant to intravascular oxygenation. The experiments confirmed that a significant buildup of PO2 occurs within the fiber bundle, which reduces the O2 exchange rate. The agreement between experiments and predictions suggests that the model captures the cardinal processes dictating gas transfer in pulsating intravascular artificial lungs. © 2000 Biomedical Engineering Society.
