    Notizen: The largest potential reservoir for the storage of potable water is in the unsaturated zone. Use of this space for the storage and retrieval of potable water is a multifaceted problem which requires application of the best talent from the scientific community.Artificial recharge has many similarities to liquidwaste disposal through deep wells. In both, the problem is to place liquid in a permeable lithologic unit at an economic rate, to predict movement and the chemical reactions and physical changes that take place while the liquid is in the reservoir. Differences between the two operations are principally in the type of fluid injected and the ultimate objective. In artificial recharge the objective is to store and retrieve water of good quality; in waste disposal the objective is to store permanently water of objectionable quality. In both artificial recharge and liquid-waste storage, the nature of the storage must be known, particularly that of the unsaturated zone. The techniques of investigation for recharge and waste disposal are generally the same.Water commonly is recharged by surface spreading through basins or by induced recharge from adjacent streams and lakes or through injection wells. Research in recharge through basins has been dominated by mathematical models based on idealized conditions and empirical relations, derived by experimental sequencing of recharge operations, and operational controls in the pretreatment of recharge water. Recharge by injection wells has been undertaken in a variety of hydrologic environments. In Israel efforts have been directed toward the analyses of diffusion and dispersion of the injected water. Much research in the United States has been directed toward the movement of bacteria and organic matter through an aquifer and toward the chemical modeling of changes in recharged water as it moves.Much more research is needed on the basic properties of aquifers, particularly in the unsaturated zone, and on all aspects of recharge-water quality. Research and the use of data produced are increasingly the responsibility of interdisciplinary teams which consider the geologic, hydraulic, and economic aspects of the system.

    Notizen: A field demonstration of surfactant-enhanced solubilization was completed in a shallow unconfined aquifer located at a Coast Guard Station in Traverse City, Michigan. The primary objectives of the study were: (1) to assess the ability of the vertical circulation well (VCW) system for controlling chemical extractants added to the subsurface; and (2) to assess the behavior of the surfactant solution in the subsurface, with a goal of maximum surfactant recovery. A secondary objective was to demonstrate enhanced removal of PCE and recalcitrant components of a jet fuel. The analytical results showed that the surfactant increased the contaminant mass extracted by 40–fold and 90–fold for the PCE and jet fuel constituents, respectively. The surfactant solution demonstrated minimal sorption (retardation) and did not precipitate in the subsurface formation. In addition, the VCW system was able to capture in excess of 95% of the injected surfactant solution. Additional field testing and full-scale implementation of surfactant-enhanced subsurface remediation should be performed.

    Notizen: Surfactant-enhanced subsurface remediation is being evaluated as an innovative technology for expediting ground-water remediation. This paper reports on laboratory and modeling studies conducted in preparation for a pilot-scale field test of surfactant-enhanced subsurface remediation. Laboratory batch and column studies evaluated the surfactant-contaminant ground-water interactions in an effort to properly design the field-scale demonstration. A series of laboratory tracer tests and numerical simulations were completed to demonstrate the effectiveness of the hydraulic system (a vertical circulation well—VCW) for capturing injected solutions in a shallow, highly conductive, unconfined ground-water formation. The results of these studies were then used to optimize the performance of the VCW system during the subsequent field-scale demonstration study which utilized the VCW for injecting and extracting a surfactant solution. Information from the simulation studies, combined with the results of the batch and column tests, was crucial for procuring regulatory approval for the field demonstration, and successful design of the field-scale demonstration.

    Notizen: There is continuing controversy over whether a degree of C4 photosynthetic metabolism exists in ears of C3 cereals. In this context, CO2 exchange and the initial products of photosynthesis were examined in flag leaf blades and various ear parts of two durum wheat (Triticum durum Desf.) and two six-rowed barley (Hordeum vulgare L.) cultivars. Three weeks after anthesis, the CO2 compensation concentration at 210 mmol mol−1 O2 in durum wheat and barley ear parts was similar to or greater than that in flag leaves. The O2 dependence of the CO2 compensation concentration in durum wheat ear parts, as well as in the flag leaf blade, was linear, as expected for C3 photosynthesis. In a complementary experiment, intact and attached ears and flag leaf blades of barley and durum wheat were radio-labelled with 14CO2 during a 10s pulse, and the initial products of fixation were studied in various parts of the ears (awns, glumes, inner bracts and grains) and in the flag leaf blade. All tissues assimilated CO2 mainly by the Calvin (C3) cycle, with little fixation of 14CO2 into the C4 acids malate and aspartate (about 10% or less). These collective data support the conclusion that in the ear parts of these C3 cereals C4 photosynthetic metabolism is nil.

    Notizen: The toxin coregulated pilus (TCP) is required for Vibrio cholerae to colonize the human intestine. The expression of the pilin gene, tcpA, is dependent upon ToxR and upon ToxT. The toxT gene was recently mapped within the TCP biogenesis gene cluster and shown to be capable of activating a tcpA::TnphoA fusion when cloned in Escherichia coli. In this study, we determined that ToxR/ToxT activation occurs at the level of tcpA transcription. ToxT expressed in E. coli could activate a tcp operon fusion, while ToxR, ToxR with ToxS, or a ToxR-PhoA fusion failed to activate the tcp operon fusion and we could not demonstrate binding of a ToxR extract to the tcpA promoter region in DNA mobility-shift assays. The start site for the regulated promoter was shown by primer extension to lie 75 bp upstream of the first codon of tcpA. An 800-base tcpA message was identified, by Northern analysis, that correlates by size to the distance between the transcriptional start and a hairpin-loop sequence between tcpA and tcpB. The more-sensitive assay of RNase protection analysis demonstrated that a regulated transcript probably extends through the rest of the downstream tcp genes, including ToxT and the adjacent accessory colonization factor (acf) genes. An in-frame tcpA deletion, but not a polar tcpA::TnphoA fusion, could be complemented for pilus surface expression by providing tcpA in trans. This evidence suggests that the tcp genes, including toxT, are organized in an operon directly activated by ToxT in a ToxR-dependent manner. Most of the ToxT expression under induced conditions requires transcription of the tcpA promoter. Further investigation of how tcp::TnphoA insertions that are polar on ToxT expression retain regulation showed that a low basal level of toxT expression is present in toxR and tcp::TnphoA strains. Overall, these observations support the ToxR/ToxT cascade of regulation for tcp. Once induced, toxT expression becomes autoregulatory via the tcp promoter, linking tcp expression to that of additional colonization factors, exotoxin production, and genes of unknown function in cholera pathogenesis.

    Notizen: Formation of a hanging dam, a large, thick sub-surface accumulation of frazil ice that blocked river flow, altered physical environmental conditions dramatically and common carp Cyprinus carpio, and brown trout Salmo trutta, evacuated the pool in which the dam developed. A mean of 80·1% of the pool volume was filled with frazil ice. Mean and bottom water velocities in the pool increased from 6 to 27 cm s−1 and from 4 to 21 cm s−1, respectively, when the hanging dam formed, and water depth decreased from a mean of 2·25 to 0·45 m. Activity levels of fish leaving the pool varied. Two carp had higher swimming activities when in the over wintering pool, compared to the period when they were out of it, and a third showed opposite results.

    Notizen: Movement of radio-tagged riverine brown trout Salmo trutta, white sucker Catostomus commersoni and common carp Cyprinus carpio in the Grand River, Ontario, was minimal during the winter except during periods of high water discharge or the break-up of river ice. Ice break-up and flooding occur when southern air masses penetrate northwards in winter and as air temperatures increase in spring. Both white suckers and common carp moved the longest distances, generally in a downstream direction, during flooding and ice break-up. Brown trout moved less during these times. All three species had positive relationships between distance moved and water discharge. During periods of high water discharge and ice break-up, white suckers and common carp used backwater habitats more and main channel habitats less than during periods of low water discharge. During high flows, brown trout tended to use runs more and pools less. The break-up of river ice and accompanying flooding resulted in several fish being stranded on the floodplain. Backwater habitats appear to be important areas of refuge for riverine fishes during periods of flooding and ice break-up.

    Notizen: This paper describes a framework for estimating the effectiveness of riparian vegetation in modifying the water temperature in streams. A physically-based model was incorporated into the framework and used to demonstrate that planting trees in strategic locations can have a substantial effect on water temperature in streams and, consequently, on the survival of target fish populations. Application of the framework suggested that equivalent plantings in different configurations can have markedly different outcomes in terms of resultant stream temperature. There is no single best solution for all stream-restoration projects but, through the use of this framework, environmental planners, designers and managers can compare scenarios and make appropriate decisions for specific streams.

    Notizen: Exponentially dividing culture forms of Trypanosoma brucei did not utilize glucose provided in the culture medium. The inclusion of 2-deoxyglucose in the medium had no effect on the growth of the trypanosomes. Glucose could be replaced by proline in the liquid phase of biphasic medium without affecting the doubling time of the organisms. Proline added to the culture medium in this way disappeared during the log phase of growth. Glucose in the culture medium was used by the trypanosomes only when the stationary growth phase had been reached. Lipid accumulated in stationary phase trypanosomes grown in glucose-containing medium, but there was no lipid accumulation in log phase organisms or in those which had been grown in proline-containing medium. Bloodstream trypanosomes transferred to liquid medium rapidly utilized glucose over the first 12 hr of culture, and this was accompanied by an accumulation of free pyruvate in the medium. The rate of glucose utilization fell off over the next 36 hr; this was accompanied by a lowering of free pyruvate in the medium and a rise in the proline oxidase activity of the trypanosomes. The possible biologic significance of proline to trypanosomes developing in the midgut of the tsetse vector is discussed.

Diphenylamine was shown to be a potent inhibitor of cyanide insensitive respiration in both bloodstream and newly established culture forms of the same isolate of Trypanosoma brucei, with the L-α-glycerophosphate oxidase system having the greatest sensitivity to the inhibitor. The NADH oxidase activity of bloodstream forms was at least twice as sensitive to diphenylamine as the corresponding activity in culture forms, suggesting different routes of NADH oxidation in the 2 forms. The oxidation of L-α-glycerophosphate was inhibited to a similar degree in both culture and bloodstream forms. L-α-glycerophosphate oxidation in bloodstream forms differed from that found in culture forms in that the bloodstream system, unlike that in the culture form, was unable to donate electrons to cytochrome c. In culture form trypanosomes there was a distinct difference in the degree of diphenylamine inhibition on the oxidation of L-α-glycerophosphate, NADH, and succinate, suggesting the participation of separate flavoproteins in the oxidation of these substrates.
