    Description: The facultative red algal epiphyte Acrochaetium sp. liberated spores preferentially and recruited more successfully in laboratory cultures when its host Gracilaria chilensis C. J. Bird, McLachlan et E. C. Oliveira was present. The same effect was also induced by cell-free medium from G. chilensis, suggesting it contained a molecular signal. Antibiotics prevented spore release in Acrochaetium sp., even when G. chilensis was present, suggesting a prokaryotic origin of the signal. Simultaneous application of N-butyl-homoserine-lactone (BHL) restored the spore-release capacity, which demonstrated that spore release was not directly inhibited by the antibiotics and indicated that bacterially generated N-acyl-homoserine-lactones (AHLs) regulate spore release. An involvement of AHL was further indicated by the fact that two different halofuranone inhibitors of AHL receptors also inhibited spore release when they were applied at relatively low concentrations. Of seven different AHLs tested, only BHL induced the effect. However, BHL was only active at relatively high concentrations (100 μM), and it was not detected in spore-release-inducing medium of G. chilensis. Another water-soluble AHL or an AHL structure analog is therefore probably the active compound in G. chilensis cultures. The data presented demonstrate that life cycle completion in Acrochaetium sp. strongly depends on bacteria, which are not always present in sufficient numbers on the alga itself. Exogenous bacteria that are associated with G. chilensis or with other potential substrates may therefore trigger timely spore liberation in Acrochaetium sp., provided that the necessary concentration of AHL is reached. This first finding of AHL perception in a red alga confirms that AHL signalling is more widespread among eukaryotes than was thought until recently. However, spore release of a second red alga, Sahlingia subintegra (Rosenv.) Kornmann, was unaffected by AHL, and the reaction observed is therefore not universal.

    Description: Summary: • Glomus intraradices is a widespread arbuscular mycorrhizal fungus (AMF), which has been found in an extremely broad range of habitats, indicating a high tolerance for environmental factors and a generalist life history strategy. Despite this ecological versatility, not much is known about the genetic diversity of this fungal species across different habitats or over large geographic scales. • A nested polymerase chain reaction (PCR) approach for the mitochondrial rRNA large subunit gene (mtLSU), distinguished different haplotypes among cultivated isolates of G. intraradices and within mycorrhizal root samples from the field. • From analysis of 16 isolates of this species originating from five continents, 12 mitochondrial haplotypes were distinguished. Five additional mtLSU haplotypes were detected in field‐collected mycorrhizal roots. Some introns in the mtLSU region appear to be stable over years of cultivation and are ancestral to the G. intraradices clade. • Genetic diversity within G. intraradices is substantially higher than previously thought, although some mtLSU haplotypes are widespread. A restriction fragment length polymorphism approach also was developed to distinguish mtLSU haplotypes without sequencing. Using this molecular tool, intraspecific genetic variation of an AMF species can be studied directly in field plants.

    Description: Picoeukaryotes are a taxonomically diverse group of organism less than 2 micrometers in diameter. Photosynthetic marine picoeukaryotes in the genus Micromonas thrive in ecosystems ranging from tropical to polar and could serve as sentinel organisms for biogeochemical fluxes of modern oceans during climate change. These broadly distributed primary producers belong to an anciently diverged sister clade to land plants. Although Micromonas isolates have high 18S ribosomal RNA gene identity, we found that genomes from two isolates shared only 90 of their predicted genes. Their independent evolutionary paths were emphasized by distinct riboswitch arrangements as well as the discovery of intronic repeat elements in one isolate, and in metagenomic data, but not in other genomes. Divergence appears to have been facilitated by selection and acquisition processes that actively shape the repertoire of genes that are mutually exclusive between the two isolates differently than the core genes. Analyses of the Micromonas genomes offer valuable insights into ecological differentiation and the dynamic nature of early plant evolution.

    Description: Sampling an intact sequence of oceanic crust through lavas, dikes, and gabbros is necessary to advance the understanding of the formation and evolution of crust formed at mid-ocean ridges, but it has been an elusive goal of scientific ocean drilling for decades. Recent drilling in the eastern Pacific Ocean in Hole 1256D reached gabbro within seismic layer 2, 1157 meters into crust formed at a superfast spreading rate. The gabbros are the crystallized melt lenses that formed beneath a mid-ocean ridge. The depth at which gabbro was reached confirms predictions extrapolated from seismic experiments at modern mid-ocean ridges: Melt lenses occur at shallower depths at faster spreading rates. The gabbros intrude metamorphosed sheeted dikes and have compositions similar to the overlying lavas, precluding formation of the cumulate lower oceanic crust from melt lenses so far penetrated by Hole 1256D.

    Description: The nitrogen cycling of Lake Cadagno was investigated by using a combination of biogeochemical and molecular ecological techniques. In the upper oxic freshwater zone inorganic nitrogen concentrations were low (up to ∼3.4 μM nitrate at the base of the oxic zone), while in the lower anoxic zone there were high concentrations of ammonium (up to 40 μM). Between these zones, a narrow zone was characterized by no measurable inorganic nitrogen, but high microbial biomass (up to 4 × 107 cells ml−1). Incubation experiments with 15N-nitrite revealed nitrogen loss occurring in the chemocline through denitrification (∼3 nM N h−1). At the same depth, incubations experiments with 15N2- and 13CDIC-labelled bicarbonate, indicated substantial N2 fixation (31.7–42.1 pM h−1) and inorganic carbon assimilation (40–85 nM h−1). Catalysed reporter deposition fluorescence in situ hybridization (CARD-FISH) and sequencing of 16S rRNA genes showed that the microbial community at the chemocline was dominated by the phototrophic green sulfur bacterium Chlorobium clathratiforme. Phylogenetic analyses of the nifH genes expressed as mRNA revealed a high diversity of N2 fixers, with the highest expression levels right at the chemocline. The majority of N2 fixers were related to Chlorobium tepidum/C. phaeobacteroides. By using Halogen In Situ Hybridization-Secondary Ion Mass Spectroscopy (HISH-SIMS), we could for the first time directly link Chlorobium to N2 fixation in the environment. Moreover, our results show that N2 fixation could partly compensate for the N loss and that both processes occur at the same locale at the same time as suggested for the ancient Ocean.

    Description: Patiria miniata, a broadcast-spawning sea star species with high dispersal potential, has a geographic range in the intertidal zone of the northeast Pacific Ocean from Alaska to California that is characterized by a large range gap in Washington and Oregon. We analyzed spatial genetic variation across the P. miniata range using multilocus sequence data (mtDNA, nuclear introns) and multilocus genotype data (microsatellites). We found a strong phylogeographic break at Queen Charlotte Sound in British Columbia that was not in the location predicted by the geographical distribution of the populations. However, this population genetic discontinuity does correspond to previously described phylogeographic breaks in other species. Northern populations from Alaska and Haida Gwaii were strongly differentiated from all southern populations from Vancouver Island and California. Populations from Vancouver Island and California were undifferentiated with evidence of high gene flow or very recent separation across the range disjunction between them. The surprising and discordant spatial distribution of populations and alleles suggests that historical vicariance (possibly caused by glaciations) and contemporary dispersal barriers (possibly caused by oceanographic conditions) both shape population genetic structure in this species.

    Description: The results of seismic studies in the near-shore, shallow waters of the south-western Kara Sea - at the Shpindler, Kharsavey and Mare-Sale sites - showed the presence of a seismic interface which can be interpreted as a submarine permafrost table. The proposed permafiost exhibits a continuous distribution and a strongly dissected top surface overlain by unfrozen sediments. The permafrost table is located at a depth of 4-6 m and 5-10 m below the sea floor at the Shpindler and Mare-Sale sites, respectively. Three dimensional modeling of the permafrost table suggests the presence of relict buried thermodenudational depressions (up to 2 km across) at a minimum sea depth of 40-45 m at the Shpindler and Mare-Sale sites. The depressions may be considered as paragenetic to thermocirques found in cliffs at the Shpindler site. At the Kharasavey site, the permafrost table has an elongated depression parallel to the modern shoreline. The maximum depression depth is 20 m below the seafloor. At present, the relict therrnocirques (Shpindler and Mare-Sale) and the elongated depression (Kharasavey) are completely filled in with sediment and are not evident in modern bottom topography.

    Description: Under the framework of the Arctic Coastal Dynamics (ACD) program two projects of the International Association for the Promotion of Co-operation with Scientists from the New Independent States of the Former Soviet Union (INTAS) focused on ACD related topics. The objectives of the first project "Arctic coastal dynamics of Eurasia: classification, modern state and prediction of its development based on GIS technology" (2002-2004) had been to develop a coastal classification and to generate GIS based map products for the coastal Zone of the Eurasian Arctic. The Russian Part of the circum-Arctic ACD classification and segmentation has been completed within this project. The overall objective of the second project "Arctic coasts of Eurasia: dynamics, sediment budget and carbon flux in connection with permafrost degradation" (2002-2005) is to quantify the material flux through coastal erosion in order to improve our understanding of the Arctic sediment and organic carbon budget. This presentation summarizes the main results of the two projects and provides an overview of more specific results which are shown in a series of Posters.
