    Description: The common marmoset (Callithrix jacchus) is increasingly attractive for use as a non-human primate animal model in biomedical research. It has a relatively high reproduction rate for a primate, making it potentially suitable for transgenic modification. Although several attempts have been made to produce non-human transgenic primates, transgene expression in the somatic tissues of live infants has not been demonstrated by objective analyses such as polymerase chain reaction with reverse transcription or western blots. Here we show that the injection of a self-inactivating lentiviral vector in sucrose solution into marmoset embryos results in transgenic common marmosets that expressed the transgene in several organs. Notably, we achieved germline transmission of the transgene, and the transgenic offspring developed normally. The successful creation of transgenic marmosets provides a new animal model for human disease that has the great advantage of a close genetic relationship with humans.     Description: alpha-klotho was identified as a gene associated with premature aging-like phenotypes characterized by short lifespan. In mice, we found the molecular association of alpha-Klotho (alpha-Kl) and Na+,K+-adenosine triphosphatase (Na+,K+-ATPase) and provide evidence for an increase of abundance of Na+,K+-ATPase at the plasma membrane. Low concentrations of extracellular free calcium ([Ca2+]e) rapidly induce regulated parathyroid hormone (PTH) secretion in an alpha-Kl- and Na+,K+-ATPase-dependent manner. The increased Na+ gradient created by Na+,K+-ATPase activity might drive the transepithelial transport of Ca2+ in cooperation with ion channels and transporters in the choroid plexus and the kidney.     Notes: Z-Dehydrophenylalanine (ΔzPhe) possessing four oligopeptides, Boc-(L-Ala-ΔzPhe-Aib)n-OCH3 (n = 1-4: Boc, t-butoxycarbonyl; Aib, α-aminoisobutyric acid), were synthesized, and their solution conformations were investigated by 1H-nmr, ir, uv, and CD spectroscopy and theoretical CD calculation. 1H-nmr (the solvent accessibility of NH groups) and ir studies indicated that all the NH groups except for those belonging to the N-terminal L-Ala-ΔzPhe moiety participate in intramolecular hydrogen bonding in chloroform. This suggests that the peptides n = 2-4 have a 4 → 1 hydrogen-bonding pattern characteristic of 310-helical structures. The uv spectra of all these peptides recorded in chloroform and in trimethyl phosphate showed an intense maximum around 276 nm assigned to the ΔzPhe chromophores. The corresponding CD spectra of the peptides n = 2-4 showed exciton couplets with a negative peak at longer wavelengths, whereas that of the peptide n = 1 showed only weak signals. Theoretical CD spectra were calculated for the peptides n = 2-4 of several helical conformations, on the basis of exciton chirality method. This calculation indicated that the three peptides form a helical conformation deviating from the perfect 310-helix that contains three residues per turn, and that their side chains of Δz Phe residues are arranged regularly along the helix. The center-to-center distance between the nearest phenyl pair(s) was estimated to be ∼ 5.5 Å. The chemical shifts of the ΔzPhe side-chain protons (Hβ and aromatic H) for the peptides n = 2-4 indicated anisotropic shielding effect of neighboring phenyl group(s); the effect also supports a regular arrangement of the Δz Phe side chains along the helical axis. © 1993 John Wiley & Sons, Inc.

    Notes: Preparation and screening of twenty new ligands, all analogs of α,α,α′,α′-tetraaryl-1,3-dioxolane-4,5-dimethanol (TADDOL), for the Ti-catalyzed asymmetric addition of methyltri(isopropoxy)titanium and diethylzinc to benzaldehyde are described. These ligands have the dioxolane ring of the TADDOL's replaced by cyclobutane, cyclopentane, cyclohexene, cyclohexane, bicyclo[2.2.1]heptene and -heptane and bicyclo[2.2.2]octene and -octane moieties; several have H-atoms or alkyl groups in place of the aryl groups, and nine of them have C2 symmetry. X-Ray crystallography and molecular mechanics are used to analyze the structure of the ligands, and two structural features appear to correlate with selectivity: (i) the torsion angle for the chelating O-atom and the ortho-C-atom of the axial Ph group (a small, ca. 19°, angle is optimum, Fig.8) and (ii) the “degree of perpendicularity” of the axial Ph group (Fig. 9). Competition experiments indicate that TADDOL 1a catalyzes both the methyltitanium and diethylzinc additions ≥ 50 times faster than the related dioxolane analogs 12a, 12c, and 12e (Scheme 7), indicating that both axial and equatorial aryl groups (see Footnote 6) are necessary for ligand-accelerated catalysis of these reactions. A refined mechanistic hypothesis is presented (Fig. 10) to explain the selectivities observed for these new ligands. Our analysis suggests that a combination of structural features appear necessary for good catalytic efficiency and high selectivity. These features, especially the rather subtle conformational effects, appear to be optimized (among the ligands tested) in the TADDOL's.

    Notes: Copolymers of 2-(3-methacryloyloxypropyl)-4,5-diphenyloxazole (3) with various monomers were prepared. It was observed that the oxazole rings attached to the copolymer backbone are converted efficiently to the corresponding triacylamino residues by photooxidation using meso-tetraphenylporphin as a photosensitizer in solution and in a thin film of the polymer. This photoinduced transformation in a copolymer 5c having N-isopropylacrylamide as a comonomer was found to be applicable to a positive working photoresist by development with an aqueous solution of amines to dissolve the exposed areas of the film. A new system for positive image formation by visible light is presented.

    Notes: N-phenyl-α-methylene-β-lactam (PML), a cyclic analog of N,N-disubstituted methacrylamides which do not undergo radical homopolymerization, was synthesized and polymerized with α,α′-azobis (isobutyronitrile) (AIBN) in solution. Poly (PML) (PPML) is readily soluble in tetrahydrofuran, chloroform, pyridine, and polar aprotic solvents but insoluble in toluene, ethyl acetate, and methanol. PPML obtained by radical initiation is highly syndiotactic (rr = 92%), exhibits a glass transition at 180°C, and loses no weight upto 330°C in nitrogen. The kinetics of PML homo-polymerization with AIBN was investigated in N-methyl-2-pyrrolidone. The rate of polymerization (Rp) can be expressed by Rp = k[AIBN]0.55[PML]1.2 and the overall activation energy has been calculated to be 87.3 kJ/mol. Monomer reactivity ratios in copolymerization of PML (M2) with styrene (M1) are r1 = 0.67 and r2 = 0.41, from which Q and e values of PML are calculated as 0.60 and 0.33, respectively.

The kinetics of α-MDMP homopolymerization with AIBN was investigated in DMSO. The rate of polymerization (Rp) can be expressed by Rp = k[AIBN]0.49[α-MDMP]1.0 and the overall activation energy has been calculated to be 73.2 kJ/mol. Monomer reactivity ratios in copolymerization of α-MDMP (M2) with methyl methacrylate (M1) are r1 = 0.71 and r2 = 0.71, from which Q and e values of α-MDMP are calculated as 0.75 and -0.43, respectively. © 1993 John Wiley & Sons, Inc.

The kinetics of the MBDOON homopolymerization with AIBN was investigated in N-methyl-2-pyrrolidone (NMP). The rate of polymerization, Rp, can be expressed by Rp - k[AIBN]0.52[MBDOON]1.1 and the overall activation energy has been calcualted to be 75.7 kJ/mol. Monomer reactivity ratios in copolymerization of MBDOON (M2) with styrene (M1) are r1 = 0.31 and r2 = 3.20, from which Q and e values of MBDOON can be calculated as 3.0 and -0.7, respectively. Ring-substituted MBDOON monomers such as 6-chloro, 6-methyl, and 7-methoxy derivatives were synthesized and polymerized with AIBN. The 6-substituted MBDOON's readily underwent radical polymerization while the 7-methoxy-MBDOON was slower to polymerize. Poly(MBDOON) is predominantly heterotactic. (rr = 35, mr = 46, and mm = 19%). The polymer releases carbon dioxide at about 200°C and is converted with some depolymerization to poly[(o-hydroxyphenyl)acetylene]. The thermolysis temperature is very much affected by the ring substituent. The onset of carbon dioxide liberation was observed at 140°C in the case of the 7-methoxyl derivative while the 6-substituents had a smaller effect on the decarboxylation temperature. © 1993 John Wiley & Sons, Inc.

    Notes: A new dual-parameter of vinyl monomers for the reactivities toward free-radicals has been evaluated on the basis of kinetic data obtained by the flash photolysis method. As a model reaction system, the reversible addition reactions of the substituted phenylthiyl radicals to vinyl monomers are selected. The absolute rate constants and the relative equilibrium constants for these reactions have been determined. The Hammett reaction constants (ρ) for the addition reaction rates were evaluated by the variation of the substituents at the para-position of the arylthiyl radicals. From the values, the polar parameters (P- values) have been evaluated. The P values of various vinyl monomers correlate with the e values of the Q-e scheme proposed by Alfrey and Price. The resonance stabilization terms of the propagating radicals were evaluated from the equilibrium constants (K) which correlate with the Q values. The K-P dual-parameter can be used in order to predict the reactivities of vinyl monomers for the primary radicals (initiation step). The monomer reactivities from the copolymerization data are also correlated with the dual parameter. There are some advantages in the new dual-parameter compared with the Q-e scheme; the separation of the resonance term from the polar term is complete and the steric factor is not included in the polar term.

    Notes: Four aryl dichlorides containing secondary amide structural units as monomers were synthesized from substituted piperazines and chlorobenzoyl chlorides. Polyamides were prepared by the nickel-catalyzed coupling polymerization of these monomers. Polymerizations were carried out in N-methyl-2-pyrrolidinone (NMP) in the presence of nickel chloride, zinc, triphenylphosphine, and bipyridine, and resulted in polyamides with inherent viscosities up to 0.38 dL/g under mild conditions. The structure of polymer was determined by IR and 13C-NMR spectroscopies. The polyamide, poly[4,4′-biphenyldicarbonyl (2,5-dimethylpiperazine)] 6d, was readily soluble in dipolar aprotic solvents and chloroform. Thermogravimetry of polyamides, poly[(4,4′-biphenyldicarbonyl piperazine)]s 6, showed 10% weight loss at around 425 and 450°C in air and under nitrogen, respectively. © 1992 John Wiley & Sons, Inc.
