    Notes: This paper describes the use of gas chromatography mass spectrometry for the confirmation of ephedrine in adulterated powdered ipecac and ipecac fluid extract. Deuteration studies of ephedrine hydrochloride and some related compounds were also performed in an attempt to postulate structures for some of the fragment ions appearing in the spectrum of ephedrine.

    Notes: The bacterial sensing system has been studied on three levels. First, a quantitative method has been devised for measuring the “action spectrum” of the bacterium in response to a sudden addition of attractant. Second, a technique has been developed for the rapid isolation of mutants defective in the transmission part of the sensing system. Third, a study of the effects of light on the transmission system reveals two components, one which generates tumbling and another which inhibits it.

    Notes: The biologically relevant conformers of enkephalin predicted by solid state, solution state, and theoretical energy studies have been compared with the published structure-activity data on these compounds. No conformational technique proposes a model consistent with all the pharmacological data; the shortcomings of each approach are evaluated. An alternative approach, which correlates the structure-activity data of opiate compounds with that of the enkephalins, is described and shown to produce a model consistent with the available structure-activity data.

    Notes: Membrane vesicles of Escherichia coli can be produced by 2 different methods: lysis of intact cells by passage through a French pressure cell or by osmotic rupturing of spheroplasts. The membrane of vesicles produced by the former method is everted relative to the orientation of the inner membrane in vivo. Using NADH, D-lactate, reduced phenazine methosulfate, or ATP these vesicles produce protonmotive forces, acid and positive inside, as determined using flow dialysis to measured the distribution of the weak base methylamine and the lipophilic anion thiocyanate. The vesicles accumulate calcium using the same energy sources, most likely by a calcium/proton antiport. Calcium accumulation, therefore, is presumably indicative of a proton gradient, acid inside.The latter type of vesicle, on the other hand, exhibits D-lactate-dependent proline transport but does not accumulate calcium with D-lactate as an energy source. NADH oxidation or ATP hydrolysis, however, will drive the transport of calcium but not proline in these vesicles. Oxidation of NADH or hydrolysis of ATP simultaneous with oxidation of D-lactate does not result in either calcium or proline transport. These results suggest that the vesicles are a patchwork or mosiac, in which certain enzyme complexes have an orientation opposite to that found in vivo, resulting in the formation of electrochemical proton gradients with an orientation opposite to that found in the intact cell. Other complexes retain their original orientation, making it possible to set up simultaneous proton fluxes in both directions, causing an apparent uncoupling of energy-linked processes. That the vesicles are capable of generating protonmotive forces of the opposite polarity was demonstrated by measurements of the distribution of acetate and methylamine (to measure the ΔpH) and thiocyanate (to measure the Δψ).

    Notes: The intracellular locations of enzymes and metabolites were determined for ornithine metabolism in Neurospora. Pulse label experiments were used to measure the rates of intracellular translocations and the sizes of compartmented pools of metabolites in the mitochondrial, cytosolic and vesicular compartments. The results indicate that rapid equilibration occurs between these pools during growth in minimal medium, although the vast majority of the ornithine is confined to the vesicular compartment. Arginine, the biosynthetic end-product of ornithine metabolism, regulates ornithine utilization through a combination of feedback inhibition, repression, and control of intracellular translocations. The last phenomenon plays a decisive role indicating that the regulation of intercompartmental translocations may be a common mechanism in rapid adaptation responses in eukaryotic cells.

    Notes: 13C spin-lattice relaxation times (T1's) are reported for C-3 of 2-methylindole (methyl,3-13C2) as a function of the concentration of added 1,3,5-trinitrobenzene at 35°C in 1,2-dichloroethane. The observed decreases in T1, with increasing concentrations of 1,3,5-trinitrobenzene, are interpreted in terms of longer time-averaged correlation times which result from (a) the formation of increasing amounts of electron donor-acceptor complex and (b) increases in viscosity. An equation is derived which makes it possible to obtain estimates of the equilibrium constant for complex formation, and the spin-lattice relaxation time of the complex, from the observed T1's and viscosity measurements. From the data obtained, values of 6.4 × 10-12 and 14.1 × 10-12 s rad-1 were calculated for the effective correlation times (at 35°C and 0.686 centipoise) and 0.21 and 0.28 nm for the effective radii of free and complexed donor respectively.

    Notes: The mass spectra of representative 1,1-dicarboxylates have been studied in order to identify fragmentation patterns typical of such compounds and thus to aid their characterization in complex mixtures. Two typical decomposition pathways were noted. One invoves the loss of a carboxylate group, and the other the ejction of an sliphatic sidechain. It is suggested that a similar gragmentation scheme is to be expected of the much more labile dialkanoates derived from ketones.
